○ 1세부: 임상시험을 통한 식물성 색소 성분 유래 인지능 개선 소재의 효능 및 안전성 평가

Ⅳ. 연구개발결과

2) 흑미 추출물의 안전성 확인함

- 약물 복용 3개월 후 이학적 검사, 혈액학적 이상소견 발생 없었음
3) 임상시험을 통한 흑미 추출물의 효능평가
- 총 53명의 대상자로 연구가 완료되었으며 대상자의 평균 나이는 64.15 ± 7.31세였고, 여성의 비율은 66%였다. APOE4 유전자를 하나라도 보유한사람은 10.4%였고 이것은 여러 문헌에 보고되는 것과 비슷한 수치였음
anthocyanin을 12주간 복용한 군과 위약을 복용한 군간의 통계적으로 유의한 인지기능 개선 효과는 없었고 특히 CERAD-K 검사의 세부항목에 개선효과가 없었음. 그러나 ADAS-cog 총점은 서로 다른 기울기로 anthocyanin 복용 군에서 인지기능의 개선 효과가 있는 경향이 있었으나 이는 통계적으로 유의하지는 않았음. 이러한 경향성은 APOE4 gene을 보유의한 군을 제외했을 경우에 더욱 두드러졌음
4) 기술이전
- 흑미 추출물을 유효성분으로 함유하는 퇴행성 뇌질환 치료 및 예방용 조성물 특허실시 (㈜한국전통의학연구소와 기술이전 계약)
○ 2세부: 농산물 유래 추출물의 신경세포 및 동물 모델을 이용한 인지능 개선소재 개발
1) 뇌해마신경세포에서의 glutamate와 amyloid beta에 의한 세포 손상에 대한 흑미, 검은콩, 오디 추출물의 효능
- 뇌해마신경세포에서 glutamate에 의한 Ca2+신호를 흑미, 검은콩, 오디추출물 모두가 농도의존적으로 억제함을 관찰함
- 뇌해마신경세포에서 glutamate에 의한 신경세포사멸을 3가지 추출물 모두에서 억제하였으나, 흑미추출물, 검은콩추출물은 15mg/ml과 30 mg/ml, 오디에서는 15mg/ml에서 유의한 효과가 나왔음
- 뇌해마신경세포에서 amyloide beta에 의한 세포사멸을 3가지 추출물 모두에서 억제하는 경향을 보였으나, 오디와 흑미추출물이 유의하게 억제함을 관찰함
2) 뇌해마신경세포에서 glutamate에 의한 세포손상에 검은콩추출물의 cyanidin-3-glucoside (C3G)가 미치는 영향
- 뇌해마신경세포에서 glutamate에 의한 Ca2+및 Zn2+ 신호의 증가와 미토콘드리아전압의 탈분극을 C3G가 유의하게 억제함을 관찰함
- 뇌해마신경세포에서 산화성 스트레스(ROS형성)을 C3G가 다른 항산화제와 마찬가지로 억제함을 관찰함
- 뇌해마신경세포에서 glutamate에 의한 세포사멸을 C3G가 억제함을 관찰함
3) 뇌해마신경세포에서 amyloide beta에 의한 세포사멸에 흑미추출물의 C3G 가 미치는 영향
- 뇌해마신경세포에서 amyloid beta에 의한 Ca2+및 Zn2+신호증가, ROS생성, 미토콘드리아전압의 탈분극을 C3G가 억제함을 관찰함
- 뇌해마신경세포에서 amyloide beta에 의한 세포사멸을 흑미추출물의 성분인 C3G와 peonidine -3-glucoside(P3G)가 억제하며 같이 사용시 더 효과가 강화됨을 확인함
4) 인지능저하 동물모델에서 흑미, 검은콩 오디추출물의 인지기능 개선에 미치는 효과
- 인지능 저하동물 생쥐에서 흑미추출물은 학습능력의 감소를 억제하였지만 검은콩 및 오디추출물은 유의한 효과가 없었음.
5) 양측 총경동맥 결찰에 의한 인지능저하 동물모델에서 흑미추출물의 뇌해마신경세포 손상에 미치는 효과
- 양측 총경동맥 결찰에 의한 인지능저하 동물모델에서 흑미추출물은 뇌해마신경세포 보호효과를 보였음
- 인지능저하 동물에서 나타나는 별아교세포의 증식을 흑미추출물이 억제함을 관찰
- 인지능저하 동물에서 glutathione peroxidase의 발현을 흑미추출물이 억제함
6) Amyloid beta 주입에 의한 알츠하이머성 치매 동물모델에서 흑미추출물의 뇌해마신경세포 손상에 미치는 효과
- 알츠하이머성 치매 동물모델에서 흑미추출물은 amyloide beta에 의한 학습능력감소를 억제하는 경향을 관찰함
- 흑미추출물은 알츠하이머성 치매 동물모델에서 나타나는 뇌해마신경세포의 사멸, 별아교세포의 증식, glutathione peroxidase의 발현에 유의한 효과가 없었음
○ 3세부:농산물 유래 인지능 개선 후보소재의 특성 규명 및 원료표준화
1) HPLC/PDA를 이용한 오디, 흑미, 검은콩의 HPLC 패턴 및 오디추출물, 흑미추출물, 검은콩 추출물의 HPLC 성분패턴을 조사하였으며 또한 UPLC-QTOF-MS를 이용한 오디, 흑미, 검은콩에 함유된 anthocyanin 성분을 MS spectrum을 통하여 분자량에 대한 정보를 이용하여 각 안토시아닌을 규명함
2) 오디, 흑미, 검은콩에서 공통적으로 높은 함량으로 측정되는 C3G를 분리하기 위해 소용량 및 대용량 고속향류크로마토그래피법을 이용하여 C3G를 분리하였음
3) HPLC-PDA를 이용한 안토시아닌 정량법을 개발하였다. 본 HPLC 정량법을 통하여 오디, 흑미, 검은콩에 함유된 C3G의 함량 차이를 비교 분석함
4) 인체적용시험을 위한 흑미농축분말을 제조하기 위해 여러 가지 제조공정을 만들었으며, 공장규모의 대량생산 및 실험실규모의 소량생산을 통하여 제조한 흑미농축분말에 함유된 C3G의 함량을 측정하여 그 함량이 일정하게 유지되는 조건을 조사하였고 최종적으로 C3G 함량을 표준화한 흑미농축분말의 제조공정을 확립하였음

○ Discovery and Biological Assay
During the first project year (2013), anthocyanins contained in crops such as black rice, mulberry and black bean were qualitatively and quantitatively analyzed by HPLC and Q-TOF MS spectroscopy and bioactivities of the extracts were evaluated by in vitro and in vivo screening model. Black rice extract was selected for the sample of clinical study because it showed the cognitive improvement activity and the extract showed the largest C3G content. According to the results, we produced the black rice extract powder through the first process of manufacturing for the clinical test.
In the second project year (2014), we established the large scale isolation process using high performance countercurrent chromatography (HPCCC) to yield cyanidin 3-glucoside, cyanidin 3-rutinoside and peonidin 3-glucoside for in vitro and in vivo test and qualitative and quantitative HPLC method. The two-phase solvent systems composed of tert methyl butyl ether-butanol- acetonitrile-water-TFA were used for the HPCCC separation. The HPCCC methods were more dominant than conventional column chromatography methods for the isolation of plant anthocyanins.
For the third project year (2015), we established quantitative and validate method to evaluate C3G content of the clinical samples (black rice extract powder). The precision was evaluated via inter and intraday variation and accuracy was checked by recovery test. We made the second manufacturing process to redeem the first manufacturing process which showed irregular C3G content. The second manufacturing process used the laboratory scale, and the black rice extract powder produced by second manufacturing process contained stable C3G content showing 5.28 mg/g black rice extract powder. The relative standard deviation was 3.58%. However, pilot scale manufacturing should be performed to convince the repeatable results.
○ Invitro neuronal cell model and animal model study
This study was designed to investigate whether 3 phytochemical extracts from black soybean, mulberry and black rice, affect cognitive function improvement in neuronal cell model and animal model.
Black rice extract, black soybean extract and mulberry extract inhibited glutamate-induced [Ca2+]i increases in cultured rat hippocampal neurons, in concentration-dependent manner. They also inhibited glutamate-induced hippocampal neuronal cell death. In addition, they reserved amyloid beta-induced [Ca2+]i increases and cell death in cultured rat hippocampal neurons. Cyanidin-3-glucoside (C3G), a major anthocyanin component of black rice extract, black soybean extract and mulberry extract, inhibited glutamate-induced [Zn2+]i increases, mitochondrial depolarization, formation of ROS in cultured rat hippocampal neurons. C3G also inhibited amyloid beta-induced [Zn2+]i increases, mitochondrial depolarization, formation of ROS. Simultaneous treatment with C3G and peonidin-3-glucoside (P3G) more effectively inhibited the amyloid beta-induced cell death than C3G and P3G alone. Only black rice extract of black rice extract, black soybean extract and mulberry extract, inhibited bilateral common carotid artery occlusion (BCCAO)-induced memory impairment in C57BL/6 mice, but other 2 extracts did not. Black rice inhibited BCCAO-induced hippocampal neuronal cell death, proliferation of astrocyte and expression of glutathione peroxidase. Black rice extract did not significantly affect amyloid beta-induced memory impairment, hippocampal neuronal cell death, proliferation of astrocyte and expression of glutathione peroxidase n C57BL/6 mice.
○ Clinical trial
Our goal was aimed to assess whether black rice extract powder containing C3G changed cognitive function in older adults with subjective memory complaints.
A 12-week randomized, controlled trial assessed cognitive outcomes in older adults (+60 year) with subjective memory complaints (n = 48) after consumption of 1.5g/day of either black rice extract powder (0.39mg of C3G per serving) or placebo with negligible anthocyanin content. ADAS-cog were measured at baseline and 12 weeks. ANOVA assessed change in cognition.
Improvements in total ADAS-cog scale were found in the black rice extract group but there was no statistical significance. A significant reduction in systolic (p = 0.038) blood pressure and a trend for diastolic (p = 0.160) blood pressure reduction was evident in the intervention group. Markers of inflammation (CRP and IL-6) were not altered.
This investigation failed to support the predicted beneficial effects of black rice extract short-term consumption on any of the neuropsychological variables included in this research. Future research is required to address several of the potential limitations of the current study, especially low C3G containing black rice extract.
