본 연구에서는 이산화탄소를 이용하여 미세조류를 배양시켜 이산화탄소를 저감하는 동시에 유용물질을 생산하는 생물공정을 개발하는 것을 목적으로 연구를 수행하였고 효과적인 연구의 진행을 위하여 고효율의 성능을 가지는 반응기와 생산성이 증가된 형질전환 균주를 개발하였음. 세부적으로 미세조류의 광합성 대사조절 최적화를 위한 유전자와 astaxanthin의 생산성을 강화하기 위한 유전자를 확보하였음. 미세조류의 형질전환을 위하여 agrobacterium 및 gene gun을 이용한 고효율의 형질전환법을 개발하였음. 본 연구에서는 이산화탄소를 이용하여 미세조류를 배양시켜 이산화탄소를 저감하는 동시에 유용물질을 생산하는 생물공정을 개발하는 것을 목적으로 연구를 수행하였고 효과적인 연구의 진행을 위하여 고효율의 성능을 가지는 반응기와 생산성이 증가된 형질전환 균주를 개발하였음. 세부적으로 미세조류의 광합성 대사조절 최적화를 위한 유전자와 astaxanthin의 생산성을 강화하기 위한 유전자를 확보하였음. 미세조류의 형질전환을 위하여 agrobacterium 및 gene gun을 이용한 고효율의 형질전환법을 개발하였음. Perfusion 공법을 통하여 최대 0.32 g/L · day의 고농도의 biomass 생산성을 기록 하였음. 효율적이고 경제성 있는 생물공정의 개발을 위하여 고분자 필름을 원료로 하는 bubble column 유형의 반응기를 개발하였고 순차적으로 scale-up을 진행하여 25L의 배양 부피를 가지는 반응기를 제작하여 실내 환경과 옥외 환경 (100L)에서 시운전을 실시하였음. 개발된 반응기의 성능을 평가한 결과 200 mg/L 이상의 astaxanthin을 획득할 수 있었으며 현재는 산업체 배가스를 이용한 생물공정 개발을 지속적으로 연구하고 있음. 회수 및 분리공정 개발로 인하여 biomass의 회수율은 95%, 유용물질의 회수율은 85%를 달성하여 연구를 성공적으로 진행하였음.

Microalgae are currently cultivated for production of various commercial products around the world in several dozen small-to medium-scale production systems, producing a few tens to a several hundreds of tons of biomass annually. The main algae genera currently cultivated photosynthetically for various nutritional products are Spirulina, Chlorella, Dunaliella and Haematococcus. Total world production of dry algal biomass for these algae is estimated at about 10,000 tons per year. About half of these productions take place in mainland China, with most of the rest in Japan, Taiwan, U.S.A., Australia and India, and a few small producers in some other countries. Biological production of valuable materials by microalgae has more productivity and price competitiveness than traditional methods such as chemical synthesis.
A unicellular green microalgae, Haematococcus pluvialis, has been focused as a microbial source of astaxanthin, which has been suggested as a food supplement for humans. H. pluvialis accumulates the highest level of astaxanthin. Astaxanthin has property as antioxidative materials and high value of 2,000 $/kg. Its unique structure, which incorporates hydroxyl and keto endings on each ionone ring, provides astaxanthin with outstanding antioxidant activity, compared to closely related carotenoids with antioxidant properties, such as β-carotene, zeaxanthin, and lutein. Therefore, H. pluvialis has high potential for application in the nutraceutical, pharmaceutical, cosmetics, food, and feed industries.
First of all, we studied to develop genetic transformation method for H. pluvialis. The Haematococcus cell synthesizes a high yield of astaxanthin using CO₂ in a photoautotrophic culture without contaminant heterotrophs: however, it takes too long to induce astaxanthin production. In this study, a highly-photosensitive mutant strain was obtained by conventional random mutagenesis to shorten induction time. Sensitivity to photoinhibition in this mutant was raised by a partial lesion in the photosystem II (PSII) of photosynthesis, thereby prompting a change in cellular morphology as well stimulating carotenogenesis (astaxanthin production). As a result, the concentrations of cell biomass and astaxanthin dramatically increased by 27% and 53% under strong light and 40% and 137% under moderate light, respectively. This Haematococcus mutant would be useful for the economical production of astaxanthin to reduce the light energy expenses in a photoautotrophic culture system, particularly in light-insufficient area, Second, to increase the production of useful materials and fixation of carbon dioxide in a microalgae, it is necessary to develop a cell that contains high astaxanthin concentration. The genetic manipulation was performed to increase the expression level of carotenoid biosynthetic genes in a microalge. In-vitro transcriptions were conducted by endogenous and heterologous promoters to select a suitable promoter for outstanding expression in H. pluvialis. It was identified that HSP70A + RBCS2 promoter induced the most powerful expression of H. pluvialis gene. Agrobacterium-mediated transformation was used to secure manipulated cells and continuous expression of GFP was also confirmed.
The strategies for high density of astaxanthin production by Haematococcus pluvialis can be divided into two categories. One is high density cell cultivation and the other is efficient induction of astaxanthin. In this study, perfusion culture process and stepwise high light illumination were introduced for high cell density cultivation and astaxanthin induction, respectively. 2.41 g/L of green vegetative Haematococcus cell was obtained by perfusion process. This was 3.01 and 1.63 times higher cell production than batch and fed-batch processes, respectively. The astaxanthin concentration produced by stepwise high light illumination was 1.26 times higher than maximum astaxanthin concentration by constant light induction. The combined effects of these two strategies had made inspiring results. Finally, 12.3 g/L of biomass was produced after astaxanthin induction and its productivity was 0.32 g/1/d that exceeded the goal of this project (0.2 g/L/d). Moreover, 602 ㎎/L of remarkable astaxanthin concentration was produced.
In this study, to achieve high cell density and price competitiveness in astaxanthin production, we have developed a disposable photobioreactor and successfully applied it to indoor & outdoor environment. The photobioreactor was made of cast polypropylene and polyethylene terephtalate (CPP) instead of traditional glass or other expensive materials, so our photobioreactor would have merits of low cost and easy installation. The photobioreactor system was optimized in terms of various parameters including size of sparger, height of diameter and air flow rate, medium composition, light intensity and photoperiodism. Moreover, the photobioreactor was applied to both indoor and outdoor environment. H. pluvialis cells that can produce astaxanthin were cultured in photobioreactors of various working volume (8 L, 15 L, and 25 L). Indoor and outdoor conditions were maintained at 20-25 ℃ temperature, under 0~300 ㎛ol·photon/m²·s light intensity. 2 % CO₂ gas was constantly supplied to each photobioreactor. Astaxanthin, which is a product of micro algae, H. pluvialis, was accumulated to 200 mg/L astaxanthin in 15 L photobioreactor and specific growth rate was 0.073. On the other hand, the astaxanthin accumulated up to 130 ㎎/L astaxanthin in 25 L photobioreactor during 76 days. Further, optimization of photobioreactor was conducted and the higher productivity of astaxanthin was achieved, compared to that in control cultivation. These results indicate that our photobioreactor can be applied both to indoor and outdoor environment and scale-up of photobioreactor could be successfully achieved.
After microalgae cultivation in outdoor & indoor condition, we tried to avail flue gas in our bioprocess system. H. pluvialis was cultivated with stepwise increasing concentration of flue gas in a week for adaptation to high level CO₂ and Nox. At first, 20 % flue gas and 80 % air were used and then flue gas concentration was increased up to 50 %. Because the H. pluvialis has two stage, green vegetative stage and red cyst stage, the experiments were conducted under each two stage conditions, green vegetative stage and red cyst stage. At the red cyst stage, there was no big difference between flue gas and pure CO₂. The dry cell weight was obtained almost 2 g/L and astaxanthin was 70 mg/L for 66 days. However, at the green vegetative stage, the induction of H. pluvialis was inhibited by fuel gas.
For development of efficient process, we developed downstream techniques for isolation of astaxanthin and separation of biomass. Fitration, centrifuge and precipitation method was considered for collecting biomass of photosynthetic microalgae, H. pluvialis. Precipitation was the lowest energy costing and was selected as optimal method in collecting biomass. The optimum crushing and extracting solvent was searched to collect carotenoids from biomass. Homogenizer (for 30 min) and ethyl acetate was the best option for crushing and extracting carotenoids from biomass. Carotenoids were saponificated to enhance astaxanthin in several conditions and were analyzed by HPLC to find the optimum saponification method. Alkali and enzyme saponification were considered as the candidates. By HPLC result, Alkali saponification was a better option for saponification. To gain the final valuable product astaxanthin, open column chromatography (Silica and Sephadex resin) and Prep-HPLC were studied for the isolation of astaxanthin. As a result, Prep-HPLC showed the highest content and purity of astaxathin.
In conclusion, the proposed technologies in this thesis are expected to be helpful for the enhancement of astaxanthin productivity in upstream and downstream process of algal technology. This study indicates that this approach can support the scale-up of photobioreactor, and it possesses a great potential to application of biological CO₂ reduction process. Additionally, strain development by genetic tranformation will increase productivity of biomass and fixation of carbon dioxide. Thus, our novel biological process like photobioreactors, separation, isolation process and genetic modified microalgae could solve environmental problem such as the green house effect because it has many advantages in views of price competitiveness, easiness of scale-up and superiority of photobioreactor.
