연구의 목적 및 내용
◆ 전사매개인자인 Med1은 세포의 증식과 세포주기 조절에 중요한 역할을 담당함. 또한 여러 암에서 단백질의 발현이 따라 암 전이와 생장에 영향을 미치나 단백질 발현 조절 기전 및 안정화, 활성화에 관한 연구가 부족함. 본 연구를 통하여 Med1이 BAP1, USP39등 다양한 탈유비퀴틴 효소에 의해서 단백질의 안정화가 조절됨을 확인함.

◆ Med1 결합단백질인 전사인자 Sp1은 세포의 증식과 분화에서 중요하게 작용함. Sp1은 sumolyaton에 의한 ubiquitination에 의해서 단백질 발현 감소와 불활성화로 인해 암전이를 증가시킴. Sp1단백질은 Med1과 함께 암세포에서 BAP과 USP39에 의해서 단백질의 발현이 조절됨.
◆ 세포의 항상성에 필요한 단백질의 회전율, 활성화, 세포내 분포등 다양한 기전에 관여하는 탈유비퀴틴효소인 DUB를 통한 암세포에서 암의 생성 및 전이에 관여하는 단백질의 발현 및 활성화를 조절할 수 있음을 확인함.
◆ 본 연구를 통하여 암 발생 및 전이에서 중요하게 작용하는 Med1/Sp1의 발현을 조절할 수 있는 탈유비퀴틴 효소인 DUB를 선정하였으며, DUB에 의해서 조절되는 Med1/Sp1의 전사활성화 기능 및 암 전이에서의 중요한 역할을 담당하고 있음을 확인함.
연구결과
◆ Med1/Sp1의 탈유비퀴틴을 조절하는 탈유비퀴틴 효소 (DUB) 선정 및 in vitro 기능 검증
암세포에서 Med1/Sp1의 탈유비퀴틴을 조절할 수 있는 탈유비퀴틴 효소를 동정하기 위하여 50여 가지의 DUB library를 통하여 Med1/Sp1 단백질의 유비퀴틴을 저해하는 DUB인 BAP1과 USP39를 선정하여Med1/Sp1 단백질의 안정화 및 활성화 조사한 결과, BAP1과 USP39에 의해서 Med1/Sp1 단백질의 발현이 안정화 되며, 전사활성화 능이 증가함을 확인할 수 있음.
◆ 암세포에서 Med1/Sp1을 조절하는 DUB의 역할 및 기전 연구
선정된 DUB인 BAP1를 통한 Med1/Sp1 단백질이 안정화 및 활성화 조절이 암세포의 생장이 증가하며 세포 사멸이 감소됨을 확인하였으며, 전사활성 능이 증가함으로써 Med1/Sp1 타켓 유전자의 발현이 조절됨을 확인함. 또한 세포의 이동성과 침윤성 능을 확인한 결과, Med1/Sp1의 단백질의 안정화 됨으로 암세포의 이동성 및 전이능이 감소됨을 확인할 수 있음. 또한 Med1/Sp1의 단백질의 인산화는 단백질의 발현의 조절 뿐만 아니라 전사 활성화에 중요한 역할을 담당하고 있으며, 단백질의 인산화를 통해 Med1/Sp1의 표적 유전자릐 발현 및 세포 생장이 조절됨을 확임함.
◆ in vivo상에서 DUB를 통한 Med1/Sp1의 역할 규명
Med1/Sp1의 단백질 발현은 세포주기 과정동안 인산화 되어 세포의 생장 및 세포 사멸에 관여함을 밝혔으며, 현재 면역세포를 이용한 암 치료제 개발에서 중요한 T세포의 전사활성화에 중요하게 작용하는 전사인자 RORg의 전사활성화를 Med1에 의해서 조절됨을 확인함.
연구결과의 활용계획
최근에는 표적 함암제 개발에 이어 면역함암제 치료제 개발이 세계적으로 활발히 이루어지고 있으며, 우리나라의 경우 4 종류의 면역함암제 치료제가 개발되어 시판되고 있으며, 해마다 면역 함암제 개발 시장이 커지고 있음. Med1/Sp1은 암 세포의 성장 및 표적유전자의 전사 활성화를 조절함으로써 암 세포의 생장 및 전이능을 조절할 수 있으며, 또한 면역 세포 특히 T세포의 분화에 관여하는 전사인자 RORg의 전사활성화를 조절할 수 있음을 확인하였음. 이러한 결과를 바탕으로 DUB에 의해 조절되는 Med1/Sp1을 통한 암세포에서의 표적 유전자와 면역세포에서의 분화 및 전사 활성화를 동시에 관여할 수 있는 항암제 개발을 위한 바탕을 제공함.

Med1, a transcription factor, plays an important role in cell proliferation and cell cycle regulation. In addition, the expression of protein in various cancers affects cancer metastasis and growth, but lacks research on regulation, stabilization and activation of protein expression. This study confirmed that Med1 stabilizes protein stabilization by various deubiquitin enzymes such as BAP1 and USP39.
Med1 binding protein, Sp1, plays an important role in cell proliferation and differentiation. Sp1 increases the metastasis by decreasing protein expression and inactivation by ubiquitination by sumolyaton. The expression of Sp1 protein is regulated by BAP and USP39 in cancer cells together with Med1.
In this study, DUB, a deubiquitin enzyme that regulates the expression of Med1 / Sp1, which plays an important role in cancer development and metastasis. UDB was selected and the transcription activation function of Med1 / Sp1 regulated by DUB and its important role in cancer metastasis.
Result
In order to identify deubiquitin enzymes capable of modulating deuterubiquitin of Med1 / Sp1 in cancer cells, BAP1 and USP39, which are ubiquitin inhibitors of ubiquitin of Med1 / Sp1 protein, were selected through more than 50 DUB libraries to stabilize Med1 / Sp1 protein As a result, activation of Med1 / Sp1 protein was stabilized by BAP1 and USP39, and transcription activation activity was increased.
I confirmed that the stabilization and activation regulation of Med1 / Sp1 protein through the selected DUB, BAP1, increased the growth of cancer cells and decreased cell death, and confirmed that the expression of Med1 / Sp1 target gene was regulated by increasing transcriptional activity. In addition, the mobility and invasiveness of the cells were confirmed, and it was confirmed that the stabilization of Med1 / Sp1 protein reduced the mobility and metastatic potential of cancer cells. In addition, the protein phosphorylation of Med1 / Sp1 plays an important role not only in regulating protein expression but also in transcription activation.
