Digestive enzymes mean all kinds of enzymes that are able to hydrolyze and digest nutrient substances, such as starch, cellulose, protein and lipid, which are called amylase, cellulase, protease, and lipase, respectively. Since digestive enzyme products contain several enzymes with different mechanisms of action, test methods for digestion activities include those for starch, cellulose, protein and lipid.
At present, the digestion activity test methods are described in the section for general testing methods of the Korean Pharmacopoeia (KP). Therefore, enzyme products included in the KP monographs are tested according to such general testing methods. However, for enzyme products included in the Korean Pharmaceutical Codex (KPC), there are different test methods for individual monographs. In addition, the digestion activity tests described in the KP include those for starch (starch saccharifying activity, starch dextrinizing activity and starch liquefying activity), protein and lipid, while test methods described in the KPC monographs apply to measurement of starch saccharifying activity, starch dextrinizing activity, lipid digestion activity, protein digestion activity, cellulose digestion activity and cellulose saccharifying activity. When considering the applicability of general testing methods in the KP to products listed in the KPC, it is necessary to update the KP and the KPC, including harmonization of categories and test methods for determination of digestion activity.
In addition, similar enzyme products, such as biodiastase 2000 I and biodiastase 2000 II, included in the KPC have different test items and test methods for digestion activities. Therefore, it is necessary to establish reasonable test methods. Further, digestion activity test methods described in the KP are very similar to those described in individual monographs of the KPC. Accordingly, definition of test methods and activity units for individual enzyme products will contribute to easy quality control of raw materials and drug products and scientific evaluation of activities. Furthermore, if the most generalized and standardized test methods are defined and applied in standardization of specifications for raw materials and finished products of various digestive enzyme products listed in the KPC, efficient quality control of such products will be realized, contributing to development of local pharmaceutical industry.
