○ 본 연구는 B. cereus group에서 종간 구별을 하기 위한 바이오마커를 발굴하고, 발굴된 마커를 이용하여 신속하게 현장적용이 가능한 real-time PCR 시험법 개발하여 다음과 같은 결과를 얻었음
1. B. cereus group 분석을 위한 시험법 현황 및 문제점 발굴
- B. cereus group 진단 검출하는 생화학적 방법에 기초한 식품공전이나 BAM의 방법은 상대적으로 복잡하고 긴 시간이 필요하고, 분자생물학적 및 유전학적 기반 바이오마커들은 낮은 검출능과 높은 위양성이 발견되므로 새로운 바
○ 본 연구는 B. cereus group에서 종간 구별을 하기 위한 바이오마커를 발굴하고, 발굴된 마커를 이용하여 신속하게 현장적용이 가능한 real-time PCR 시험법 개발하여 다음과 같은 결과를 얻었음
1. B. cereus group 분석을 위한 시험법 현황 및 문제점 발굴
- B. cereus group 진단 검출하는 생화학적 방법에 기초한 식품공전이나 BAM의 방법은 상대적으로 복잡하고 긴 시간이 필요하고, 분자생물학적 및 유전학적 기반 바이오마커들은 낮은 검출능과 높은 위양성이 발견되므로 새로운 바이오마커의 개발이 필요함
2. B. cereus group 균주에 대한 염기서열 분석 및 유전체 데이터 확보
- B. cereus group 기준, 표준 및 야생 분리균주들 111개 및 non-Bacillus 음성대조균 9종을 포함하여 120종을 확보하여 실험에 사용하였음
- NCBI GenBank에서 B. cereus group에 속하는 289개 유전체 정보에 대한 생물정보학적 분석을 수행한 결과 B. cereus group은 유전체적 상동성이 매우 높게 나타났으며, 유전체, 16S rDNA, MLSA 및 유전자에 기반한 cladogram 분석은 전통적 분류기준으로 구별된 Bc 및 Bt 종간 및 종내 분류와 일치하지 않고 서로 섞여 그룹화됨
3. B. thuring iensis 기준균주의 전장유전체(whole genome) 분석을 세계 최초로 완료함
- B. thuringiensis 기준균주인 ATCC 10792는 염색체는 5.43Mb이고 5461개 유전자를 가지며, 5개의 플라스미드를 보유하였음
- 유전체 비교 분석은 ATCC 10792 genome이 Bt reference genome보다 Bc reference genome과 더 높은 상동성을 보임
4. lipoprotein 유전자에 대한 DNA 서열 분석과 cladogram 분석
- B. cereus 그룹 특이적 유전자로서 발굴된 lipoprotein 유전자의 DNA 서열 정보를 GenBank에 등록된 균주들에 대하여 얻은 후 cladogram 분석한 결과 B. cereus 그룹 균주들을 크게 16그룹으로 구분하였음.
- 각 그룹에는Bc와 Bt가 역시 혼재되어 있으므로 B. cereus group의 종간 및 종내 유연관계가 매우 가까우면서도 다양함을 나타냄
- 실험에 사용한 B. cereus 및 B. thuringiensis 기준균주, 표준균주 및 야생균주들에 대하여 lipoprotein 유전자의 DNA 서열화를 수행하고, cladogram 분석한 결과에서도 Bc 및 Bt 균주들의 종간 및 종내 유연관계가 매우 유사하면서 서로 혼재되어 나타났음
5. 유전체 정보 분석을 통해 B . cereus group 종간 구별이 가능한 바이오마커 3종(lipoprotein, methytransferase, X RE) 및 알려진 유전자 4종(g roE L, gyrB , motB , cry2)을 발굴 및 개발하였음.
- 120종류의 균주에 대한 B. cereus group 특이 마커인 lipoprotein(lipo-4), methyltransferase (MT-17), groE L, gyrB, motB 각각의 유전자 특이적 PCR primers의 검출능은 각각 95%, 85%, 97%, 95%, 95%이었고, 특이도는 각각 95%, 86%, 98%, 96%, 96%였음
- lipoprotein과 다른 마커들로 duplex PCR을 하면 B. cereus group 검출력이 100%에 도달함
- Bt 특이 바이오마커로 발굴한 XRE와 cry2의 검출능은 각각 93%, 81%이고, 특이도는 93%와 83%이였음.
6. B . cereus group 종간 구별을 위한 현장 적용 real-time PCR 시험법 개발
- 선발된 B. cereus group 바이오마커들의 real-time PCR 결과 10 fg DNA/reaction 농도까지 양성이었으며, 발굴된 바이오마커를 이용한 검출용 Real-Time PCR 조건을 최적화하였음.
- B. cereus를 인위적으로 오염시킨 상추, 시금치, 김밥을 대상으로 수행한 Real-time PCR 시험법의 현장 적용 가능성 검증하여 3.4x103∼1.5x104 CFU/g시료(또는 2∼10 CFU/PCR반응)에서도 양성으로 검출할 수 있었음.
7. 종합하면, B. cereus group은 생화학적, 유전학적, 유전체학적으로 매우 유사하므로 "B. cereus sensu lato"의 종 개념을 도입할 필요가 있으며, 본 과제에서 개발한 PCR primers 기반의 바이오마커들을 이용하면 세균 속에서 B . cereus group을 구별ㆍ확인할 수 있음.
본 용역과제를 통하여 얻은 위의 연구결과들은 다음과 같이 활용될 수 있을 것으로 사료됨.
1. B . cereus group 종간 구별 시험법 확립 기초 근거 자료로 활용될 수 있음.
2. B. cereus group 세균에 대한 오염원 및 오염도 파악을 위한 기초자료로 활용될 것으로 사료됨.
3. B. cereus group 종간 구별 시험법 확립 및 현장적용 시험법 개발을 통해 식품안전관리 정책 수립에 활용될 수 있을 것으로 사료됨.

This project was conducted for the development of Biomarkers and experimental methods for classification of Bacillus cereus group species.

2. Research on the comparative genomic analysis of B. cereus group
- bioinformatics and genomic analysis with 289 genomes obtained from NCBI were conducted to make phylogenetic tree and cladograms, which revealed B. cereus group as B. cereus sensu lato. 120 strains of B. cereus group and other bacteria were used.
3. Research on de novo assembly and comparative analysis of whole genome in B. thurigiensis type strain ATCC 10792
- ATCC 10792 has a chromosome of 5,43 Mb containing 5461 CDS, and 5 plasmids.
- The comparative analysis reveals that the genome of ATCC 10792 has high identities and similarities with B. cereus ATCC 14579 rather than B. thuringiensis reference genome konkukian 97-27.
4. Research on DNA sequencing and phylogenetic analysis of lipoprotein, a noble specific protein in B. cereus group
- sequencing results of lipoprotein gene showed that B. cereus group has high variation in the cladogram and was therefore grouped in 16 clades, and each group consisted of both B. cereus and B. thuringiensis.
5. Research on establishing biomarkers specific to B. cereus group
- 7 PCR primer sets were developed and evaluated as biomarkers specific to detect only B. cereus group or B. thuringiensis.
- The detection rates of those primers targeted to lipoprotein, methyltransferase, groEL, gyrB , and motB genes were 95%, 85%, 97%, 95% and 95% respectively among 120 bacteria including 111 strains of B. cereus group
- XRE gene-targeted PCR primers had high specificity(93%) to identify solely B. thuringiensis among B. cereus group and other non-Bacillus spp.
6. Research on the development of real-time PCR method to differentiate B. cereus group from samples of foods or vegetables in the field
- real-time PCR detection methods with lipo-4, groEL, gyrB or XRE primers specifically detected B. cereus group or B. thuringiensis
- The detection limits of real-time PCR with each 3 primers specific to B. cereus group was 10 pg of genomic DNA, and also was 1-10 CFU of bacteria/reaction in the artificially contaminated samples with B. cereus or B. thuringiensis.
Therefore, the above research outcomes were considered to be applied as described below:
- to be used as fundamental data supporting to establish the method to differentiate species of B. cereus group from non-Bacillus spp..
- to be used as fundamental data necessary for evaluating which sources of samples contaminated and which levels of contamination by B. cereus strains
- to be used as supportive data to revise microbiological anaytical manuals of "Food Code" and to standardize the manual to improve accuracy and reliability of microbiological detections
