연구의 목적 및 내용

연구의 목적 및 내용
본 연구에서는 혈관의 안정화를 야기하는receptor tyrosine kinase인 Tie2의 활성화 조절 기작을 Tie2의 세포외부위의 구조변화의 측면에서 연구하고자 한다. 즉, Tie2의 작용제인 Angiopoietin1(Ang1)과 길항제인 Angiopoietin2(Ang2)의 Tie2 활성화 정도 차이가 어디에서 기인하는지 구조적으로 규명하여, 이들의 신호전달로 인한 신생혈관형성이나 염증 반응 시 혈관투과성의 조절이 이루어지는 메카니즘을 규명하고자 한다. 이렇게 규명된 Tie2의 세포외부위의 구조변화와 활성 조절의 관계를 활용하여, Tie2 길항제인 Ang2를 작용제로 변화시켜 혈관의 안정성 및 혈관 투과성을 감소시킬 수 있는 방법을 고안하고 이 방법을 질병 모델에서 검증하고자 한다. Tie2 단백질의 세포외부위에는 세포내부의 kinase 도메인의 자발적인 다량체 형성을 억제하는 부위가 있다는 선행 연구를 바탕으로, 리간드인 Ang1은 Tie2 결합시 Tie2 세포외부부위의 구조변화를 야기하여 Tie2를 활성화 시킬 것을 예상하였다. 따라서 본 연구에서는 Tie2 단백질의 구조변화를 측정할 수 있는 FRET 기반 실험법을 개발하고 이를 이용하여 Ang1과 Ang2에 의한 Tie2 단백질의 구조 변화를 예측하고자 하며, 이를 바탕으로 해당 분야의 오래된 질문인, 두 리간드가 서로 다른 기능을 하는 이유에 대한 해답을 명쾌하게 제시하고자 한다.
연구결과
1) Fluorescence Resonance Energy Transfer (FRET)을 이용한 구조 변화 측정법 개발 및 2) Ang1/Ang2에 의한 Tie2 구조 변화 측정
Tie2의 N 말단에 tag된 ACP에 특이적으로 반응하는 녹색의 형광 시그널과 적색 형광으로 표지한 세포막 사이의 FRET 시그널을 실시간으로 관찰함으로써 두 형광 간, 즉 Tie2의 N 말단과 세포막 사이의 거리를 측정하는 기법을 확립한 뒤, 작용제인 Ang1과 길항제인 Ang2에 의한 Tie2의 구조변화를 관찰하였을 때, FRET efficiency가 Ang1에 의해서는 감소하고, Ang2에 의해서는 증가하는 것을 확인하였다. 이는 곧, Ang1에 의해서는 Tie2의 N말단이 보다 더 세포막과 멀어짐을, 그리고 Ang2에 의해서는 세포막과 더 가까워짐을 예측할 수 있는 결과이다.
3) 단일 분자 영상화 (single-molecular imaging)를 통한 Tie2의 구조 변화 예측
입사소각 X선 산란법을 통해 (포항공과대학 가속기 연구소에 있는 SAXS laser beam을 이용하여) Tie2의 세포외 부위 (Tie2-ECD)와 Tie2_ECD-Ang2_RBD의 복합물 구조 모두 folding되어 있는 구조를 확인할 수 있었고, 또한 Tie2-ECD의 1/4이나 되는 크기의 리간드가 결합하였음에도 불구하고 오히려 더 구부러진 듯한 구조를 확인할 수 있었다. 이는 곧, 길항제인 Ang2에 의하여 Tie2의 구조가 보다 구부러지게 됨으로 세포막과 보다 가깝게 되어지는 구조를 갖게 된다고 예측할 수 있는 결과이다.
연구결과의 활용계획
제안된 연구는 신생혈관형성에서 핵심적인 역할을 하고 있는 Tie2의 새로운 신호전달 기작에 대한 이해가 주된 연구 목표이다. 또한 본 연구는 생물학적 현상의 작용 기작 규명에 그치지 않고, 연구 결과를 바탕으로 기존 치료제에 비해 보다 우월한 성능을 가진 치료제 개발의 실마리를 제공하는 것을 목표로 하고 있다. 따라서 본 연구는 Tie2 biology의 보다 심도 있는 이해를 가능하게 할 뿐 아니라, 본 연구실과 제약사와의 협력을 통해 혈관의 불안정성이 하나의 원인이 되는 암, 염증, 혈관성 안과질환 등의 실제적인 치료제의 개발까지 가능하게 할 수 있을 것으로 기대된다.

The proposed research will study the mechanism of activation of Tie2, a receptor tyrosine kinase that causes stabilization of blood vessels, in terms of structural changes of Tie2. In other words, the difference of activation mechanism of Tie2 between Angiopoietin1 (Ang1), an agonist of Tie2, and Angiopoietin2 (Ang2), an antagonist, is structurally identified and the regulation of vascular permeability upon angiogenesis or inflammation responses caused by the signaling pathway. Using the relationship between the structural changes of the extracellular domain of Tie2 and the regulation of activity, we would devise and verify a method to reduce the stability and permeability of blood vessels by changing Ang2.
Based on a previous study, the ligand Ang1 induced structural changes in the extracellular region of Tie2, resulting in activation of Tie2, suggesting that there is a structural hindrance on extracellular region of Tie2 that inhibits the formation of spontaneous multimerization of the kinase domain. In this study, we have developed FRET-based method to measure the structural change of Tie2 protein and predict the structural change of Tie2 protein by Ang1 and Ang2 using this method. Based on this, we want to present a clear answer to why the two ligands function differently.

By measuring in real time the FRET signal between the green fluorescence signal and the red fluorescence-labeled cell membrane that specifically react with the ACP tagged at the N-terminus of Tie2, the distance between the two fluorescent sites, ie, the N-terminus of Tie2 and the cell membrane , We found that the FRET efficiency was decreased by Ang1 and increased by Ang2 when the structural change of Tie2 was observed by the agonist Ang1 and the antagonist Ang2. This suggests that the N terminal of Tie2 is more distant from the cell membrane than that of Ang1, and that it is closer to the cell membrane than that of Ang2.
3) Prediction of structural change of Tie2 by single-molecular imaging
The SAXS laser beam at the Accelerator Research Institute of Pohang University of Science and Technology showed that both the extracellular domain of Tie2 (Tie2-ECD) and the complex structure of Tie2_ECD-Ang2_RBD were folded, moreover ligand bounded-structure showed more bent structure, although ligand has a size of 1/4 of that of the Tie2-ECD. That is, it can be predicted that the structure of Tie2 becomes more bent due to the antagonistic agent Ang2, so that the structure becomes closer to the cell membrane.
Expected Contribution
The main research objective of the proposed study is to understand the novel signaling mechanism of Tie2, which plays a key role in angiogenesis. In addition, the present study aims to provide a clue to the development of therapeutic agents with superior performance over existing therapeutic agents based on the results of research rather than identifying the mechanism of action of biological phenomena. Therefore, this study not only enables a deeper understanding of Tie2 biology, but also cooperates with our laboratories and pharmaceutical companies to develop practical therapies such as cancer, inflammation and vascular ophthalmic diseases, which are one cause of vessel instability.
