연구개발성과
- 제대혈 유래 중간엽줄기세포를 전처리하여, 치료 및 표적 효능 물질이 증가하도록 만듦. 이로부터 동물모델에서 급성 척수손상 치료효능이 우수한 나노베지클을 제작하였음. 나노베지클의 특성 및 저온 저장 가능성을 확인하였고, 치료 및 표적 효능 물질은 각각 50% 이상 증가하였음.

- Final objective: In this study, we developed exosome-mimetic nanovesicles derived from umbilical cord blood mesenchymal stem cells, which possessed enhanced targeting efficiency and therapeutic efficacy, for spinal cord injury as a off-the-shelf therapeutic agent.
- Conventional therapeutic agents (e.g. stem cells, chemical reagents) have showed insignificant effects, on the other hand, our nanovesicles showed remarkable therapeutic effects and can be repeatedly used for patients immediately after hospitalization.
Results
- In this study, we pretreated umbilical cord blood-derived mesenchymal stem cells to increase therapeutic and targeting factors. Then we produced nanovesicles from pretreated mesenchymal stem cells which possesssed remarkable therapeutic efficacy for acute spinal cord injury. We determined the characteristics of nanovesicles and the therapeutic/targeting factors increased more than 1.5 folds. and confirmed the possibility of the storage at low temperature.
- With the treatment of nanovesicles, the apoptosis of neuronal cells and the inflammation of macrophages were decreased, whereas the angiogesis of endothelial cells were increased in vitro. In addition, the targeting efficiency of nanovesicles to inflammatory endothelium were increased by pretreatment.
- We successfully made the animal models for acute spinal cord injury. The pretreated nanovesicles exhibited the 2.5-fold increased targeting efficiency compared to non-treated nanovesicles. In addition, pretreated nanovesicles showed the attenuation of inflammation, the decrease of the neuron apoptosis, and increased angiogenesis, which finally resulted in enhanced neurobehavioral ability of spinal cord injured mice.
Expected Contribution
- Technical aspect: Showing a new paradigm as an advanced concept of therapeutic agent. Providing enhanced targeting efficiency and therapeutic efficacy compared to conventional therapeuitc agents. Our nanovesicles can be applied to an expanded area including ischemic diseases (myocardial infarction and stroke).
- Social Aspect: Improving the quality of life of the patient who will be suffered from permanent impairment by spinal cord injury.
- Industrial Aspect: Reducing social and economical expenses. Our nanovesicles will be a valuable brand-new therapeutic agent for spinal cord injury treatment.
