연구목적
- 종양에서 일어나는 glycolytic switch/oncogenic metabolism은 tumor growth 및 aggressiveness와 밀접한 연관성을 가진 중요한 현상임에도 불구하고 그 기작은 명확히 밝혀져 있지 않음.

연구목적
- 종양에서 일어나는 glycolytic switch/oncogenic metabolism은 tumor growth 및 aggressiveness와 밀접한 연관성을 가진 중요한 현상임에도 불구하고 그 기작은 명확히 밝혀져 있지 않음.
- 본 연구진은 선행연구결과, 발생 유전자인 homeobox gene Distal-less(Dlx)-2가 1) COXVIc 와 COX19의 발현 억제를 통하여 미토콘드리아 산소호흡을 억제하고 glycolytic switch를 유도함을 관찰하였으며, 2) TGF-β에 의한 glycolytic switch 현상에 관여함을 밝혔음. 또한 3) oncogenic metabolism 관련 Dlx-2 target 분자로 OAT, PDHB, PFKFB2, HK3, H6PD, SUCLG2, acetyl-CoA carboxylase, 17β-HSD, DGKI, glutaminase, HIF-1α 등을 발굴하였음.
- 본 연구에서는 Dlx-2와 Dlx-2 target 분자를 중심으로 TGF-β에 의한 oncogenic metabolism에서의 기능을 validation함으로써 항암치료 분자 target을 발굴하고 이를 이용하여 분자암치료 기술개발에 활용하고자 함.
연구방법
MCF-7, MDA-MB-231 등 다양한 암세포에서 western blotting, real time RT-PCR, 형광현미경 분석 및 ChIP assay를 통해 EMT를 확인하고 기작을 조사하였음. 또한 mitochondrial respiration, COX activity 및 glycolytic switch, mitochondrial oxidative phosphorylation의 in situ 분석을 통해 glycolytic switch/mitochondrial repression을 확인하였음. 뿐만 아니라, human 암조직에서 target gene 발현을 조사하였으며 Xenograft mouse를 이용하여 target gene에 대한 in vivo validation을 수행하였음.
연구 결과
1차년도 연구결과 1) Dlx-2/Snail 신호전달계에 의해 TGF-β-induced EMT가 조절됨을 관찰하였으며, 2) 이 과정에서 oncogenic metabolism 관련 Dlx-2 target 분자로 GLS를 발굴하였음.
GLS 불활성화 및 glutamine metabolism 억제에 의해 TGF-β 및 Dlx-2/Snail-induced EMT가 조절됨을 밝혔으며, 또한 3) Dlx-2/GLS 불활성화 및 glutamine metabolism 억제가 p53-micro RNA 발현 조절을 통해 Snail 발현을 억제함으로써 EMT를 억제함을 밝혔음. 뿐만 아니라 4) multicellular tumor spheroid(MTS) model system에서 Dlx-2/GLS 불활성화는 EMT 및 암세포 성장을 억제함을 확인하였음.
2차년도 연구 결과 1) GLUD1, GOT1/2, ME1, IDH1/2 불활성화를 통해 glutamine 대사를 억제 시 TGF-β에 의한 EMT가 억제됨을 확인하였음. 반면 GLS2는 영향을 미치지 않음을 관찰하였음.
또한 2) oncogenic metabolism 관련 Dlx-2 target 유전자인 GLS/PFKFB2/H6PD/ACACB를 불활성화 시 TGF-β에 의한 EMT 및 glycolytic switch, mitochondrial repression 현상이 억제됨을 밝혔음. 뿐만 아니라 3) Human 암조직 breast, colon 및 ovarian cancer tissue의 다양한 stage에서 Dlx-2, GLS, Snail, PFKFB2, H6PD, ACACB 발현이 증가되어 있는 반면, p53, Snail-targeting miRNAs, COXVIc은 발현이 감소되어 있음을 관찰하였음. 4) Dlx-2 target 분자를 대상으로 유전자 발현조절 및 관련 신호전달 기전을 분석한 결과 PFKFB2는 Dlx-2-glutamine metabolism-Snail cascade를 통해 발현이 조절되며, H6PD는 Snail에 의해 부분적으로 조절되고 ACACB는 Dlx-2-glutamine metabolism을 통해 조절되지만, Snail-independent pathway를 통해 조절됨을 확인하였음.
3차년도 연구결과 1) 2 차년도에 in vitro 상에서 Dlx-2 target 분자(GLS1, PFKFB2, H6PD, ACACB)의 불활성화가 tumor progression에 미치는 영향을 확인하였으므로 3차년도에는 in vivo에서 그 영향을 확인한 결과 동물모델에서 GLS1/PFKFB2/H6PD/ACACB 불활성화에 의해 Xenograft mouse의 tumor growth 및 lung metastsis가 억제됨을 확인함으로서
GLS/PFKFB2/H6PD/ACACB가 tumor growth 및 metastasis에 중요하게 작용함을 밝혔음. 또한 2) Dlx-2에 의한 oncogenic metabolism 억제물질 screening system을 구축하기 위해 먼저 in vitro 상에서 TGF-β에 의한 EMT에 미치는 GLS1, PFKFB2, ACACB inhibitor의 농도별 영향을 확인하였으며, 3) 동물 모델에서 기존 임상 항암제로 알려진 GLS inhibitor인 BPTES를 처리한 결과 Xenograft tumor 성장 및 metastasis가 억제됨을 밝혔음. GLS, Dlx-2, Snail를 대상으로 protein structure 분석 및 억제 물질에 대한 target site를 결정하는 등 Dlx-2에 의한 oncogenic metabolism 억제후보물질을 도출하였음.
기대효과
- TGF-β-Dlx-2에 의한 glycolytic switch/oncogenic metabolism 기작에 대한 연구는 새로운 암 연구 분야를 개척하는 작업으로, 암발생 기작의 규명에 기여할 것으로 기대하며, 많은 후속 연구를 파생시킬 것으로 기대함.

- Unlike normal cells, cancer cells exhibit glycolytic switch/oncogenic metabolism that contribute to tumor growth and aggressiveness. However, its mechanism remains largely unknown.
- We previously observed that homeobox gene Distal-less(Dlx)-2 1) regulates expression of COXVIc and COX19, induces mitochondrial repression and glycolytic switch, 2) and may be invovled in TGF-β-induced glycolytic switch. 3) We also previously identified OAT, PDHB, PFKFB2, HK3, H6PD, SUCLG2, acetyl-CoA carboxylase, 17β-HSD, DGKI, glutaminase, HIF-1α as target genes of Dlx-2-induced oncogenic metabolism.
- Based on these results, we will investigate the role(s) of Dlx-2 and Dlx-2 target genes in TGF-β-induced oncogenic metabolism to improve our understandings of tumor development and progression and to develop molecular targets of cancer therapy.
Methods
We investigated EMT and the molecular mechanism by western blotting, real time RT-PCR, and immunofluorescence in various cancer cells, including MCF-7 and MDA-MB-231, and glycolytic switch/mitochondrial repression by analyzing mitochondrial respiration, COX activity and glycolytic switch, mitochondrial oxidative phosphorylation in situ. We also investigated target gene expression in diverse human cancer tissues and established xenograft model for in vivo validation of target genes.
Results
■ The first year: 1) We found that Dlx-2/Snail cascade is implicated in TGF-β-induced EMT, 2) and identified GLS1 as target genes of Dlx-2-induced oncogenic metabolism. We also found that GLS1 shRNA and glutamine metabolism inhibition prevent TGF-β- and Dlx-2/Snail-induced EMT/glycolytic switch, 3) and shRNA for Dlx-2 or GLS1, and glutamine metabolism inhibition decreased Snail mRNA levels through p53-dependent upregulation of Snail-targeting microRNAs, that results in inhibition of EMT. 4) In addition, using multicellular tumor spheroid(MTS) model system, we found that shRNA for Dlx-2 or GLS1 suppress MTS culture-dependent EMT and growth.
■ The second year: 1) We found that glutamine metabolism inhibition through shRNA for GLUD1, GOT1/2, ME1, IDH1/2, but not GLS2, prevent TGF-β-induced EMT, 2) and shRNA for Dlx-2 target genes (GLS, PFKFB2, H6PD, ACACB) prevent TGF-β-induced EMT/glycolytic switch/mitochondrial repression. 3) We also observed that expression of Dlx-2, GLS, Snail, PFKFB2, H6PD, and ACACB are increased, while expression of p53, Snail-targeting miRNAs, and COXVIc are decreased in breast, colon and ovarian cancer tissues at different stages. 4) In addition, we established regulatory mechanism of Dlx-2 target genes: expression of PFKFB2 is regulated by Dlx-2-glutamine metabolism-Snail cascade, H6PD is partly regulated by Snail, whereas ACACB is regulated by Dlx-2-glutamine metabolism in a Snail-independent manner.
■ The third year: Because in the second year, we examined effects of shRNA for GLS/PFKFB2/H6PD/ACACB on tumor progression in vitro, we performed an in vivo validation study in the third year. 1) First, we found that shRNA for GLS/PFKFB2/H6PD/ACACB suppressed xenograft tumor growth and metastasis, indicating the important role of Dlx-2 target genes (GLS, PFKFB2, H6PD, ACACB) in tumor growth and metastasis. 2) To establish the screening system for inhibitor of Dlx-2-induced oncogenic metabolism, first, we examined the effects of inhibitor for GLS1, PFKFB2, ACACB on TGF-β-induced EMT in vitro. 3) Next, we found that BPTES, a GLS inhibitor, suppresses xenograft tumor growth and metastasis in vivo. Finally, we analysed protein structure of GLS, Dlx-2 and Snail, and identified their target site to contribute to development of new drug that target oncogenic metabolilsm.
Expected Contribution
- Molecular studies on TGF-β-Dlx-2-mediated regulation of glycolytic switch/oncogenic metabolism will not only discover the potential mechanism for tumor progression but also improve follow-up research.
- In the long run, we expect that our study on oncogenic metabolism may contribute to development of molecular cancer therapeutics and new drug candidates.
