□ 연구목표
연구자는 우연히 25 mM Kcl이 상처 치유를 촉진하고, fibroblast에서 collagen의 생성을 촉진시킨다는 것을 알게 되었음.
이에 25 mM Kcl이 fibroblast에서 발현되는 potassium channel에 작용하여, fibroblast 내의 어떤 signal pathway를 통해서, collagen을 더 많이 생성시키는 지를 연구하기로 함.

즉 K channel activator를 이용하여 hypertrophic scar를 예방 및 치료하고, K channel inhibitor를 이용하여 자외선으로 인해 생성되는 주름을 개선할 계획임.
□ 연구내용
1. Kcl이 fibroblast의 collagen 생성을 증가시키는 signal pathway (ERK, NF-kB, Protein tyrosine kinase, JNK, PKC, PKA, NFAT) 조사
2. 발현량이 증가한 signal pathway에 관련된 단백질의 inhibitor (Gö 6983, Gö 6976, H-89, PD98059, SP600125, NFAT inhibitor, Bay 11-7082)를 fibroblast에 처리하여, collagen 생성량 (western blot analysis of procollagen, sircol collagen assay)을 봄.
3. Reporter gene (SBE, 3-TP, AP-1, 3-KB) analysis
4. Fibroblast에서 발현되는 potassium channel (TREK-1, TREK-2, TRAAk, TWIK-1, TASK-1, TASK-2) 의 존재를 RT-PCR, real-time PCR, western blotting으로 확인
5. K channel inhibitor (iberiotoxin, charybdotoxin)와 activator (NS1619)를 fibroblast에 처리하여 collagen 생성을 봄.
- 또한 특정한 signal pathway의 inhibitor를 처리하여, collagen의 생성량을 측정.
6. K channel activator를 이용한 hypertrophic scar의 예방 및 치료
- Rabbits의 귀에 연골이 노출되는 상처를 만든 후, 28일이 지나면, 비후성 반흔이 생김.
- NS1619 1 mM를 처리하여 hypertrophic scar의 예방 혹은 치료함
- 조직 (in vivo)에서 특정 signal patihway의 발현 양상
7. K channel inhibitor를 이용한 mouse model에서 생성된 주름의 개선
- 마우스의 등에 UVB를 조사하여 주름 모델을 만듦.
- Skin replica를 통한 주름 분석
- H&E와 Masson’s trichrome staining
□ 기대효과(응용분야 및 활용범위 포함)
Fibroblast의 potassium channel을 조정하여 collagen 합성을 조절할 수 있다면, 임상에서 획기적인 성과가 기대됨.
1. Collagen 생성 증진
Collagen의 합성을 증가시켜
- 상처 치유를 촉진시키고,
- 자외선에 의해 생성되는 주름을 방지하고,
- 나이가 들어 생기는 피부의 collagen 결핍으로 인한 피부 노화를 방지함.
이는 학문적 발달 뿐 아니라, 의약품과 화장품 등 기능성 제품의 개발로 이어져, 엄청난 이익 창출이 가능함.
2. Collagen 생성 억제
Collagen의 합성을 감소시킨다면, 임상에서 획기적인 성과가 기대됨. 예를 들면 Cirrhosis (liver), Idiopathic pulmonary fibrosis (lungs), Endomyocardial fibrosis or Old myocardial infarction (heart), Keloid or hypertrophic scar (skin), Scleroderma/systemic sclerosis (skin, lungs) 등에 사용될 수 있을 것임.

We firstly will measure the change of the collagen amounts after treating the protein inhibiting (or activating) the related signal pathway
We will also examine the potassium channel expressed in fibroblast.
Moreover, we will research whether K channel inhibitor can increase the collagen production after treating in fibroblast, and K channel activator can inhibit the collagen production.
With the result of studies, we will try to apply it in treatments for illness caused by the abnormality of collagen production by regulating K channel.
In other words, we are planning to use K channel activator for preventing and treating hypertrophic scar, and K channel inhibitor for the treatment of the UV-induced wrinkles.

If we control the collagen production in fibroblast using the potassium channel modulator, various and epoch-making outcomes will be expected.
1. Enhancement of collagen production
The increase of collagen production in fibroblast
- enhance wound healing
- prevent or reduce UV-induced wrinkles
- prevent skin aging caused by collagen deficiency.
Our study may make a contribution to not only the improvement of medical knowledge, but also the development of manufactures such as cosmetics, which finally can produce enormous profits.
2. Suppression of collagen production
If our study can clarify the fact that the potassium channel modulator decrease the collagen production, the modulator can be used in various disease, such as Cirrhosis (liver), Idiopathic pulmonary fibrosis (lungs), Endomyocardial fibrosis or Old myocardial infarction (heart), Keloid or hypertrophic scar (skin), Scleroderma/systemic sclerosis (skin, lungs).
Our study can produce enormous profits in terms of industry and medical knowledge.
3. Researching new signal pathway may lead to breakthrough on inventing a new inhibitor or activator of signal pathway related to collagen production and development of medical knowledge.
