1. 농업환경 및 농자재 중 독소형 미생물의 오염실태 및 안전관리 모델 개발

Ⅳ. 연구개발결과
1. 농업환경 및 농자재 중 독소형 미생물의 오염실태 및 안전관리 모델 개발
생산단계의 토마토, 딸기 농업환경에서 유해미생물의 오염도를 분석하였다. 생산단계의 조사지점별 유해균 분포를 조사하기 위하여 생산량 상위10%이내의 주산단지의 딸기농가 30농가, 토마토 40농가를 선정하여 전국조사를 실시하였다. 시기별 생산단계의 지점별 유해균 분포를 조사하기 위하여 토마토와 딸기 주산지의 17농가를 대상으로 1년 동안 총 4284점의 시료를 수집하여 유해미생물의 오염도 변화를 조사하였다. 딸기 재배기간 중 B. cereus 와 S. aureus 의 발생량을 조사한 결과 휴지기에 감소하였다가 재배시기에 다시 증가하는 경향을 나타내었다. 토마토 재배기간 중 B. cereus 와 S. aureus 의 발생량을 조사한 결과 B. cereus는 토양 및 대기온도가 높은 7∼8 에 감소하였다가 이후 다기 증가하는 경향을 나타내었다. S. aureus 의 경우 고온기 뿐만 아니라 휴지기인 10∼12월에 감소하였다가 정식 이후 다시 증가하였다. 조사한 모든 지점에서 위해균이 분포하고 있음이 확인되었으나 그 밀도가 식중독을 일으키기 위한 최소량 보다 적었다. S. aureus로 확인된 60개의 분리균주에 대하여 sea, seb, sed 독소 유전자형을 확인한 결과 1개의 분리균주에서 sed 독소 유전자가 확인되었다. 또한 토마토 표면에서 S. aureus의 밀도 변화는 인공접종 후 시간별로 조사한 결과 접종 후 24시간 이내에 급격하게 감소하였으므로 자연상태에서 토마토 과실 표면에 S. aureus가 오염되더라도 토마토과실표면에서 S. aureus의 밀도가 증가할 가능성은 낮을 것으로 예상된다. B. cereus 와 S. aureus 발생량과 온도와의 상관관계를 분석한 결과 딸기, 토마토 재재환경에서 토양 및 피복재 표면 온도와 이들 균의 발생량은 음의 상관관계가 있었으므로 토양 내 유해균은 휴지기에 열소독으로 사멸 가능할 것으로 예상되었다.
2. 토마토 및 딸기의 유통-소비 단계별 독소형 미생물 생장특성 구명
딸기와 토마토를 많이 재배하는 지역의 유통 환경을 조사한 결과 재배지에서 소비지에 이르기까지 일정 수준의 온ž습도 변화가 관찰되었다. 따라서 딸기와 토마토에 S. aureus를 접종하여 생장 패턴을 조사하고 카이네틱 모델을 개발한 결과 저장 온도에 의한 rate 값에는 큰 차이가 없었으며, 다이나믹 모델 개발 결과 딸기와 토마토에 오염된 S. aureus 생장에는 유의적인 변화가 관찰되지 않았다. B. cereus 영양세포와 포자를 접종한 딸기 역시 S. aureus의 결과와 유사하였으며 토마토에 B. cereus를 접종한 경우 생장이 전혀 관찰되지 않아 카이네틱 모델을 개발할 필요가 없다고 판단되었다. 세척수에 따른 딸기의 S. aureus, B. cereus 저감화 효과분석 결과 구연산, 전해수 침지 시 큰 효과가 관찰되지 않았으며, 토마토는 염소수 100 ppm에 6분 침지 후 S. aureus는 3.5 log CFU/g, B. cereus는 3 log CFU/g의 저감화 효과가 관찰되었다.
3. 토마토 및 딸기에 오염된 독소형 미생물 위해정보 분석 및 위해성평가
B. cereus, S. aureus는 그램 양성균으로서 포자를 형성하고 자연계에 널리 분포한다. B. cereus의 최적 생육 온도는 28 ~ 35℃, 최저는 4℃, 최고는 48℃이다. 용혈성(Beta hemolytic) 이고, enterotoxin에 따라 구토형과 설사형으로 나뉜다. 일반적으로 식품 내 105-8 CFU/g 범위의 감염량인 것으로 알려져 있다. 병징의 경우 일반적으로 구토, 설사 등 경미한 증상을 나타내지만, 화농성감염증, 괴저, 수막염, 봉와직염 등이 보고된 바 있다. S. aureusdml 최적 생육온도는 35℃이며 7 ~ 47.8℃ 범위 내에서 생장할 수 있다. Staphylococcus 속의 enterotoxin은 single-chain 단백질이며 SEA, SEB, SEC1,2,3, SED, SEE 등 5종류가 보고되었고 최근에는 SEG, SEH, SEI가 보고되었다. 병징으로는 메스꺼움, 경련성 복통, 구토, 설사 등이 있을 수 있고, 심한 경우에는 탈수현상, 두통, 근육통 등이 발생한다고 알려져 있다. 유통단계별, 주산지별, 시기별 토마토, 딸기의 위생지표세균 및 유해미생물 오염도를 조사하였다. 토마토의 시료수는 생산지 310점, 판매지 308점, 딸기의 시료수는 생산지 375점, 판매지 267점 분석하였다. 생산단계 위생지표세균 오염도 검정 결과, 지역별 차이보다는 각 농가별 오염도 편차가 큰 것으로 나타났다. 시기별로는 기온이 높아지는 시기로 갈수록 위생지표세균(일반호기성세균)의 오염도가 높아지는 경향을 나타났고 대장균군/대장균의 경우에는 시기에 따른 오염도 변화에서 유의성을 나타내지 않았다. 유통단계별 오염도를 분석한 결과, 생산단계 보다는 판매단계(대형매장, 재래시장)의 위생지표세균의 오염도가 높아지는 경향을 나타내었다. 분석한 토마토, 딸기에서는 E. coli, E. coli O157:H7, Salmonella spp., L. monocytogenes는 검출되지 않았다. B. cereus, S. aureus의 정량적 분석 결과 평균 오염도는 각각 0.25, 0.005 Log CFU/g인 것으로 나타났고, 오염비율은 각각 28.5, 0.5%인 것으로 나타났다. 생산단계 토마토, 딸기의 B. cereus, S. aureus의 초기오염도를 beta 분포를 이용하여 초기오염 확률분포를 산출하였다. 유통, 판매, 소비 과정 중 B. cereus, S. aureus의 밀도변화를 확률적 시뮬레이션과 우리나라 국민의 토마토, 딸기 섭취량 산출 등을 통해 식중독 발생 가능성을 예측한 결과, 딸기-S. aureus는 평균 6.586-9, 딸기-B. cereus는 평균 7.642-9, 토마토-B. cereus는 평균 6.778-9로 나타났으며 토마토 -S. aureus는 토마토에서 사멸곡선을 나타내는 관계로 위해도결정 실시에서 제외하였다.

The purpose of this study was to analyze microbiological hazards for cultivation environments and personal hygiene of strawberry and tomato farms at the growth and harvesting stage. Samples were collected from thirty strawberry farms and forty tomato farms located in Korea and tested for Staphylococcus aureus and Bacillus cereus. To investigate the change in the distribution of the S. aureus and B. cereus, a total of 4284 samples including air born, soil or medium, mulching film, harvest basket, groves and irrigation water etc. were collected from eight strawberry farms and nine tomato farms during one year. As a result, total S. aureus and B. cereus in all samples were detected. Among the total bacteria of strawberry farms, S. aureus (glove: 0 ∼ 2.1 log CFU/100cm2, harvest basket: 0∼3.0 log CFU/100cm2, soil or culture media: 0 ∼ 4.1 log CFU/g, mulching film: 0 ∼ 3.8 log CFU/100cm2), B. cereus (glove: 0 ∼ 2.8 log CFU/100cm2, harvest basket: 0 ∼ 4.8 log CFU/100cm2, soil or culture media: 0 ∼ 5.3 log CFU/g, mulching film: 0 ∼ 4.5 log CFU/100cm2) were detected in all samples. The total bacteria of tomato farms, S. aureus (glove: 0 ∼ 4.0 log CFU/100cm2, harvest basket:0∼ 5.0logCFU/100cm2, soil or culture media:0∼6.1logCFU/g, mulching film: 0∼ 4.0logCFU/100cm2), B. cereus (glove: 0 ∼ 4.0 log CFU/100cm2, harvest basket: 0∼4.3 log CFU/100cm2, soil or culture media: 0∼5.9 log CFU/g, mulching film: 0∼4.7 log CFU/100cm2) were detected in all samples. The contamination of S. aureus and B. cereus were detected in soil, mulching film and harvest basket from planting until harvest to processing, with the highest count recorded from the soil. But S. aureus and B. cereus were not detected in irrigation water samples. The incidence of S. aureus and B. cereus in hydroponics culture farm were less than those in soil culture. In particular, the amount of S. aureus and B. cereus detected in strawberry and tomato farms were less than the minimum amount required to produce a toxin that induces food poisoning. In this way, the degree of contamination of food poisoning bacteria was lower in the production environment of the Korea strawberry and tomato, but problems can be caused by post-harvest management method. These results will be used as fundamental data to create a manual for sanitary agricultural production environmental management and post-harvest management should be performed to reduce the contamination of hazard microorganisms.
Quantitative Microbial Risk Assessment for Toxin-Producing Bacteria in Tomato and Strawberry.
As interest in well-being, the consumption of organic fruits and vegetables has been increased. Fresh fruits and vegetables sanitation is important in all process stages because of consumption immediately without heating. In fact, Staphylococcus aureus and Bacillus cereus have been detected in domestic strawberries and tomatoes. Thus it is necessary to study in order to perform a scientific risk assessment and establish safety management standards. The objective of this study was to develop a dynamic model of toxin-producing bacteria and to evaluate antimicrobial effect to reduce initial contamination of the bacteria on strawberries and tomatoes. The distribution of temperature and humidity in strawberries and tomatoes were measured. Also the kinetic behavior of S. aureus and B. cereus in strawberries and tomatoes as a function of storage temperature and time were investigated. Dynamic model as a predictive model was developed, and the goodness of fit of the developed model was evaluated. In addition, enterotoxins of S. aureus and B. cereus were measured. To analyze the effect of reduction of the bacteria in the washing water, strawberries were immersed in citric acid, electrolyzed water and tomatoes were immersed in electrolyzed water, ozonated water, and chlorine water. As a result, temperature and humidity in distribution of strawberries and tomatoes were generally constant, and the rate (μmax) was not significantly different at all storage temperatures. Also, the development of dynamic model fitted S. aureus cell counts in strawberries and overpredicted B. cereus cells in strawberries and S. aureus cells in tomatoes. Vegetative cells and spores of B. cereus inoculated in strawberries were similar result of S. aureus. However, B. cereus growth in tomatoes was not observed. Therefore, a dynamic model of B. cereus in tomatoes was not developed. The effect of washing water on microbial reduction in strawberries was not observed during the immersion. In case of tomatoes, immersion at 100 ppm chlorine water for 6 min showed 3.5 log CFU/g reduction of S. aureus and 3 log CFU/g reduction of B. cereus. These results can be used as scientific evidence for quantitative microbiological criteria and will also be used to establish safety manegement guideline of toxin-producing bacteria in strawberries and tomatoes.
