□ 연구의 목적 및 내용
금을 기반으로 해서 만들어진 발광하는 나노닷은 이제 겨우 주목을 받기 시작했음에도 불구하고, 금 나노닷의 합성과 성질을 이해하기 위한 눈부신 발전이 있어오고 있다. 그러나, 금 나노닷을 기반으로 하는 형광 물질은 아직 경쟁력을 갖추지 못하고 있다. 금 나노닷의 화학적, 광물리학적 성질을 향상시키기 위한 탐구는 여전히 진행 중 이다. 그러나, 금 나노닷이 여러 영역에서 발광 할 수 있을지 혹은 발광 양자수율이 극적으로 향상이 될 수 있을지는 아직 명확하지 않다. 이 프로포절에서, 매우 밝고, 안정한, 작

□ 연구결과
1. 금 나노닷은 인시츄로 세포 내 뿐만 아니라 짧은 펩타이드를 통해서 수용액에서 합성: 적색 발광을 지니고, 큰 스토크 이동을 가짐. 7%의 양자수율, μs라이프타임, 낮은 발광 밝기. 그러나, 4℃ 수용액 상태에서 1년 이상의 우수한 선반수명을 지님.
2. 펩타이드로 보호되어 만들어진 금 나노닷의 합성 및 그 광물리학적 특성연구
3. 나노닷의 구조와 그들의 광학적 특성사이의 관계 연구: 산소는 금 나노닷의 형성을 촉진하고 있으나 결정적인 요인이 아님을 밝혀냄. 시스테인은 금 나노닷을 안정화 시키기 위해 우수한 작용기 그룹이나 없어서는 안 될 그런 것은 아니었음. 친수성의 금나노닷을 만들기 위하여 물과 글리세롤을 함께 사용하는 것이 좋은 용매임을 밝혀냄.

6. 금의 촉매활성 연구와 에칭도움에 의한 나노닷 생성과 같은 금 관련 물질에 대한 연구: 수용액 내에서 나노입자 표면과 반응물 사이의 정전기적 상호작용이 촉매 역할에 있어 보다 결정적인 역할을 하고 있음을 입증.
□ 연구결과의 활용계획
우리 그룹은 최초로 펩타이드로 보호된 금 나노닷의 합성과 그 광물리학적 특성에 관한 체계적인 연구를 하였다. 이런 나노닷은 생체적합성 조건에서 우수한 안정성을 가지며, 기타 많은 다른 금속을 기반으로 하는 나노닷의 발광 물질을 압도한다. 게다가, 금나노닷의 발광 세기는 그 나노닷을 둘러싸고 있는 마이크로 환경의 점성도에 민감하고, 이는 단단한 혹은 점성적인 환경이 금 나노닷의 발광을 촉진함으로 금 나노닷에 장점을 제공한다. 이런 모든 특성들은 금 나노닷이 생체환경 내에서 광학적 인비보 이미징내에서 세포막의 단단함 혹은 점성도의 센서 혹은 프로브로 사용 할 수 있는 가능성을 제시하고 있다.

Even though luminescent nanodots made of gold just started to attract attention, there has been significant progress in understanding the syntheses and properties of gold nanodots. However, fluorophores based on gold nanodots are not so competitive yet. Exploration is still underway to improve their chemical and photophysical properties. It is potential but not clarified that gold nanodots can emit at varied wavelength as other nanodots do, as well as that the luminescence quantum yield can be dramatically improved. In our proposal, we tried to develop new protection groups for gold nanodots to obtain highly bright, safe, stable and small size gold nanodots, and investigate the relations between nanodot sizes, structures, and photophysics of nanodots, with a final goal of competitive biological applications. Our research will greatly broaden the new fluorophore tools for optical imaging as well as biological sensoring.
□ Result
We have systemically investigated the synthesis and photophysical properties of peptide-protected gold nanodots. 1. gold nanodots can be generated not only in cell in situ, but also under the protection of short peptides in aqueous solution. These gold nanodots emits red light, with a large Stokes' shift. A 7% quantum yield and microsecond lifetime indicate a low brightness of the gold nanodot emission. However, the shelf lifetime of such nanodots is outstanding, more than a year in aqueous solution. 2. the synthesis and photophysical properties of peptide-protected gold nanodots. 3. Studying on the links between the nanodots structure and their properties. Oxygen can promote the formation of gold nanodots, but not critical. Cysteine is an excellent functional group, but not indispensible, to stabilize gold nanodots. Both water and glycerol are good solvents for the preparation of hydrophilic gold nanodots. 4. Peptide-protected gold nanodots are surprisingly stable in cell culture medium, such as DMEM in physiological condition. 5. To improve the synthesis efficiency of gold nanodots, we prepared gold nanodots in reverse micelles. Different from that in aqueous solution, gold nanodots in reverse micelles are sensitive to temperature. The lower the temperature, the higher the emission intensity. Either increasing the rigidness of water pool by lowering the water/oil ratio of the reverse micelles, or increasing the viscosity of the aqueous solution by adding glycerol, increases the emission intensity of gold nanodots. Such a phenomenon suggests that a rigid, or viscous environment promoted the emission of gold nanodots. 6. We also undertook research on gold-related materials. We demonstrated that electrostatic interactions between the nanoparticle surface and reactants in solution showed a more predominant impact on the catalytic activity.
□ Expected Contribution
We believe our research was the first comprehensive study on the synthesis and photophysical properties of peptide-protected gold nanodots. It showed excellent stability in physiological condition, surpassing many metal-based luminescence imaging materials. Moreover, the emission intensity of gold nanodot is sensitive to the viscosity of the microenvironments surrounding the nanodots, offering further merits to the gold nanodots. They are potentially useful for designing probes or sensors of viscosity or membrane rigidness in physiological optical in vivo imaging.
