1. 민들레의 항염증 활성 규명
민들레 잎과 뿌리의 주요 구성성분을 분석한 결과, Vit C는 잎이 뿌리에 비해 다소 높은 함량을 나타내었고, 베타-카로텐은 잎에 10.8 mg/100 g의 함량을 나타낸 반면 뿌리에는 0.5 mg/100 g으로 함량이 매우 낮은 것으로 나타났으며, 총 식이섬유 함량은 뿌리의 식이섬유 함량이 다소 높았다. 총폴리페놀과 총플라보노이드는 잎이 17627.3 mg/100 g과 13421.7 mg/100 g으로 뿌리에 비해 월등히 높은 함량을 나타내었다. 민들레 잎과 뿌리의 아미노산 조성을 비교한 결과, 잎의 경우 glutamic acid, arginine, lysine, serine 등의 함량이 높았고, 뿌리는 glutamic acid, lysine, glycine, aspartic acid 등의 함량이 높았다. 무기질은 잎이 뿌리에 비해 높은 무기질 함량을 나타내었고, 잎의 경우 K, Ca, P, Mg 등이, 뿌리의 경우에는 K, P, Na, Ca 등의 함량이 높은 것으로 나타났다.
민들레 잎과 뿌리로부터 70% MeOH 추출물과 용매별 분획을 제조한 후 NO, PGE₂, cytokines(TNF-α, IL-1β, IL-6) 등의 생성에 대한 저해율을 측정함으로써 이들의 항염증 활성을 탐색하였다. LPS로 유도된 RAW 264.7 cell의 NO 생성에 대한 민들레 잎과 뿌리의 추출물과 분획의 저해효과를 조사한 결과, 70% MeOH 추출물과 n-BuOH, 물 분획은 NO 생성억제 효과가 없었지만 chloroform과 ethyl acetate 분획은 큰 억제효과를 나타내었다. 민들레 잎과 뿌리의 추출물과 분획의 PGE₂ 그리고 TNF-α, IL-1β, IL-6 등의 cytokines 생성에 대한 저해 효과에서도 NO 생성 억제효과와 동일한 경향을 나타내었다. NO, PGE₂ 및 cytokines의 생성을 크게 억제하는 것으로 확인된 민들레 잎의 chloroform, ethyl acetate 분획의 iNOS와 COX-2의 발현에 대한 억제 효과를 western blot을 통해 확인한 결과, chloroform의 처리에 의해 iNOS와 COX-2의 발현이 농도의존적으로 저해되었다. Ethyl acetate 분획에 의해서도 iNOS의 발현이 농도의존적으로 감소된 것으로 나타났지만 COX-2의 발현은 크게 감소시키지 않았다. Pro-inflammatory mediators인 NO, PGE₂와 cytokines 그리고 NO와 PGE₂의 조절단백질은 외부 자극에 의해 MAPKs와 JAT-State system의 발현을 통하여 활성화된다. 민들레 잎의 MeOH 추출물과 chloroform, ethyl acetate의 분획이 MAPKs의 생성에 대한 영향을 확인하기 위해 LPS로 유도된 phosphorylated ERK1/2와 JNK1/2 MAPK의 발현을 western blot을 통해 확인한 결과, chloroform과 ethylacetate 분획을 처리한 macrophage의 phosphorylated ERK1/2와 JNK1/2의 발현이 농도의존적으로 크게 감소하는 것으로 나타나 민들레 잎의 chloroform과 ethyl acetate 분획의 처리는 MAPKs의 phosphorylation을 억제하여 염증을 매개하는 인자들의 생성을 저해하는 것으로 나타났다.
항염증 효과가 뛰어난 chloroform과 ethyl acetate 분획의 활성물질을 silica gel column chromatography, TLC와 prep-LC를 통해 정제하였고 LPS로 유도된 NO 생성 억제효과를 측정하였다. Silica gel chromatography를 통해서 분획물을 75:25의 chloroform과 MeOH의 혼합 용매로 용출시켜 NO 활성이 뛰어난 물질로 분리하였다. 이 물질을 다시 TLC를 통해 4개의 fraction으로 분리했고 이 중 높은 활성을 보인 1번째와 2번째의 fraction을 다시 silica gel column chromatography를 통해 8개의 fraction으로 분리하였으며, 높은 활성을 나타낸 2번째 분획물을 C18 column을 사용한 HPLC로 분석한 결과 하나의 주요 피크가 나타났다. 따라서 이를 prep-LC를 통해 분리해내어 LC/MS와 NMR을 통해 구조 동정을 하였다.
2. 민들레 항염증 활성물질의 소재화
민들레 잎으로부터 항염증 활성 물질을 분리하여 소재화하기에 적합한 민들레 잎의 적정 열수추출 및 농축조건을 조사하였다. 60~90°C의 추출 온도에서의 추출물의 특성을 조사한 결과, 60°C 추출물의 수율이 가장 높았고, flavonoid의 함량은 온도가 증가함에 따라 증가하였지만 항염증 활성은 감소하는 경향을 나타내었다. 가장 높은 항염증 활성을 나타낸 60°C 추출물의 유용성분의 추출을 증가시키기 위해 세포벽 분해 효소를 처리한 결과 민들레 잎에서 추출된 flavonoid 함량과 추출 수율이 1.5배 증가하고 항염증 활성도 감소하지 않는 것으로 나타나 세포벽 분해효소 처리가 항염증 활성물질의 추출증가에 상당한 영향을 미치는 것으로 나타났다. 또한 효소 처리에 의한 민들레 잎으로부터 추출한 항염증 활성물질을 농축시키기 위해 column chromatography를 실시한 결과, Amberlite XAD-2와 Sepabeads SP-850에 흡착된 물질 모두 항염증 활성이 매우 높은 것으로 나타났지만 Sepabeads SP-850에 흡착된 물질의 수율이 2배 이상 높았기 때문에 민들레 잎의 항염증 활성물질을 농축하는데 Sepabeads SP-850 resin이 훨씬 효과적이었다.
민들레 잎으로부터 분리한 항염증 소재의 피부염 및 관절염 유발 동물모델에서의 항염증 효과를 조사하였다. TPA와 croton oil로 실험동물의 귀에 염증을 유발하고 염증 유발 전과 후의 귀 두께를 측정하여 함염증 소재의 귀 부종 억제 효과를 측정하였다. TPA처리 염증모델에서 대조군인 indomethacin 처리군의 경우 귀 두께가 15.2 ㎛ 증가한 반면 항염증 소재 처리군은 처리량 1~100 mg일 때 귀 두께가 39.1~23.9 ㎛ 증가하였고 처리량이 증가할수록 농도의 존적으로 귀 두께가 감소하는 것으로 나타났다. Croton oil 처리 염증모델에서도 100 mg 농도에서 가장 큰 억제능력을 보였고 농도가 증가할수록 높은 효능이 있는 것으로 나타났다. 민들레 잎의 항염증 소재가 관절염 유발 모델에서의 항염증 효과를 검증하기 위해 염증 유발후 항염증 소재의 9일 동안의 투여에 따른 족 부종의 부피, squeaking, 체중 분배비율 등을 측정한 결과 항염증 소재 투여에 의해 족 부종 부피 등의 행동양식이 크게 개선되는 것으로 나타났으며, 투여량이 증가할수록 높은 효능을 나타내었다.
민들레 잎의 항염증 소재의 온도와 pH에서의 안정성을 확인하기 위하여 항염증 소재를 30~90°C, pH 3~11에서 처리한 후 LPS로 유도된 macrophage의 NO 생성 저해율을 측정한 결과 온도가 증가하여도 항염증 소재의 활성에 영향을 끼치지 않았지만 pH가 9 이상으로 높아지면서 활성이 감소하였다. 항염증 소재의 여드름균 (Propionibacterium acnes, Streptococcus epidermidis) 대한 항균성은 없었으며, 항산화성은 상당히 높은 것으로 나타났다. 민들레 항염증 소재의 독성 유무를 조사하기 위해 고농도의 단회 투여 및 장기복용에 의한 동물에서의 독성을 시험하였다. 고농도 (2000 mg/kg)의 단회투여에서 독성이 나타나지 않았으며, 장기복용에 따른 독성시험 결과 항염증 소재 투여에 의해 특정한 행동변화와 이상증상을 관찰되지 않았고, 혈액학적 및 혈액생화학적 검사에서도 이상이 발견되지 않았다. 부검결과 대부분의 장기에서는 이상이 발견되지 않았지만 항염증 소재 투여군의 신장에서 이상증상이 발견되어 신장에 대해서 조직병리학적 검사를 실시하였지만 이상이 없는 것으로 판명되었다.
3. 민들레를 이용한 기능성 제품 개발
민들레 음료 제조에 적합한 민들레 잎의 덖음조건을 확립하기 위하여 덖음처리에 의한 민들레 잎과 열수추출액의 이화학적 및 관능적 특성을 조사하였다. 열수추출액의 아미노태질소, 총폴리페놀 등이 크게 감소한 반면 환원당의 경우에는 생잎에 비해 더 많은 양이 추출되는 것으로 나타났다. 열수추출액의 향미묘사분석에 의해 떫은맛, 쓴맛, 풀냄새, 탄맛, 단내 등 5가지 관능적 특성이 도출되었고, 떫은맛, 쓴맛, 풀냄새가 처리구간 유의적인 차이를 나타내었으며, 관능적 기호도 측정 결과 덖음횟수가 증가할수록 색과 맛에 대한 기호도가 증가하여 종합적인 기호도도 크게 증가하는 것으로 나타났다. 적정 추출조건을 확립하기 위하여 4회 덖음처리한 민들레 잎을 60, 75, 90°C에서 열수추출하여 추출온도 및 추출횟수별 추출액의 이화학적 특성을 비교한 결과 모든 항목에서 추출온도에 따른 값의 차이가 거의 나타나지 않은 반면 추출횟수가 증가할수록 각 특성값은 크게 감소하는 것으로 나타났다. 민들레의 추출온도별 추출액에 대한 향미묘사시험을 통해 떫은맛, 쓴맛, 풀냄새, 탄맛, 구수한 맛 등 5가지 관능적 특성을 도출하였고 90°C 추출액은 쓴맛이 감소하고 구수한 맛이 증가하는 것으로 나타났다. 이상의 결과를 토대로 하여 민들레 음료 개발을 위한 적정 전처리 및 추출조건을 덖음온도 180°C, 덖음횟수 4회, 추출온도 90°C, 추출횟수 2~3회로 결정하였다. 민들레 음료 제조를 위한 추출액의 적정 음용농도를 결정하기 위하여 농축액을 희석하여 관능검사한 결과 0.3 °Bx가 적정 음용농도이었으며, 색이나 맛의 변화를 최소화하면서 음료의 맛에 크게 영향을 미치지 않는 안정제를 검토하였다. 낮은 음용농도 때문에 민들레 고유의 향미가 약한 것으로 나타나 이를 보완하기 위해서 향 첨가시험을 실시하여 국화향을 선발하였다.
민들레 항염증 소재를 이용하여 여드름 치료용 및 관절염 예방용 기능성 제품을 개발할 목적으로 함께 첨가할 생약재의 항염증 활성을 측정한 결과 오디, 인진쑥, 초피나무의 열수추출 물의 NO 생성 억제율이 우수하여 항염증 소재와 함께 첨가할 생약재로 선정하였다. 항염증 소재가 여드름균에 대한 항균효과를 나타내지 않았기 때문에 화장품, 연고 등의 여드름 환자용 제품을 개발하기 위해서는 여드름균에 대한 항균성을 갖는 항균제가 필요하다. 포도씨 추출물, 자몽씨 추출물, 파인-오렌지 등의 천연 항균제를 이용하여 여드름균에 대한 항균력을 측정한 결과 포도씨 추출물(GS-1000)이 가장 우수한 항균력을 나타내었다. 민들레 항염증 소재와 항염증 활성이 우수한 것으로 확인된 초피나무, 인진쑥, 오디 추출물 그리고 여드름균에 대한 항균력이 뛰어난 포도씨 추출물 (GS-1000)을 혼합하여 여드름 환자용 크림, 연고, 비누를 개발하였다. 민들레 항염증 소재와 초피나무, 인진쑥, 오디 추출물을 혼합하여 관절염 예방용 환과 과립차를 개발하였다.

1. Investigation on anti -inflammatory activity of dandelion
The nutrient components of leaves and roots of dandelions were analyzed, and leaves and roots showed differences for various characteristics. Leaves of dandelion had higher vitamin C, β-carotene, total polyphenol and flavonoid content compared to dandelion roots, but roots had higher dietary fiber content. According to the result of measurement of their mineral and amino acid composition, leaves of dandelion had high content in K, Ca, P, Mg in case of mineral, and glutamic acid, arginine, lysine, serine in case of amino acid, and roots of dandelion had high content in K, P, Na, Ca in case of mineral, and glutamic acid, lysine, glycine and aspartic acid in case of amino acid.
Dandelion leaves and roots were extracted with 70% MeOH and their extracts were fractionated sequentially with various solvents. And then anti-inflammatory activities of these extracts and fractions were determined by measuring inhibition on production of NO, PGE₂ and cytokines. The inhibition of LPS-induced NO production on RAW 264.7 macrophages of extract and its fractions from dandelion leaves and roots was evaluated. Conclusively, the 70% MeOH extract, n-BuOH and water fractions had little effect on inhibition of NO production, however, the chloroform and ethyl acetate fractions were most effective. The extracts and fractions from dandelion leaves and roots inhibited production of PGE₂, and cytokines such as TNF-α, IL-1β and IL-6 with the same tendency of inhibition of NO production. The chloroform and ethyl acetate fraction from dandelion significantly suppressed production of NO, PGE₂ and cytokines. Furthermore, the expressions of LPS-induced iNOS and COX-2 on RAW 264.7 macrophages of the fractions were detected by western blotting. As the results, the expressions of iNOS and COX-2 were remarkably decreased in a dose-dependent manner by treatment of the chloroform fraction. It was showed that the expression of iNOS decreased in a dose-dependent manner by the ethyl acetate fraction, but it slightly suppressed the expression of COX-2. Pro-inflammatory mediators such as NO and PGE₂, cytokines and regulating enzymes of NO and PGE₂ were activated depending on the external signals through the expressions of MAPKs and JAT state systems. Western blot of expressions of LPS-induced phosphorylated ERK1/2 and JNK1/2 MAPK was performed to determine whether the MeOH extract, chloroform and ethyl acetate fractions were related to the modulation of the activation of MAP kinases. As the results, the chloroform and ethyl acetate suppressed dose- dependently the induction of LPS-induced phosphorylation of ERK1/2 and JNK1/2. From this finding, it is assumed that the chloroform and ethyl acetate fractions in LPS-treated macrophages suppressed inflammatory mediators by inhibiting phosphorylation of MAPKs.
Bioactive compounds that increased anti-inflammatory activity were fractionated and separated from chloroform and ethyl acetate fractions by silica gel column chromatography, TLC on silica and prep-LC. The anti-inflammatory activity of each preparations was determined by measuring of LPS-induced NO production. Fractions were separated by eluting with chloroform and MeOH mixtures (75:25) from silica gel column chromatography, and the separated fractions showed increased anti-inflammatory activity. These fractions were more separated into 4 fractions by TLC. Among these 4 fractions, the first and second fractions that showed significantly increased activity were purified by silica gel column chromatography again. The second of 8 fractions from the silica gel column chromatography showed the highest activity. From this finding, the last second fraction was analyzed by HPLC using C18 column, resulting that the one major peak was detected. Finally, the compound of the major peak was separated by prep-LC, and then it was identified by LC/MS and NMR.
2. Materialization of anti-inflammatory compounds from dandelion
To materialize anti-inflammatory compounds from dandelion leaves, we studied optimal conditions of hot water extraction and concentration. We investigated the properties of extracts on various temperatures between 60 and 90 °C. As the result, extraction yields at 60°C were the highest as 39.2±1.3% and flavonoid contents were increased by heating. However, anti-inflammatory activities were remarkably decreased by increasing temperature. According to the results that the highest anti-inflammatory activities of water extract at 60 °C were demonstrated, cell wall degrading enzyme treatment was carried out to increase extraction yield of anti-inflammatory compounds. The enzyme treatment resulted in increased yields and flavonoids from dandelion leaves as 1.5 times and anti-inflammatory activities were not decreased. Also, column chromatography was used for concentration of anti-inflammatory compounds from extract of dandelion leaves treated with enzyme. It demonstrated that anti-inflammatory activities of adsorbed compounds on Amberlite XAD-2 and Sepabeads SP-850 were significantly increased. But yields of adsorbed compounds were increased over two times on Sepabeads SP-850 compared to Amberlite XAD-2. Thus Sepabeads SP-850 were more effective than Amberlite XAD-2 to concentrate the anti-inflammatory compounds of dandelion leaves.
To verify the anti-inflammatory activity of anti-inflammatory compounds from dandelion leaves, their in vivo anti-inflammatory effect in dermatitis and arthritis model was investigated. TPA-induced or croton oil-induced acute edema was developed in the mouse ears, and they were applied to both sides of inflamed ears. It was found that they could significantly reduce the ear swelling, compared to that of non-treated control. In the case of DA-100 treatment, its anti-inflammatory effect was comparable to that of indomethacin, a non-steroidal anti-inflammatory drug. In order to verify the anti-arthritic effect of anti-inflammatory compounds from dandelion leaves, several behavioral parameters such as paw volume, squeaking score, and weight distribution ratio were investigated in a carrageenan-induced arthritis rat model. Their therapeutic effect was observed on 9th day after the arthritis induction, as compared to saline-treated control group. Oral administration of them significantly alleviated apparent symptoms of paw volume, squeaking score, and weight distribution ratio in rats. Therefore, it could be concluded that topically applied anti-inflammatory compounds from dandelion leaves exhibited its potentials as a new drug candidate with an effective anti-inflammatory activity.
Inhibitory activity on NO production of LPS induced macrophage of anti-inflammatory compounds from dandelion leaves was determined to evaluate the stability of anti-inflammatory activity with temperature of 30~90℃ and pH of 3~11. Applied temperature did not affect, but pH above 9 reduced the activity. Antibacterial activity of anti-inflammatory compounds on acne-causing bacteria(Propionibacterium acnes, Streptococcus epidermidis) were also determined and its activity was not detected. Toxicity of anti-inflammatory compounds was determined with high dose one-time trial and for long-term trial with rats. One-time high dose(2000 mg/kg) tiral did not show any toxicity. Long term trial also did not show any symptom of specific behavior abnormality and had no concern from serum biochemical analysis data. Autopsy did not present any disorder from organs examined but kidney and kidney were also proven to be normal with histopathological observation.
3. Development of functional products using dandelion
To establish the roasting conditions of dandelion leaves for the manufacture of beverages, we investigated the physicochemical and sensory characteristics of dandelion leaves and their hot water extracts by roasting treatment. As the number of roasting times increased, the amino nitrogen and total polyphenol contents of the roasted dandelion leaf hot water extracts also decreased greatly, and their L-, a-, and b-values showed much lower values than those of the raw leaf extracts. However, a greater amount of reducing sugars was extracted in the roasted leaves than in the raw leaves, suggesting that the roasting process allows for easier extraction of the compounds in dandelion leaves. Five sensory characteristics, including astringent taste, bitter taste, green flavor, burnt taste, and sweet odor, were deduced through quantitative descriptive analysis of the hot water extracts. Among them, astringent taste, bitter taste, and green flavor showed significant differences between roasting treatments. The sensory evaluation results show that as the number of roasting times increased, the palatability of the hot water extracts increased greatly, in terms of color and taste.
In order to establish extraction condition of dandelion leaves, physicochemical properties of extracts of 4-times roasted dandelion leaves extracted with different extraction temperatures and extraction times were investigated. There was no difference in values of measurement items with different extraction temperatures, but as the extraction times increased, values of physicochemical properties decreased greatly. Five sensory characteristics, including astringent taste, bitter taste, green flavor, burnt taste, and savory taste, were deduced through quantitative descriptive analysis of the hot water extracts. Bitter taste of extracts decreased and savory taste of extracts increased with increase of extraction temperature. Taken together, it was concluded that roasting temperature of 180°C, roasting of 4 times, extraction temperature of 90°C and extraction of 2~3 times were optimum pretreatment and extraction condition for development of dandelion beverage. As a result of sensory test to determine concentration of extracts relevant to drink, 0.3 °Bx was optimum concentration. Additives of vitamin C, sodium ascorbate and sodium bicarbonate et al. were added to dandelion extracts to minimize the changes in color and taste of beverages. To complement the low intensity of natural dandelion flavor, various flavors were examined, and chrysanthemum flavor was selected.
Water extract of mulberries, mugwort, Zanthoxylum piperitum DC. had excellent inhibition activity on NO production and were most desirable subsidiary material for development of a functional food products effective on acne and arthritis with the anti-inflammative compounds from dandelion leaves. Cosmetics and ointment for acne patients were developed with grape seed extract(GS-1000) which were most effect antimicrobial compounds on acne bacteria among known natural antimicrobial agents and extracts from extract of Zanthoxylum piperitum DC., mugwort, mulberry. Pills and granule type products for arthritis patients were also developed with the anti-inflammative compounds from dandelion leaves and extract of Zanthoxylum piperitum DC., mugwort, mulberry.
