1. 다소비 농산물의 식중독균 모니터링 DB 구축 및 유전적 다양성 조사
총 14종의 농산물을 대상으로 총 1,970점의 시료에 대해 위생지표세균(일반호기성세균, 대장 균군)과 유해미생물인 B. cereus, S. aureus을 정량적으로 조사하였고, E. coli, E. coli O157:H7, Salmonella spp., L. monocytogenes에 대해서는 정성적으로 분석하였다. 일반호기성 세균의 경우 0.01 ~ 7.18 Log CFU/g 범위의 밀도로 분포하였고 대장균군의 경우에는 총 1,970점의 시료 중 801점(40.7%)의 시료에서 검출되었고 밀도 범위는 0.01 ~ 5 Log CFU/g 이상인 것으로 나타났다. B. cereus는 224점의 시료(11.3%)에서 검출되었고 그 범위는 0.01 이하에서 2.56 Log CFU/g이었다. S. aureus는 21점의 시료(0.1%)에서 검출되었고 그 범위는 0.01 ~ 1.69 Log CFU/g이었다. E. coli는 총 106점의 시료(5.3%)에서 양성으로 나타났고, E. coli O157:H7, Salmonella spp., L. monocytogenes에 대해서는 양성 시료가 없었다. 시험대상 작물에서 분리된 총 439개 B. cereus 균주에 대해 enterotoxin인 hemolysin BL(hblC,D,A)과 nonhemolytic enterotoxin(nheA,B,C) 유전자 보유여부를 검정한 결과 hblA, hblB, hblC 모두를 가지고 있는 균주는 총 29개 균주이고 nheA, nheB, nheC 모두를 가지고 잇는 균주는 30개 균주인 것으로 나타났다. E. coli의 경우 총 112 균주에 대해 ETEC의 LT, ST, EHEC의 stx1, stx2, EIEC의 ipaH, EPEC의 bfpA, eaeA, EAEC의 aggR 유전자 보유여부를 검정한 결과 2개의 균주에서 EPEC의 eaeA 유전자 보유가 확인되었고, 다른 110개 균주에서는 모두 보유하지 않은 것으로 나타났다. 시험대상 작물에서 분리된 B. cereus, S. aureus, E. coli 균주를 대상으로 B. cereus, S. aureus에 대해서는 14종의 항생제, E. coli에 대해서는 13종의 항생제에 대한 내성을 검정한 결과 B. cereus의 경우 Cefoxitin, Cephalothin, Amoxicillin-clavulanic acid, Ceftriaxone 항생제에 대해 각 87, 77, 90, 64% 비율의 균주가 저항성을 나타내는 것으로 나타났다. S. aureus는 Cefoxitin, Clindamycin, Amoxicillin-clavulanic acid, Ceftriaxone에 대해 각 37, 37, 40, 33% 비율의 균주가 저항성을 나타내는 것으로 나타났다. E. coli는 Telithromycin, Cephalothin, Streptomycin에 대해 각 96, 61, 30% 비율의 균주가 저항성을 나타내는 것으로 나타났다.
2. 농산물 중 혐기성 식중독균 분석법 확립
농산물 중 enterotoxin 생성 Clostridium perfringens를 분석하기 위하여 먼저 토마토, 고추, 상추, 들깻잎에 오염된 Clostridium perfringens를 stomacher, sonication, pulsifier법으로 전처리하여 회수율을 비교한 결과 pulsifier법이 가장 회수율이 높았다. 또한 cpa, cpe에 대한 특이 프라이머를 각각 4종, 5종을 평가하여 가장 특이도와 민감도가 높은 프라이머를 1종씩 선발하였다. C. perfringens의 증식에 적합한 증균배지를 선발하기 위하여 영양 세포와 포자를 열, 저온, 산에 노출한 후 회복력을 비교한 결과, 영양세포 회복력정도는 cook meat broth ≥ liver broth > FTG ≥ WC broth > TSB 순이었고, 포자의 회복력은 회복력 정도는 cook meat broth ≥ liver broth > FTG ≥ WC broth > TSB 순으로, 포자나 영양세포 모두 cook meat broth가 가장 적합하였다. 또한 8종의 선택배지를 사용하여 민감도, 특이성을 분석한 결과, 8종의 배지 모두 민감도 100%였으나 특이도는 TSC, TSN, TSCF, SPS, SFP, OPSP, CP, CCP에서 각각 82.8%, 86.2%, 74.7%, 79.3%, 70.1%, 92.7%였다. 따라서 농산물 중 enterotoxin 생성 Clostridium perfringens를 분석하기 위해서는 cook meat broth에서 증균하고 pulsifier로 전처리한 후 cpa, cpe에 대한 특이 프라이머로 유전자 분석하여 양성인 시료를 TSC나 CCP 배지로 분리하면 보다 효율성을 높일 수 있을 것이라 사료된다.
농산물 중 Campylobacter jejuni, Campylobacter coli의 분석시 선택배지에서 위양성결과가 높아 분석에 어려움이 있다. 이를 개선하기 위하여 Bolton broth에 rifampicin(10 mg/L), potassium tellurite(2.5 mg/L), cefsoludin(10 mg/L), novobicin(8 mg/L), sulfamethoxazole(50 mg/L)을 첨가하여 효과를 평가하였다. 그 결과 rifampicn 첨가시 특이성이 높일 수 있었다. 최적의 rifampicn 첨가 농도를 결정하기 위하여 0 mg/L, 2.5 mg/L, 5.0 mg/L, 7.5 mg/L, 10.0 mg/L, 12.5mg/L를 Bolton broth에 처리하였으며 그 결과, 12.5 mg/L를 첨가하였을때 위양성결과를 보인 Acinetobacter calcoaceticus, Ochrobacterium anthropi, Ochrobacterium intermedium, Pseudomonas aeruginosa가 저해되었다. 한편 rifampicin 등 여러 종류에 내성을 가진 E. coli는 저해되지 않았다. 또한 개발된 배지 결합사용하였을 때 보다 효율성을 높일 수 있는 선택배지를 선발하기 위하여 특이성이 비교하였다. 그 결과, Campy Food agar > mCCDA > Preston agar > Brilliant Campylobacter agar > Wang agar > Abeyta-Hunt 순으로 Campy Food agar와 mCCDA가 가장 효과적이었다. 개발된 증균배지와 mCCDA, CFA 배지를 사용하여 농산물과 축산물 중 Campylobacter jejuni, Campylobacter coli을 분석하였을 때 ISO 분석법보다 현저하게 특이성을 높아지는 결과를 얻을 수 있었다.

This study investigated microbial contamination levels on fourteen kinds of agricultural products from farms stage to evaluate potential hazards associated with foodborne illness. A total of 1,970 samples were collected in major cultivating area from 2013 through 2015, and analyzed to enumerate aerobic bacterial counts, coliforms, Bacillus cereus and Staphylococcus aureus. In addition, the prevalence study for four kinds of pathogens (Escherichia coli, E. coli O157:H7, Salmonella spp. and Listeria monocytogenes) was performed on each sample. Aerobic bacterial counts ranged from 0.01 to 7.18 log CFU/g, with the highest bacterial cell counts recorded for watermelon. Coliforms were detected in 801 samples (40.7%) with a minimum of 0.01 log CFU/g and a maximum of more than 5 log CFU/g. B. cereus was detected in 224 samples (11.3%) ranging from <0.01 to 2.56 log CFU/g among total samples analyzed. S. aur us was detected in 21 samples (0.1%) with a minimum of 0.01 log CFU/g and a maximum of 1.69 log CFU/g. E. coli was detected in 106 samples (5.3%) among 1,970 samples. E. coli O157:H7, Salmonella spp. and L. monocytogenes were not isolated from any of the samples. Total 439 isolates of B. cereus were isolated from positive samples, and all isolates were evaluated for their genetic diversity and for their toxigenic profiles, i.e., the presence of hblA, hblC, hblD, nheA, nheB and nheC. The HBL complex (hblA, hblC, and hblD) was found in 29 strains (6.6%). The NHE complex (nheA, nheB, nheC) was found in 30 strains (6.8%). Total 112 isolates of E. coli were isolated and evaluated for their toxigenic profiles (LT and ST for ETEC; stx1 and stx2 for EHEC, ipaH for EIEC, bfpA, eaeA for EPEC and aggR for EAEC). The eaeA gene was detected in two strains isolated from barley. All strains except for two strains from barley were negative for pathogenic genes. The strains of B. cereus, S. aureus and E. coli isolated from samples were analyzed their antibiotic susceptibility using the Kirby-Bauer disc diffusion method. The B. cereus strains were highly resistant to cefoxitin (87%), cephalothin (77%), amoxicillin-clavulanic acid (90%) and ceftriaxone (64%). The S. aureus strains were resistant to cefoxitin (37%), clindamycin (37%), amoxicillin-clavulanic acid (40%) and ceftriaxone (33%). The E. coli strains were highly resistant to telithromycin (96%), cephalothin (61%) and streptomycin (30%). The microbial contamination levels of several agricultural products determined in this study may be used as the fundamental data for microbiological risk assessment(MRA).
We also conducted study on establishing the method for detection of Clostridium perfringens, Campylobacter jejuni and Campylobacter coli from produce. For detection method improvement of C. perfringens, PCR method was established. And newly designed enrichment broth-subsequent plating media combination was assessed on sensitivity and selectivity for Campylobacter detection in produce. In the results of establishment of the method for detection of C. perfringens, the primers designed by E. Augustynowicz and Yasuhiro were optimum in detection of cpa and cpe. The detection limit from pulsified samples was lower than that from sonicated and pummeled samples. And the detection limit of DNA extraction method using commercial kit was lower than that of boiling method. The recovery rate of stressed cell of five tested media was in the following order: cook meat broth ≥ liver broth > FTG ≥ WC broth > TSB. The specificity of TSC, TSN, TSCF, SPS, SFP, OPSP, CP, CCP was 82.8%, 86.2%, 74.7%, 79.3%, 70.1%, 92.7%, respectively. In the results of establishment of the method for detection of C. jejuni/coli, no significant differences in C. jejuni/coli growth were found between enrichment media with various levels of rifampicin. In addition, it was a combined use of R-Bolton broth and CFA that showed highest specificity (99.2 %). When applied to food samples, the combination decreased false-positive results by 58.3%, 55%, 16.7% and 8.3% for romaine lettuce, chicken, tomato and Korean leek respectively compared to a standard method. It was demonstrated that combination of R-Bolton broth and CFA is useful to detect Campylobacter in food with suppressing non-Campylobacter bacteria.
