연구목표
본 연구의 최종 목적은 한국, 스웨덴 및 미국 연구팀들의 전문분야를 적용한 공동연구를 통해 폴리글루타민 확장 퇴행성 뇌질환을 유발하는 단백질을 연구하여 병의 발병 기작을 이해하고, 병을 진단하는 바이오마커 및 치료제를 개발하는데 있다.
연구개발내용
- Huntingtin 단백질의 구조 및 기능 연구: 헌팅턴씨 병은 헌팅턴에 폴리글루탐산이 확정되어 일어나는 병으로, 헌팅턴씨 병의 원인이 되는 Huntingtin 단백질의 정상상태와 병 유발상태의 3차원 구조의 도메인을 동정하고, 초저온 전자현미경을 이용
연구목표
본 연구의 최종 목적은 한국, 스웨덴 및 미국 연구팀들의 전문분야를 적용한 공동연구를 통해 폴리글루타민 확장 퇴행성 뇌질환을 유발하는 단백질을 연구하여 병의 발병 기작을 이해하고, 병을 진단하는 바이오마커 및 치료제를 개발하는데 있다.
연구개발내용
- Huntingtin 단백질의 구조 및 기능 연구: 헌팅턴씨 병은 헌팅턴에 폴리글루탐산이 확정되어 일어나는 병으로, 헌팅턴씨 병의 원인이 되는 Huntingtin 단백질의 정상상태와 병 유발상태의 3차원 구조의 도메인을 동정하고, 초저온 전자현미경을 이용하여 구명하였다. 최근에 규명된 Huntingtin-HAP40 와 비교하여 Huntingtin 단백질이 매우 flexible 한 conformation을 가진다는 것을 규명하였고, 정상상태인 Q23 과 병 유발 상태인 Q78 의 구조적 차이점을 규명하였다. 2단계에서는 N-terminal 도메인의 폴리그루탐산 확장이 C-terminal 도메인의 구조를 변화시키는 기작을 규명할 계획이다. 또한, Huntingtin 단백질과 결합하고 병 유발에 중요한 단백질을 동정하고 이들과의 결합구조를 규명할 계획이다.
- Huntingtin variant 의 therapeutic 이용을 위한 연구: Huntingtin 단백질은 세포내에서 exon12가 없어진 형태의 Huntingtin (Httn d12)이 만들어지는데, Httn d12 에는 caspase site 가 비활성화 되어 있어 toxic 한 헌팅턴의 N-terminal fragment를 만들지 못한다. 생체네 Huntingtin을 이러한 Huntingtin variant를 유전자 조작기법으로 만드는 방법 (ProQR therapeutics)을 이용한 헌팅틴 치료방법을 개발하는 가운데 Huntingtin d12와 Huntingtin 분자 동등성을 물리화학적 방법을 이용해 연구하였고, 2단계에서는 Huntingtin d12 의 3차원 초저온 현미경 구조를 규명하여 Huntingtin variant를 이용한 신약개발의 기틀을 마련할 예정이다.
- Huntingtin PTM 과 그 기능 연구: Huntingtin 의 PTM 은 헌팅턴 병의 pathogenesis 에 중요한 것으로 알려져 있다. 본 연구에서는 Huntingtin 을 인산화 시키는 kinase 들과 이들의 타깃을 동정하였고, 이 중에서 Ser2550 의 인산화는 Huntingtin단백질의 안정성을 조절하는 것으로 밝혀내었다. 2단계에서는 Huntingtin 의 다른 PTM 이 Huntingtin 의 기능과 헌팅턴 병의 pathogenesis 에 관여하는 역할을 규명할 계획이다.
- Huntingtin 의 기능을 조절하는 물질 개발: 헌팅턴 병을 유발하는 Q78 형태의 Huntingtin의 기능을 modulation 하는 aptamer를 개발하고 이들의 효과를 연구하였다. 이 aptamer 는 Huntingitn 의 병을 조절하는 기반 기술로 사용가능하다.
- Ataxin-1 과 CIC 구조 및 기능연구: SCA1 폴리그루타민 병을 유발하는 Ataxin-1 과 Ataxin-1 결합단백질이면서 pathogenesis 에 중요한 CIC 의 초기 전자현미경 구조를 규명하였고, CIC 이 DNA 에 결합하는 형태의 x-선 구조를 규명하였다. 또한 Ataxin-1 과 CIC 의 결합을 제어하는 물질 개발을 수행중에 있다. 이와 더불어 Ataxin-1 PTM을 프로파일링을 수행하였다. 2단계에서는 지속적으로 Ataxin-1/CIC 의 3차원 구조를 규명하고 PTM 의 역할에 대해 연구할 계획이다.

To understand the pathogenesis of the polyglutamine (polyQ) tract neurodegenerative diseases, and to develop diagnosis and therapeutics for the diseases, we will investigate the causative proteins focusing on Huntington’sdisease (HD) and Spinocerebellar Ataxia type1 (SCA1), by collaborating among the groups having different expertise from Korea (KPI), Sweden (FPI) and United States (co-FPI).

- Structural and functional analysis of Huntingtin protien: To investigate the structures of Huntingtin, we mapped the domain within Huntingtin. In addition, we determined cryo-EM structures of normal Huntingtin Q23 and disease Huntingtin Q78 showing that the polyQ expansion at the N-terminal induces conformational change of the C-terminal domain. Based on the recent atomic resolution structure of Huntingtin-HAP40 and our own structures, we concluded that the Huntingtin is flexible structures adoping various conformations. In the phase II, we will continue to identify proteins interacting with Huntingtin and pursue to determine the structures of the complexes.
- Characterization of Huntingtin variant (Httn d12) for therapeutic development: Httn d12 is a natural Huntingtin variant, which does not produce toxic N-terminal fragment due to the absence of caspase6 site.
- Investigation of the roles of Huntingtin PTM: As the post-translational modifications (PTMs) of Huntingtin have been implicated in HD pathogenesis, we identified PTMs and their responsible enzymes for Huntingtin protein and investigated the roles of the PTMs. From this study, we identified that Ser2550 is phosphorylated by PKA and the phosphorylation regulates the staibility of Huntingtin protein. In phase II, we will continute to investigate the roles of other PTMs in HD pathogenesis.
- Developing aptamers modulating Huntingin activity: To develop means to modulate the disease specific activity of Huntingtin, we developed an aptamer that specifically binds to Huntingtin Q78 and abrogate the PRC2-stimulating activity of Huntingtin Q78.
- Structural and functional analysis of Ataxin-1/CIC complex: To investigate the molecular basis of SCA1 pathogenesis, we investigated the structures of Ataxin-1/CIC complex. First, we determined initial low resolution structure of Ataxin-1/CIC and the crystal structure of the DNA binding domain of Ataxin-1/CIC with the target DNA. Second, we investigated the PTMs of Ataxin-1. In phase II, we aim to determine high resolution structures of Ataxin-1/CIC and the function of this complex in the context of SCA1 pathogenesis.

Expected Contribution
The outcomes of the proposed research will provide structural and biochemical information of the polyQ disease proteins providing a platform for developing means to diagnose and to cure the diseases. In addition, this research will provide mechanistic understanding on the effect on polyQ expansion on the causative proteins. Furthermore, we will provide therapeutic targets and means to diagnose and to cure the diseases. Furthermore, this GRL will consolidate the collaboration among Korea, Sweden, and United States for understanding neurodegenerative diseases.
