Ⅳ. 연구개발결과

Ⅳ. 연구개발결과
○ 전국에서 발효식품 126점을 수집하였으며, 이로부터 효모를 분리하였고, 기존 식품에서 분리한 효모와 함께 1,000주를 실험하였다. SO2 내성, 에탄올 내성, β-glucosidase, glucanase, protease, ureas의 효소활성능, 황화수소 생성능, 바이오제닉아민 생성능, cerulenin 및 TFL 저항성 실험을 통해 환경내성이 우수하고 효소활성능 높으며, 황화수소와 바이오제닉아민 비생성 효모를 선발하였다. 선발 효모 동정한 결과 Saccharomyces cerevisiae, Wickerhamomyces anomalus, Hanseniaspora uvarum, Hanseniaspora opuntiae, Pichia kudriavzevii , Candida tropicalis로 나타났다. 효모의 특성을 파악하기 위해, 당화액에 효모를 넣어 발효한 후, pH, 산도, 아미노산도, 유기산, 기호도 등을 조사하였다. 향기 패던에서 S. cerevisiae 와 non-Saccharomyces 는 서로 다른 것으로 나타났다. 당화액을 사용하여 S. cerevisiae와 C. tropicalis, P. kudriavzevii , H. opuntiae 효모를 1:1, 1:100, 1:10000의 비율로 혼합 발효한 결과, 1:100과 1:10000에서 효모의 특징이 나타났다. 전자현미경으로 선발한 효모의 외관을 관찰하였으며, 내산성, 내당성, 내염성, 내열성을 조사하였다. 또한 효모를 종균으로 사용하여 양조적성을 평가한 결과, non-Saccharomyces의 알코올 함량은 4~9%, S. cerevisiae 효모의 알코올 함량은 10~14%이며, 효모에 따라 pH, 당도, 산도, 유기산 함량이 다른 것으로 나타나 다양한 발효음료 개발 가능성이 있는 것으로 판단된다.
○ 종균 선발을 위한 발효식품 자원은 장류, 김치류를 포함하여 총 89점을 수집하였고, 수집된 자원으로부터 젖산균 739점, 고초균 833점, 효모 227점을 분리하였다. 분리된 균주의 효소활성검정 결과 α-amylase 93점, Protease 137점, Lipase 279, Fibrinolytic enzyme 196점의 활성을 확인하였다. 활성이 우수한 균주를 대상으로 동정을 실시하여, 청국장 제조용 고초균인 AFY-2와 김치 제조용 유산균인 AFY-3을 최종 선발하였다. AFY-2는 비독성 균주로 청국장의 풍미와 PGA 함량이 우수한 것이 특징이고, AFY-3은 내염성 및 저온생육이 우수하여 김치 맛 개선 및 아삭한 식감을 높여주는 것이 특징이다. 개발된 종균은 실용화를 위해 평창의 부일농산과 동해의 (주)락천식품에 각각 유상기술이전을 실시하였다.
○ 전통발효식품 유래 미생물을 분리하기 위하여 전국으로부터 44점의 발효식품을 수집하였고, 수집된 자원으로부터 3,540점의 미생물을 분리하였다. 분리한 미생물로부터 유용효소 활성 및 유용물질 생산능이 우수한 균주를 선발하기 위하여 단백질 분해능, 지질 분해능, 젖산 생성능, 초산 생성능, GABA 생성능, 유해균 생육 억제능 등을 평가하였다. 평가 결과, 지질 분해능 우수 균주 6점, 단백질 분해능 우수 균주 2점, 젖산 생성능 우수 균주 3점, 초산 생성능 우수 균주 1점, GABA 생성능 우수 균주 1점, 유해균 생육 억제능 우수 균주 1점을 선발하였다. 선발한 균주들의 유용효소 및 유용물질 생산능을 정량적으로 평가한 결과, MBE/L360, 865, 1050, 2205 균주가 최종 선발되었다. 선발한 4점의 균주의 배양조건에 따른 대사체 분석을 실시한 결과, 배양 조건에 따라 대사체가 상이하게 나타나는 것을 확인하였으며, 이를 활용하여 대사체 라이브러리를 구축하였다. 추후 이러한 결과를 바탕으로 종균의 활용성을 증대시키며 표준화된 고품질의 전통발효식품을 생산 할 수 있을 것으로 기대된다.

The objective of this study was to investigate what types of useful starter are produced by 1,000 wild type yeast strains isolated from Korean fermented foods. We selected the wild yeasts that had SO2, alcohol tolerance and enzyme activities (β-glucosidase, protease etc). The selected wild yeasts were identified as Saccharomyces cerevisiae, Wickerhamomyces anomalus, Hanseniaspora uvarum, Hanseniaspora opuntiae, Pichia kudriavzevii , Candida tropicalis. We characterized 12 yeasts of the genera Saccharomyces, Wickerhamomyces, Hanseniaspora, Pichia, Candida, and evaluated their enological potential. We investigated the pH, organic acid, preference degree of the wild yeasts by alcoholic fermentation. The aroma pattern of the S. cerevisiae and non-Saccharomyces were each investigated another. We selected non-Saccharomyces yeasts to combine with starter cultures of S. cerevisiae in mixed fermentation trials. The kinetics of growth and fermentation, and analytical profiles of the alcoholic beverages produced indicate that these non-Saccharomyces strains can be used with S. cerevisiae starter cultures to increase volatile compound production, and to increase or reduce the titratable acidity and organic acids of the final products, depending on yeast species and inoculum ratio used. The overall effects of the non-Saccharomyces yeasts on fermentation and alcoholic beverages quality were strictly dependent on the Saccharomyces/non-Saccharomyces inoculum ratio that the differences of fermentation conditions. We observed the morphology of the wild yeasts with an Scanning Electron Microscope (SEM). And we tested the tolerance of acid, sugar, salinity and temperature. Almost all Saccharomyces genera yeasts were showed to have a tolerance at 10% ethanol, but non-Saccharomyces genera yeasts displayed a low tolerance. The alcoholic beverages fermented by non-Saccharomyces yeasts demonstrated higher levels of soluble solids, titratable acidity, amino acids, and lower ethanol content, when compared with the alcoholic beverages fermented by Saccharomyces genera yeasts. Our results may contribute to a better understanding of the microbial interactions on wild yeasts fermentation.
Total 1,799 of food microorganisms (Lactic acid bacteria, 739 isolates, Baillus subtilis 833 isolates, Yeast 227 isolates) were isolated from Korea traditional fermented food in order to select useful starter. Bacillus subtilis AFY-2 producing high polyglutamic acid (PGA) in Chungkookjang and Leuconostoc mesenteroides AFY-3 enhancing Kimchi taste and crunch texture were selected as final starters. We provided two starters to local food companies at a cost.
We isolated 3,540 bacterial strains from korea traditional fermented foods in different province. To screen useful strains, we evaluated enzyme activities of isolates such as lipase, protease, fibrinolytic and antipathogenic activities and productivity of useful products such as acetic acid, lactic acid and GABA. Based on the results, we selected useful microorganisms and these strains were deposited at the KCTC and KCCM. Among them, MBE/L360, 865, 1050, 2205 strains were selected to investigate metabolome. By analyzing metabolic changes of selected strains in different culture conditions, we assumed bio-active materials and constructed metabolite library.
