바이오필름은 미생물의 군집체로 세포외 다량체 기질(extracellular polymeric substances, EPS)이라는 분비된 기질(self-secreted matrix) 속에 막의 형태로 형성된다. (Stewart and Franklin, Nat. Rev. Micriobiol., 2008) 3차원적 구조물인 바이오필름은 미생물이 생활할 수 있는 미세 환경(microenvironment)을 구축하고 미생물을 보호하는 역할을 하며 EPS 내에서의 확산(diffusion) 현상을 통해 미생물이 살아가는데 필요한 산소 및 영양분을 공급한다 (Flemming et al., J. Bacteriol., 2007). 바이오필름 형성과 제어를 위해서는 먼저 이 미세 환경을 구축하고 제어하여야 한다.
본 과제는 바이오필름의 형성 제어 및 방제를 가능하게 하는 시스템 구현을 위한 기술 개발을 목표로 한다. 이에 따라 본 연구팀은 먼저 물리적으로나 화학적으로 제어가 용이하며 적은 양으로도 실험이 가능한 미세 유체 시스템(microfluidic system)을 디자인 및 개발하고자 한다. 미세 유체 시스템 디자인 및 개발과 미세 유체 시스템 내의 바이오필름 형성법 개발을 시작으로 미세 유체 시스템 최적화, 물리적, 화학적 요소의 적용으로 바이오필름 형성 제어 및 방제 방법을 개발하려 한다. 이 미세 유체 시스템을 사용하여 보다 현실적인 3차원적 바이오 필름의 형성에 필요한 다양한 물리적 혹은 화학적인 요소들을 밝히고, 더 나아가, 주어진 환경에서 바이오필름이 어떻게 형성될지 체계적으로 예측할 수 있기를 기대한다. 자연에서 바이오필름이 다양한 환경에서 존재하듯이 다양한 조건에서 바이오 필름을 형성시키고 그 과정을 이미징(imaging)을 통해 모니터링(monitoring)한 후 3차원적 바이오필름 분석을 통해 효과적이고 체계적인 바이오필름의 형성, 제어, 제거 방법을 개발하고자 한다.
미세 유체 시스템 내에서의 바이오필름의 형성 제어 및 방제를 가능하게 하는 시스템 구현을 목표로 진행중인 본 연구는 현재 1년 4개월차 연구를 완료 중이며, 처음 1년차 동안의 연구내용으로 제안되었던 미세 유체 소자 디자인 개발 및 바이오필름 형성법 개발을 진행하였다. 미세 유체 소자의 디자인은 기존의 평평한 바이오필름 형성 모사를 위한 디자인을 기반으로 하이드로겔을 사용하는 삼차원 바이오필름 미세환경 모사를 가능하게 하도록 고안하였으며 이를 바탕으로 2차년도에는 유동과 화학 요소 분포 제어를 위한 디자인을 최적화 했다. 마스터를 제작하여 미세 유체 소자 공정과정을 확립하였고 공정된 미세 유체 소자에 박테리아를 주입하여 바이오필름 형성의 초기 단계인 미세 유체 소자 내에서의 세포 흡착 현상이 나타나는 것을 확인하였다. 그리고 Agarose gel과 박테리아를 함께 배양하여 약제의 영향을 조사하였다. 마지막으로, 바이오필름에 대한 모델을 새롭게 제시하였다. 향후 바이오필름의 투과성을 고려한 생장 및 저감 과정 모델링을 실험과 병행함으로 상호보완적인 바이오필름의 삼차원적 형성 제어 및 방제법 개발을 목표로 한다.

In recent years, researchers have noted the importance of bacterial biofilm research for its application in environmental and medical fields (Kohusova et al., Environ. Monitoring & Assess., 2011). Since bacteria can readily adapt to the given environment, biofilm can be formed in natural environment or anywhere that is hydrated (Flemming, Water Sci & Tech., 1993). For example, biofilm may be formed in industrial setting such as water pipeline, waste water treatment system, or in human organs. Because these undesired formation of biofilm may lead to the negative effects ranging from clogging to infections, it is important to understand the governing mechanisms in the formation of biofilm process, in order to be able to control, disperse and prevent it (Palmer et al., J. Indust. MIcrobiol. Biotech., 2007).
Furthermore, the delicate control of biofilm formation process is necessary to use biofilm during the organic filtering process in wastewater treatment. Therefore, being able to fine tune the formation and suppression of biofilm can lead to the development of numerous applications, depending on the necessary conditions.
Biofilm is a colony of microorganisms in a thin film form, and it is enclosed in a self-secreted matrix called, extracellular polymeric substances (EPS) (Stewart and Franklin, Nat. Rev. Micriobiol., 2008). The microenvironment of biofilm exists as a three dimensional structure, and it provides protection and support for the microorganisms of a biofilm through diffusion of oxygen and nutrients in the EPS (Flemming et al., J. Bacteriol., 2007). Proper mimicking of the microenvironment as well as its control is crucial for forming and controlling the biofilm.
This research aims to develop a system that enables biofilm formation control and prevention. Thus, we plan to design and develop a microfluidic system that allows a precise control of physical and chemical cues. The project may start with designing the microfluidic system and forming biofilm in microfluidics, and will continue in optimizing application of physical and chemical cues for biofilm formation control. Doing so, different physical and chemical cues responsible for the formation of biofilm will be identified, and the final goal of predicting the formation of biofilm in a given environment may be achieved. Moreover, we will be able to form biofilm under various conditions, similar to how biofilm exists in a diverse environment naturally. Finally, the entire process will be monitored and analyzed through different imaging techniques such that effective and systematic methods for biofilm formation, removal and prevention control will be developed.
The objective of this study is to materialize a microfluidic system for control and prevention of biofilm. We developed a microfluidic device and protocols for biofilm formation until now. The design of the microfluidic platform is devised to mimic 3D microenvironment for biofilm, and we optimized the design for that purpose. This system controls flow and chemical distributions. Moreover, we confirmed bio-adhesion of bacteria, the initial stage of formation of biofilm, in the device. After that, we seeded bacteria with agarose gel, and investigate the efficacy of antibiotics. Finally, we introduced a new model for biofilm. Our future works are to conduct experiments and to develop models for control and prevention strategy of biofilm.
