□ 연구의 목적 및 내용 혈관은 혈관내피세포외에도 혈관주변세포, 성상세포, 미세교세포 등 다양한 세포들로 구성되어 있으며, 따라서 병적 상황, 특히 당뇨 상황에서 이들 혈관을 구성하는 세포들간 상호작용에 대한 분자기전을 이해하는 것이 본 과제의 중요한 목적이다. 이를 위해 본 과제에서는 1) 혈관내피세포와 혈관주변세포 및 성상세포의 상호작용 규명하고, 2) 혈관내피세포와 미세교세포의 상호작용 규명, 3) 혈관내피세포와 혈관주변 미세환경의 상호작용 기전에 근거한 타겟 타당성 검증 연구를 수행하였다.

□ 연구결과 1) 혈관내피세포와 혈관주변세포 및 성상세포의 상호작용 규명 • 고혈당 조건에서 혈관내피세포, 성상세포, 미세교세포 cytokine 발현을 분석하여, 혈관내피세포 유래 Ang2 cytokine 발현 증가 동정하였다. • Ang2에 의한 성상세포 및 혈관주변세포의 세포사멸 현상을 확인하고, 관련 신호전달기전을 동정하였다. • 당뇨유발 마우스 망막에서 성상세포의 세포사멸과 혈관투과도와의 관련성을 분석하였다. 2) 혈관내피세포와 미세교세포의 상호작용 규명 • 고혈당에 노출된 미세교세포에서 IL-6 cytokine 발현 증가를 동정하였다. • IL-6에 의해 혈관내피세포 투과도 증가현상과 관련 신호전달기전을 동정하였다. • LPS등에 의해 미세교세포 분화도 변화에 의한 혈관투과도 변화 분석하였다. • 당뇨유발 마우스 망막의 미세교세포 역할 분석하였다. 3) 혈관내피세포와 혈관주변 미세환경의 상호작용 기전에 근거한 타겟 타당성 검증 • 동정된 cytokine의 작용기전과 이를 타겟으로 한 항체, 억제제 등을 사용하여 실제 당뇨유발 마우스 망막에서 혈관손상 및 혈관투과도 변화를 분석하였다. • 성상세포 및 미세교세포의 변화와 혈관손상 및 혈관투과도 변화의 관련성 분석하였다.
□ 연구결과의 활용계획 본 연구과제 수행 중 고혈당에 노출된 미세교세포는 IL-6 사이토카인 분비를 통해 혈관내피세포의 STAT3신호전달을 활성화시키고, 이것이 혈관투과도와 연관이 있다는 결과를 얻었으며, 이는 혈관내피세포 STAT3의 새로운 타겟으로써의 중요성을 보여주는 중요한 결과로써, 혈관내피세포의 특이적 기능에 대한 연구가치가 높다고 판단되며, 이에 후속연구를 진행할 예정이다.

□ Purpose&contents Blood vessels are composed of various cells such as vascular endothelial cells, pericytes, astrocytes, and microglia. Therefore, it is important to understand the molecular mechanism of interaction between cells constituting these vascular microenvironment in vairous pathological situations. In this study, we investigated 1) the interaction between vascular endothelial cells and pericytes and astrocytes, 2) the interaction between vascular endothelial cells and microglia, 3) the target validation based on the cellular interaction in pathological conditions.
□ Result 1) the interaction between vascular endothelial cells and pericytes and astrocytes • we analyzed the cytokine expression in endothelial cells, astrocytes, and microglia in hyperglycemic conditions. • we found the Ang2-induced cell death in astrocytes and pericytes and identified the relating signaling pathway. • we analyzed the relationship between the cell death and the vascular permeability in the diabetic mouse retinas. 2) the interaction between vascular endothelial cells and microglia • we found the increased expression of IL-6 cytokines in microglia exposed to hyperglycemia. • we found the IL-6-induced increase in vascular leakage and its related signaling pathways. • we analyzed the changes in vascular permeability due to changes in microglia differentiation by LPS • we analyzed the role of microglia in diabetic mouse retinas 3) the target validation based on the cellular interaction in pathological conditions • we analyzed the mechanism of the identified cytokines and the effect of neutralizing antibody or inhibitor for the identified targets on the vascular injury in the diabetic mouse retinas. • we analyzed the relationship between astrocytes and microglia and changes in vascular permeability in mouse retinas.
□ Expected Contribution In this study, microglial cells exposed to hyperglycemia activated STAT3 signaling in vascular endothelial cells through IL-6 cytokine expression, and this was associated with vascular permeability. As an important results of its significance as a target, it is supposed to study the specific function of STAT3 in vascular endothelial cells in the further studies.
