1. 선행연구 조사 결과
환경 내 금속 나노물질의 노출평가와 관련된 선형연구 조사 결과, 제조 나노물질의 사용량이 최근들어 증가됨에 따라 이들이 환경 내로 유출되고 있는 것으로 나타났으나, 실제 환경 매질에서 존재하는 나노물질에 대한 정확한 정성·정량적인 결과는 거의 보고되지 않고 있었다. 현재까지 수질 환경매질에 노출된 나노물질의 평가는 주로 SEM, TEM, UV-vis 등에 의해 이루어지고 있는 것으로 파악되었다.

Ⅳ. 연구 결과
1. 선행연구 조사 결과
환경 내 금속 나노물질의 노출평가와 관련된 선형연구 조사 결과, 제조 나노물질의 사용량이 최근들어 증가됨에 따라 이들이 환경 내로 유출되고 있는 것으로 나타났으나, 실제 환경 매질에서 존재하는 나노물질에 대한 정확한 정성·정량적인 결과는 거의 보고되지 않고 있었다. 현재까지 수질 환경매질에 노출된 나노물질의 평가는 주로 SEM, TEM, UV-vis 등에 의해 이루어지고 있는 것으로 파악되었다.
나노물질국제기구(OECD, ISO) 및 EU 등 주요 선진국의 연구 동향 조사한 결과, 나노물질에 대한 연구는 국제적으로 경제협력개발기구(OECD), 국제표준화기구(ISO) 및 국제전기기술위원회(IEC)를 중심으로 안전성평가 및 국제 표준화 작업이 추진 중에 있다. 현재, 국제표준화기구(ISO), 국제전기기술위원회(IEC) 산하의 기술위원회(ISO/TC229, IEC /TC113)에서 나노기술 표준 제정 추진 중으로 ISO/TC229(한국 등 44개국 참여)는 산하의 4개 작업반(WG)을 설치하여 나노물질작업장의 보건 안전지침, 나노용어표준 등 국제 표준 10종 제정 완료 및 11년도 27종 표준 제정 추진이며, 2009년 9월 OECD 화학물질위원회 산하에 제조나노물질작업반(WPMN)이 설립하여 국제협력으로 나노물질의 안전성조사가 진행 중이다.
2. 자연발생 나노물질의 특성 평가 방법론 제시
현재 국내·외 자연발생 나노물질은 광산지역, 화산분출 또는 열수분출 지역, 토양 및 수계(하천, 호수, 해수) 등이 대표적인 발생지역으로 알려져 있으며, 점토광물, 금속 산화물, 앨러페인, 이모고라이트 등의 무기물질과 휴믹 물질을 포함한 자연유기물질과 polysaccharide 등의 유기나노물질이 대표적인 자연발생 나노물질인 것으로 알려져 있다. 현재까지 수행된 자연발생 나노물질 모니터링 사례로는 자연발생 철산화물의 거동에 대한 모니터링 연구정도이며, 자연발생 나노물질과 제조나노물질의 구별을 위하여 표면특성(코팅물질의 종류 및 특성), 원소 조성(원소별 농도 및 원소 간 농도비), 그리고 형태학적 특성 등을 이용한 정성 및 정량기법과 동위원소 조성비를 이용되고 있는 것으로 나타났다.
자연발생 나노물질과 제조나노물질의 구별을 위한 시범연구에서 티탄철석으로부터 기인한 자연발생 TiO2와 업체에서 사용되는 제조된 TiO2를 구별하기 위하여 두 물질 간 원소 조성비, 형태학적 특성 등을 비교해본 결과, Ti, Al, Fe 등의 농도가 상이하였으며, 제조나노물질은 구형에 가까운 형태이나 자연발생 나노물질은 불규칙한 형태 또는 반자형, 타형 등으로 나타났다. 따라서 원소의 조성과 형태학적 특성을 이용하면 자연발생 나노물질과 제조나노물질을 구별할 수 있을 것으로 판단되었다.
3. 주요 제조나노물질 배출원에 대한 모니터링 시범연구
주요 제조나노물질 배출원에 대한 모니터링 연구를 위한 대상 배출원을 선정하기 위하여 환경부 나노물질 유통량 조사 DB(2015년 6월 기준)를 검토하여 제조나노물질 취급업체는 AgNP 14곳, TiO2 115곳, ZnO 78곳에 대하여 아래와 같은 기준에 의하여 모니터링 시범연구를 위한 개황조사를 위하여 7개 업체를 선정하였다.
개황조사를 위해 선정된 나노물질 배출원의 모니터링 연구 수행을 위해 나노물질 관련 제품생산업체에서 발생되는 폐수(처리 전후의 유입수와 처리수)를 채취하여 수화학적 특성을 확인하고 나노입자의 존재 형태에 대한 분석을 수행하였다. 7개 업소(AgNP 1점, TiO2 4점, ZnO 3점)를 대상으로 1차(7월-8월), 2차(8월-9월)에 걸쳐 발생폐수와 처리수를 채취하여 나노물질 모니터링을 수행하였다.
AgNP 배출업소로 파악된 A사의 Ag 함량은 약 0.003㎍/L 수준으로 나타났으며, 공정에서 불산(HF)과 염산(HCl) 등을 주로 사용하는 공정의 특성상 공정에서 사용되고 있는 AgNP가 용해되어 입자상으로 배출되지 않는 것으로 파악되었다. TiO2 나노물질 배출업소로 파악된 B사, C사, D사, E사에 대한 폐수처리장 유입수와 처리수 내 Ti 함량 분석 결과, 업체에 따라 자체처리장의 유입수에서는 수십 ㎍/L ~ 수백 ㎍/L, 자체처리장의 처리 후 배출되는 처리수는 수십 ㎍/L의 Ti 농도를 보였다. 또한 시료 중에 Ti의 크기를 알아보기 위해 다단계 여과처리(무여과, 0.45 ㎛, 0.2 ㎛, 3 kDa 필터)에 의한 B사, C사, D사, E사의 시료의 입자 분석 결과, 유입수와 유출수에 주로 0.45 ㎛ 이상의 Ti 입자로 나타났으며, 일부는 0.45 ㎛ 또는 0.2 ㎛ 이하의 입자상 Ti와 용존상 Ti로 존재하는 결과를 보였다. ZnO 나노물질이 배출될 것으로 파악된 3개의 업체인 E사, F사, G사에 대하여 Zn 관련 분석을 수행한 결과, 대상업체들의 유입수에서는 수백 ㎍/L ~ 수천 ㎍/L, 자체처리장의 처리 후 배출되는 처리수에서도 수 ㎍/L ~ 수천 ㎍/L의 Zn 농도를 보였다. 다단계 여과처리(무여과, 0.45 ㎛, 0.2 ㎛, 3 kDa 필터)에 의한 E사, F사, G사의 입자 분석 결과, E사의 경우 유입수와 처리수 내에는 0.2 ㎛ 이하의 Zn 입자가 주로 존재하는 것으로 나타났으며, F사의 경우 유입수는 대부분 0.45 ㎛ 이상의 크기를 갖는 Zn 입자가 주로 존재하며 처리수에는 0.2 ㎛ 이하의 Zn 입자가 존재하는 것으로 나타났다. 또한 G사의 경우 유입수의 Zn 입자는 대부분 0.45 ㎛ 이상의 크기로 주로 존재하며, 처리수에는 대부분 용존상 Zn이 존재하는 것으로 나타났다.
개황조사 결과를 통해 업체 외부로 배출되는 처리수 내 0.45 ㎛ 이하의 입자상 물질이 존재하며, 주변 인근하천까지 노출 연관성이 있는 업체를 고려하여 집중모니터링 대상 2개 업체(B사, C사)를 선정하였으며, 2개 업체와 처리수가 유입되는 인근 공공하수처리시설(B사의 처리수가 유입되는 P, S 공공하수처리시설과 C사의 처리수가 유입되는 Y 공공폐수처리시설)과 이들 공공하수처리시설의 처리수가 방류되는 B사 인근 하천(B-1, B-2)과 C사 인근 하천(C-1)에 대한 입자상 Ti에 대한 집중모니터링을 수행하였다.
B사와 C사 및 주변 하천에 대한 집중모니터링 결과, 입자상 Ti는 B사와 C사의 자체처리시설 유입수에서는 비교적 높은 함량의 입자상 Ti가 검출되었으나, 처리수 내 입자상 Ti의 농도는 크게 감소하는 것으로 나타나 자체처리 및 공공하수처리시설에서 효과적으로 제거고 있는 것으로 나타났으며, 인근 하천으로 유입되는 방류수에서는 미량만이 존재하며, 또한 인근 하천수와 혼합된 후 응집침강 등의 다양한 작용에 의하여 입자상 농도는 급격히 감소하는 것으로 나타났다.
4. 화평법 상 위해우려 제품 군 내 나노물질 분석을 위한 기술 표준화 방법 연구
본 연구에서는 위해우려 제품군 내 나노물질 분석 표준화를 위하여 위해우려 제품을 제형과 매질의 특성에 따라 3가지 형태로 분류하였다. 분류 기준은 매질의 분해 방법에 의한 것으로서 물에 용해되는 제품은 수용성 시료, 물 및 유기용매에 용해되는 제품은 비수용성 시료, 용해가 되지 않는 시료는 고체상 시료로 분류하였다.

위해우려 제품 내 나노물질의 분석 절차는 크게 3가지 단계를 수행하는 것으로 하였다. 첫 번째 단계는 제품 내 금속물질의 전함량을 측정하는 단계로서 이 과정에서 금속 물질의 함유 여부를 판별한다. 첫번째 단계에 의해 제품 내 금속 물질이 있을 경우 두 번째 단계를 통해 금속물질이 입자상으로 존재하는지 용존상으로 존재하는지를 판별한다. 이를 위하여 두 번째 단계에서는 제품을 10 kDa 필터가 장착된 용기를 이용하여 고속원심분리한 후 용존상 금속물질의 함량을 정량적으로 측정한다. 입자상 금속물질의 함량은 금속물질의 전함량과 용존상 함량의 차이를 이용하여 산정한다. 두 번째 단계를 통하여 제품 내 입자상 금속물질의 존재가 확인 될 경우 세 번째 단계를 수행한다. 세 번째 단계는 입자상 금속물질의 크기 및 크기분포를 TEM을 이용하여 확인하는 단계이며, 이를 위하여 제품의 매질분해, 원심분리를 이용한 입자상 금속물질의 회수, 회수된 입자상 금속물질의 분산 과정을 수행하게 된다.
금속물질의 전함량 측정을 위하여 산분해 조건을 확인한 결과 AgNP 및 TiO2 물질의 경우 최적의 산분해 조건은 질산 또는 질산+황산을 이용하여 마이크로파 산분해하는 것이었다. SiO2의 경우 질산, 불산을 이용하여 마이크로파 산분해한 후 붕산을 이용하여 잔류 불산의 영향을 배제하였을 때 SiO2의 분석이 가능한 것으로 평가되었다.
금속물질의 용존상 함량의 측정을 위한 시료는 수용성 시료와 비수용성 시료이다. 수용성 시료의 경우 제품을 증류수 희석한 후 10 kDa 필터가 장착된 용기를 이용하여 고속 원심분리하는 방법을 통해 용존상 금속물질을 99% 이상 회수 가능한 것으로 나타났다. 비수용성제품 시료는 디클로로메탄을 이용하여 제품을 희석한 후 메탄올을 첨가하여 주입된 디클로로메탄의 함유량이 15% 이하가 되었을 때 10 kDa 필터를 이용하여 고속원심분리 여과법을 사용할 수 있었으며, 이 과정을 통해 회수된 용존상 금속물질의 회수율은 95% 이상으로 나타났다.
제품 내 함유된 입자상 금속물질의 크기 및 크기분포를 TEM 측정하기 위하여 3가지 단계를 수행하였다. 첫 번째 단계는 제품 매질을 분해하는 과정으로 이를 위하여 용매 분해법 및 회화법을 사용하였다. 용매 분해법은 증류수에 의하여 제품 매질이 용해되지 않는 비수용성 제품의 매질을 분해하기 위한 실험법이며, 용매 분해법에 사용되는 유기 용매는 입자상 금속물질의 용해 및 변형이 발생되지 않으면서 제품 매질을 분해할 수 있어야 한다. 연구 결과, 용매 분해법을 사용하기 위한 최적의 용매는 디클로로메탄인 것으로 확인되었으며, 디클로로메탄은 AgNP, TiO2 및 SiO2의 입자상 금속물질의 변형 및 용해 없이 비수용성 시료의 매질을 안정적으로 분해할 수 있었다. 회화법은 제품의 매질이 용해되지 않는 고체상 시료의 전처리 방법이며 회화법에 의한 입자상 금속물질의 용해 및 변형이 없어야한다. 회화법을 AgNP 및 TiO2, SiO2의 입자상 금속물질에 적용한 결과 최적의 회화 온도는 600℃에서 2시간 반응하는 것으로 나타났다. 이때 TiO2 및 SiO2의 손실 및 변형은 확인되지 않았다. 다만 AgNP의 경우 100~200℃에서 휘발되는 것을 확인하였기에 AgNP의 경우 회화법을 적용하기 어려웠다. 제품 매질 분해 후 입자상 금속물질을 회수하기 위한 방법으로 원심분리 방법을 사용하였다. 원심분리 방법은 시료의 점성, 입자상 금속물질의 밀도 및 지름, 원심력 등에 의하여 원심분리 시간이 달라진다. 이론적인 원심분리 시간을 계산하여 별첨하였다. 원심분리에 의한 입자상 금속물질의 회수는 별첨에 표시된 이론적 원심분리 시간보다 2배의 안전계수를 적용하여 수행하여야만 입자상 금속물질이 회수되는 것으로 확인되었다. 결론적으로 제품 매질 분해에 의해 매질에서 탈리된 입자상 금속물질을 회수하기 위해서는 별첨에 의거한 이론적 원심분리 시간의 2배에 해당되는 시간으로 원심분리 하였을 때 입자상 금속물질의 회수율을 95% 이상이 된다. 회수된 입자상 금속물질을 TEM 관찰하기 위한 마지막 단계로서 입자의 분산과정이 필요하다. 입자의 분산은 NIST(Nationnal Institute of Standards and Technology)와 CEINT에 의해 연구된 실험방법을 사용하였다. NIST와 CEINT에 연구된 자료에 의하면 입자의 분산을 위해 0.05% BSA용액을 이용한 후 초음파 분산하는 방법을 제시하였다.
본 연구에서 제시된 실험법의 적절성을 확인을 위해 표준물질 매질 첨가 실험을 수행한 결과, 표준물질 매질 첨가법에 의해 평가된 나노물질의 회수율은 95% 이상이었으며, 나노물질의 변형 및 손실은 없었다. 또한 금속성분의 전함량, 용존상 함량 및 회수된 입자상 함량의 회수율은 모두 95% 이상으로 나타나 본 연구에서 제시한 시험법에 의해 입자상 금속물질의 정성 및 정량분석이 가능한 것으로 판단되었다.
위해우려 제품군 내 22개 제품을 대상으로 시험법을 적용하였다. 1단계 금속물질의 전함량 분석에서 은은 8개 제품, 티타늄은 6개 제품, 그리고 규소는 8개 제품에서 검출되었다. 2단계 입자상 함량 분석에서 티타늄 함유 제품과 규소 함유 제품에서 입자상 함량이 높게 검출되었다. 은을 함유한 8개 제품 중 6개 제품에서 은의 입자상 함량이 검출되었으며, 2개의 제품에서는 은이 모두 용존상으로 존재하였다. 입자상 은이 검출되지 않은 2개 제품을 제외한 20개 제품에 대해 3단계 입자 크기 및 크기분포 측정 시험을 실시하였다. 본 연구에서 제시한 시험법을 통해 17개 제품에서 입자의 크기 및 크기분포를 측정하였으며, 관찰된 입자는 100nm 이하의 나노입자에 해당하는 크기를 나타내었다. 3개의 제품에서는 TEM 현미경 관찰 시 모든 입자가 응집된 형태로 존재하며, 응집체를 구성하는 1차 입자의 경계면이 뚜렷이 나타나지 않아 크기 측정이 불가능하였다.

As a result of increasing development and usage of nanomaterials within a number of countries, such as USA, Japan, EU, and Korea, a issue on their environmental release and exposure has been continuously raised. For the reliable tracing of released nanomaterials, research and development of relevant monitoring techniques is a prerequisite. Up to date, however, it has been insufficient. For this reason, this study has been launched. The objectives of this study were: (1) to investigate the research trend of environmental exposure of nanomaterials, (2) to suggest the methodology to distinguish between naturally-occurring and manufactured nonamterials through literature reviews and a preliminary study, (3) to conduct a demonstration study on the monitoring of representative releasing sources of manufactured nanomaterials, (4) to improve and enhance methodologies for characterize and analyze nanomterials, and (5) to propose a standard method to analyze nanomaterials contained in potentially-hazardous commercial products. The final goal was to prepare full-scale researches on the environmental exposure of nanomaterials through gathering and serving basic data, establishing methodologies, and suggesting strategies and policies.
Above of all, we reviewed overseas and domestic literatures on environmental exposure of nanomaterials as well as the R&D trend of nanomaterials within a number of advanced countries. The literature survey indicates that a variety of nanomaterials have been released into environments, but there have not been reported reliable results of field scale researches on identifying and detecting nanomaterials in real environments. Several international organizations, such as OECD, ISO, and IEC have tried to evaluate the safety of manufactured nanomaterials, to establish legislation to regulate commercial nanomaterials, and to prepare standard methods to measure and characterize nanomaterials.
A number of literatures were reviewed to investigate the methodologies to distinguish naturally-occurring and manufactured nanomaterials, and the review results suggest that there have been reported four typical methods, such as surface property (type and characteristics of coating materials), elemental composition (individual concentrations of each element and compositional ratio between elements), morphological feature, and isotopic composition. We conducted a preliminary study to confirm the methodology suggested in the literature. Titanium oxide (TiO2) nanoparticle which are naturally formed from ilmenite (FeTiO3, FeO·TiO2) was targeted, and 9 stream water samples which were likely to contain TiO2 nanoparticles were taken. Also, manufactured TiO2 nanomaterials were obtained from two domestic factories. And then, those two kinds of TiO2 nanomaterials analyzed and compared in terms of total concentration and relative concentration of each size fraction using ICP-MS. In addition, their particle properties (size, size distribution, shape, elemental composition) were observed using SP-ICP-MS and TEM/SEM-EDS. The preliminary results confirmed that naturally-occurring and manufactured TiO2 nanomaterials could be distinguished by comparison of morphological features and elemental composition (concentrations each element and compositional ratios between elements).
A demonstration study on representative releasing sources of nanomaterials was conducted to establish techniques for monitoring of nanomaterials released into environments. Stepwise researches, such as general and detailed survey were planed and undertaken. For the general study, 7 target factories were selected from total 207 enterprises, based on the consideration of type of nanomaterials dealt with, production of wastewater containing nanomaterials, and feasibility of sampling. Target nanomaterials were Ag, TiO2, and ZnO. Two kinds of samples, such as influent and effluent (before and after treatment by facility within the factory ) were sampled and analyzed with regards to total concentration and relative concentration of each size fraction using ICP-OES and ICP-MS. Also, particle properties, such as size and size distribution were measured using SP-ICP-MS. The results suggest that Ag nanomaterials were not detected because of usage of fluoric acid (HF) and hydrochloric acid (HCl) during the manufacturing process. In th case of ZnO nanomaterials, nano-sized particles were not detected and most of them were observed to be dissolved. In terms of TiO2 nanomaterials, finally, nano-sized fractions were detected in two plants, and those two sites were considered to be targeted in the detailed survey. In the detailed survey, 7 kinds of samples were obtained: influent and effluent (before and after treatment by facility within the factory), influent and effluent (before and after treatment by public wastewater treatment system), sludge (obtained from public wastewater treatment system), and nearby stream waters (before and after mixing with effluent from public wastewater treatment system). The results of detailed survey indicate that even though the original wastewater produced from manufacturing process appeared to contain a relatively high concentration of particulated TiO2 nanomaterials, most of them seemed to be removed through treatment systems. In addition, a small amount of particulated TiO2 nanomaterials contained in effluents was investigated to be released into nearby streams, but it is likely that their concentration was drastically decreased after confluence as a result of aggregation and sedimentation caused by various interactions occurring within the stream environments.
Another task of this study was conducted to develop analytical methods for particulate metallic material contained in the detergent product group. Therefore detergent product group were classified into three type of readily soluble matrix and readily soluble matrix in non-polar solvent and not soluble matrix. Advance three-step analysis of the particle metal material; first, analysis of total metal; second, analysis of dissolved metal; third, measurement of particle size. The content of particulate metal material can be obtained at the difference in the metal components of the dissolved phase in an amount of total metal components. Total metal analyzes of titanium and silver was the best of the microwave decomposition method using nitric acid. In the case of silicon it was possible to measure when using the boric acid dilution after microwave decomposition with hydrofluoric acid and nitric acid. Measurements of the dissolved metal material, the liquid is filtered using a high-speed centrifugal separation method using a 10 kDa filter was performed to measure in ICP. Measurement of the size of the particulate metallic material was carried out in three step. The first step utilizes a pyrolysis or dissolution, a step of decomposing the matrix of the product. The second step utilizes a centrifugal separation, a step of recovering the particulate metal material. The final step, after recovered particulate metal material the dispersion, a step of measuring the size using a TEM. Set result of verifying the experimental methods in the present study, the total metal material showed a recovery rate of 99%, the dissolved metal materials has become 95%. Further, it was possible to create the size and size distribution of the particulate metal material using the experimental method of the present study. In this process, damage and deformation of the nano metal material has not been confirmed. In conclusion, using the experimental methods of the present study, it was found that the particulate metallic material in detergent product group can be stably analysis.
Consequently, this study showed the methodology and procedure for monitoring nanomateirlas released into environments. In addition, a standard method was proposed to qualitatively and quantitatively analyze metal nanomaterials contained in potentially-hazardous commercial products. In particular, the proposed standard method should be taken into account when policies as well as regulatory frames on manufactured nanomaterials are established. More works seem to be required to gather more data on the nanomaterials released into environments and improve each step of the standard method proposed in this study.
