본 연구는 최근 새롭게 출현하는 신·변종 위해우려 미생물에 대한 국가 차원의 체계적 대응 연구의 일환으로써 설계되었다. 국내 미관리 위해우려 주요 세균·바이러스·기생충의 식품 중 검출 시험법을 확립하고, 국내 유통식품과 생산환경 관련 오염실태를 조사하여 발생 가능한 새로운 식중독을 사전에 예방할 수 있는 대응책을 마련하고자 하였다. 이에 본 연구진은 위해세균, 위해바이러스, 위해기생충 전문 연구팀이 세부 연구기관으로 구성되어, 1) 국내 유입 가능 신종 미생물을 조사하고, 2) 연구 우선순위를 결정, 우선 연구대상에 대한 연차별 탐색조사를 기획하였으며, 3) 식품 대상 표준시험법이 부재한 우선 연구대상 미생물 종별 검출 시험법을 확립하여, 4) 국내 유통 식품과 생산-유통단계별 오염실태를 분석하였다. 또한, 5) 국내외 위해정보를 심층분석한 위해기술서, 6) 개발·검증한 시험법의 이해와 활용이 용이하도록 시험법 가이드라인을 개발하고, 7) 후순위 연구대상 미생물에 대해서는 간이형 위해정보카드를 작성하여 위해소통 강화에 기여하고자 하였다.
연구결과로 국내 유입 가능한 위해우려 미생물의 위해정보 제공현황 DB를 구축하고, 국내 전문가들의 의견을 수렴하기 위해 1) 국제기구와 EU 등 선진국 6개국 이상, 총 21개 기관에서 제공하는 262건의 미생물 위해정보 제공현황을 분석하여 식품 매개 위해우려 미생물 85종의 후보군을 확보하였으며, 최근 이슈와 식품 검출 필요 종 7종(세균 2종, 바이러스 2종, 기생충 3종)을 주관부서 협의 하에 추가하여 연구대상 후보 미생물 총 92종을 선정하였다. 2) 국내 감염의학, 수의학, 식품안전 분야별 전문가 설문을 통해 연구대상 후보군의 우선 관리대상에 대한 의견을 수렴하였다. 또한, 3) 위생 선진국과 위생 취약국 각 4개국(총 8개국)의 학계, 기관의 식품안전 전문가 설문을 통해 식품 중 위해우려 미생물에 대한 종별 중요도와 신규 위해우려 미생물에 대한 의견을 수렴하고 국가별 관리현황과 전통 식문화로 인한 위험 식품군을 파악하였다. 4) 개발한 연구 우선순위 결정 체계도로 후보군의 우선순위를 도출하여 13종(세균 8종, 바이러스 1종, 기생충 4종)의 우선 연구대상 1순위 목록을 확보하였으며, 5) 위의 1)-4) 일련의 과정을 통해 발굴한 후보군 중 주관부서와 협의 하에 최종 시험법 개발대상 12종[세균 5종(Coxiella burnetii, Streptococcus dysgalactiae, Vibrio mimicus, Streptococcus suis, Vibrio alginolyticus), 바이러스 3종(Tick-borne encephalitis virus, Severe fever with thrombocytopenia syndrome virus, Crimean-Congo hemorrhagic fever virus), 기생충 4종(요코가와흡충, 간질, 거대간질, 톡소포자충)]을 확정하였다. 6) 해당 연구대상 12종에 대하여 식품 전처리법, 핵산 추출법을 포함한 분자생물학적 진단법과 배지배양법을 개발하여 정성·정량법을 확립하였다. 종별 검출빈도가 높거나 식중독 발병사례가 있는 식품을 대상으로 인위접종 검증실험을 진행하고, 식약처 표준절차에 따라 실험실 간 교차검증을 수행하여 국가 표준시험법으로서의 토대를 마련하였다. 7) 종별로 오염 가능성이 큰 식품군을 선별하여 국내 유통 농·축·수산물 2,789건에 대한 모니터링을 실시하였고, 연구대상 12종 중 4종을 제외한 8종(C. burnetii, S. dysgalactiae, V. mimicus, S. suis, V. alginolyticus, Severe fever with thrombocytopenia syndrome virus, 요코가와흡충, 톡소포자충)이 자연산 굴 등 가열 섭취해야하는 농·축·수산물에서 검출되었다. 8) 연구대상 식품의 생산환경(낙농장, 미나리 재배지, 자연산 굴채집장)에서 총 1,093건의 탐색조사를 실시하였으며, 자연산 굴 채집장의 해수에서 톡소포자충이 검출되었다. 9) 연구대상 12종에 대하여 전문 학술정보를 포함하는 전문가용, 안전한 소비를 위한 간략한 위해정보를 포함하는 소비자용 각 1건씩 총 24건의 위해기술서를 개발하고, 10) 현장 활용이 용이한 형태의 시험법 가이드라인을 개발하였다. 마지막으로, 연구우선순위가 낮게 책정된 미생물 76종을 대상으로 간이형 위해정보카드를 제작하였다.
종합적으로 본 연구는 국내 유입이 가능한 신종 위해우려 미생물에 대한 국가 관리체계를 강화하기 위해 국가 관리 대상으로 제안될 수 있는 식품 매개 인체병원성 미생물에 관한 심층적인 연구를 수행함으로써 미예측 식중독 사고의 사전 예방에 이바지하고자 하였으며, 불규명 미생물 식중독의 원인 규명률 증진과 역학조사에 이용하고자 하였다. 또한, 국가별 신종 위해우려 미생물 관련 위해정보를 수집하고 맞춤형 DB를 구축, 유통 식품과 식품생산환경 중 위해우려 미생물의 오염실태 조사 결과를 확보하여 국가적 차원의 위해정보 빅데이터를 구축·확립하였다. 또한, 식품 중 존재하는 신종 위해우려 미생물(세균, 바이러스, 기생충)을 신속하고 정확하게 검출할 수 있는 시험법을 개발하였고, 우선관리의 필요성이 있는 위해우려 미생물에 관한 전문가용과 소비자용 위해기술서, 시험법 가이드라인, 간이형 위해정보카드 개발을 통해 연구성과를 활용·확산하고 지식을 보급하는데 기여하고 있다. 국내·외 전문가 대상 설문조사, 국내 유통 식품에 대한 위해우려 미생물의 검출, 인체 병원체 대상 식품 중 검출법 신규 개발 등의 연구수행 내용을 기반으로 학술성과 발표를 활발하게 진행함으로써 관련 분야에서 국제적으로 선도할 수 있는 결과를 확보하였다.

This study was designed as a part of a systematic study on the national level to respond to newly emerging food-borne pathogens. The purpose of this study was 1) to establish detection methods for bacteria, viruses and parasites in food that are not currently regulated in Korea, and 2) to prepare countermeasures in advance to prevent possible food poisoning by these novel microorganisms by examining their prevalence in domestically distributed food and food production environment. To accomplish these goals, research teams of which the main research fields are pathogenic bacteria, virus, or parasites joined to form a consortium for this project. Also, survey expert team (Gallup Korea) joined to gather domestic and oversees experts’ opinions on the relevant issues. The research contents of this study were as follows: 1) investigation on the potentially pathogenic microorganisms which are not nationally managed as food-borne pathogen, but could enter our country, 2) prioritization of the microorganisms in order to design annual research plans, 3) establishment and validation of the detection methods for potentially hazardous microorganisms with high priority, and 4) analysis on the prevalence of domestically distributed foods and food production-distribution environments. In addition, 5) risk profiles including the in-depth risk information on the target microorganisms, 6) test guidelines for the developed detection methods, and 7) the simplified risk information cards for low-risk microorganisms were developed to strengthen the risk communication.
To establish a DB of risk information on microorganisms that can be introduced into Korea and to collect the opinions of domestic experts, first of all 1) we have screened the 262 cases of microbial hazards provided by a total of 21 institutions including the international organizations and oversees national authorities. As a result, we have secured a candidate group of 85 types of microorganisms related to food-borne hazards, and 7 species (2 bacteria, 2 viruses, 3 parasites) which had recent issue regarding food safety or required a detection method targeting food matrix. A total of 92 candidate microorganisms were selected. Also, 2) opinions on the high-priority research targets were collected by survey on experts in each field of infectious medicine, veterinary medicine, and food safety in Korea. In addition, 3) through a survey on food safety experts in academia or institutions of total 8 countries (4 advanced countries, 4 developing countries regarding food hygiene), the opinions and suggestions on each microorganism, national management status and traditional food groups with risks were identified. 4) By deciding the priorities of candidate groups with the prioritization decision tree developed by our research team, the first-priority pathogens of 13 species (8 bacteria, 1 virus, 4 parasites) were secured. 5) Based on above results from 1) - 4), following 12 species of first-priority pathogens or the ones with recent issues on food safety/food detection methods were designated as the research targets in this study: 5 bacteria (Coxiella burnetii , Streptococcus dysgalactiae, Vibrio mimicus, Streptococcus suis, Vibrio alginolyticus), 3 viruses (Tick- borne encephalitis virus, Severe fever with thrombocytopenia syndrome virus, Crimean-Congo hemorrhagic fever virus), and 4 parasites (Metagonimus yokogawai , Fasciola hepatica, Fasciola gigantica, Toxoplasma gondii). 6) Qualitative and quantitative detection methods targeting the 12 species were established by developing food preparation method, molecular analysis methods including nucleic acid extraction and PCR, and medium culture methods. Artificial inoculation tests were conducted for verification of the developed detection methods, and cross-validation was performed between laboratories according to the standard procedure of the Ministry of Food and Drug Safety, thereby forming the foundation as a national standard test method. 7) Food groups with high risk of contamination were selected by species and 2,789 cases of domestically distributed agricultural, livestock, and aquatic products were monitored of the prevalence of the target species. Eight species out of the 12 research subjects (C. burnetii, S. dysgalactiae, V. mimicus, S. suis, V. alginolyticus, Severe fever with thrombocytopenia syndrome virus, M. yokogawai , T. gondii) were detected in agricultural, livestock, and aquatic products. All food types with positive detection results of the target species could be decontaminated through heating and cooking before consumption. 8) Total of 1,093 samples were collected from the production environment (dairy farm, water parsley plantation, wild oyster collection site) of the target food types. T. gondii was detected in the seawater of the wild oyster collection site. 9) Also, 2 types of risk profiles were developed for each research subject: (i) a type for experts including professional academic information and (ii) and another type for consumers including risk information for safe consumption. 10) In a form that is easy to use in the fields, detection method guidelines were developed. 11) Finally, simple risk information cards were designed for 76 microorganisms which were decided as low-priority in this research and thus excluded from the main research subjects.
Overall, this study aims to prevent novel and emerging food poisoning events and decrease the occurrence of food poisonings induced by unknown causative agents through proposing the human pathogens with high plausibility of causing food poisoning as new targets of national regulation. DB of risk information customized to the domestic situation and big data of prevalence of the microorganisms of concerns in domestic foods and environments were established. In addition, we have developed detection methods that could rapidly and accurately detect the research targets in this study (bacteria, virus, and parasites) present in food and verified the methods through cross-validation tests. Also, contribution was made on the utilization and dissemination of the research results and knowledge by developing the risk profiles, test guidelines and the simple risk information cards. This is a leading research in related fields supported by actively publishing academic results based on the results obtained in the research such as the surveys on domestic and foreign experts, the prevalence of microorganisms with concern in foods and environments, and novel detection methods in foods for the research subjects.
