전통적으로 식품산업에 널리 이용되어 오던 젖산은 최근에 환경오염에 대한 관심증가로 인해 새로운 용도, 생분해성 고분자의 원료로서 각광받게 되었다. 따라서 기존의 석유화학 플라스틱에 대응할 정도의 가격으로 젖산을 생산하기 위해서는 효율적이고 경제성 있는 발효법의 개발이 요구된다. 가장 먼저 생각해 볼 수 있는 비용절감 요인은 발효배지에 있으므로 일반적으로 널리 쓰이는 고가의 yeast extract (YE)를 대체하기 위해 여러가지 다른 복합 질소원들을 탐색해 보았다. 결론적으로 YE를 완전히 대체할 수 있는 복합 질소원은 없었으며 다음에는 부분적으로 대체하기 위한 접근법을 시도했다. 즉 YE는 일반적으로 B 비타민의 함량이 다른 질소원에 비해 풍부하므로 순수한 B 비타민을 소량 첨가하여 생산성 개선 효과를 검토해 보았다. YE에 직접 비타민을 첨가하는 것은 생산성 향상 효과가 10\% 내외에 그쳤지만 soy 유래 질소원인 N-Z-Soy peptone의 경우는 2배 이상의 향상된 젖산 생산을 보였다. 따라서 N-Z-Soy peptone과 YE의 혼합배지에 25 mg/L 정도의 B 비타민을 첨가하여 같은 질소농도의 YE 배지와 비슷한 젖산 생산성을 얻어 부분적으로 YE를 대체할 수 있음을 증명했다. 식품 용도와는 달리 고분자 합성 원료로서의 젖산생산은 생산성뿐만 아니고 고분자 중합시 영향을 미치는 젖산의 광학순도와 acetic acid, formic acid, ethanol 등의 부산물 생성이 최소화되어야 한다. 따라서 젖산발효에 가장 큰 영향을 미치는 pH 제어도 이를 고려해야 하므로 회분식 발효를 통해 부산물 생성에 미치는 pH의 영향을 검토했다. 생산성은 5.0에서 6.5까지의 pH에 따라 증가했지만, 생산성과 부산물 최소화를 모두 고려한 최적 pH는 6.0이었다. 생산성을 향상시키기 위해서 반응기내 젖산균의 농도를 높게 유지해야 한다는 것은 널리 주지되어 온 사실이다. 특히 membrane filter에 의한 세포 재순환 방법은 현저한 생산성 향상효과를 보여왔는데 이와 같은 고농도 환경이 부산물 생성에 미치는 영향은 연구된 바가 드물다. 따라서 hollow fiber filter를 이용한 세포 재순환 발효에서 cell 농도의 영향을 검토했는데 결론적으로 반응기내 cell 농도가 높아도 포도당 제한이 걸리지 않으면 뚜렷한 부산물의 생성은 보이지 않았다. 부산물 생성에는 낮은 희석율에서 유래되는 포도당 제한이 더 심각한 영향을 미쳤다. 고농도 세포 배양을 위한 대표적인 다른 방법은 고정화이다. 일반적으로 calcium alginate bead를 이용한 고정화가 많이 시도되 왔지만 본 연구에서 bead보다 capsule 형태의 구조가 1.5배 정도 더 높은 농도의 젖산균을 고정화 할 수 있었다. 그러나 calcium alginate capsule은 반복 사용에서 생산되는 젖산으로 인해 구조의 불안정성을 보여 capsule 바깥으로 cell이 새어 나오는 현상을 보였다. 여러가지 강도 개선 실험을 통해 Chitosan으로 처리된 Barium alginate capsule은 반복적인 사용에서도 capsule의 형태를 유지하여 바깥으로 빠져나오는 cell의 농도가 현저하게 줄었으며 약 2.7 g/L$\cdot$h의 생산성을 유지하였다.

Recently new applications of lactic acid, such as a monomer for biodegadable plastics, have a potential to greatly expand the market for lactic acid if more economical processes could be developed. With the aim to produce lactic acid at a reduced medium cost, various complex nitrogen sources were examined in batch cultures of Lactobacillus casei subsp. rhamnosus. Among the sources tested, yeast extract (YE) enhanced lactic acid production most significantly. The addition of B vitamins of less than 25 mg/L enhanced lactic acid production to some extent in all cases except the media supplemented with nitrogen sources of casein origin. Batch fermentations were performed in order to find an optimum pH for both lactic acid production and by-products minimization. While a pH range of 6.0 - 6.5 was optimal for cell growth and lactic acid production, superior results were achieved at pH 6.0 when both maximum lactic acid productivity and minimum by-product formation were considered. Among the techniques that can increase cell concentration in the bioreactor, cell recycle using a hollow fiber filter and cell immobilization were studied. A continuous fermentor with a partial cell recycle was employed in order to investigate the physiological effects of cultivating L. casei at a high cell density in terms of by-products formation such as acetic acid, formic acid, ethanol, and \% D-lactic acid. The increase of by-products formation in the recycle fermentor was more significantly connected with substrate limitation resulted from a lower dilution rate rather than the cell concentration. The immobilization method of cells was developed using alginate capsules that possess an interphasic membrane and a liquid-core. The capsules were found to offer more space for cellular growth than gel-core beads, and allowed 1.5-fold higher cell concentration than the latter. However, the Ca-alginate structure was unstable during repeated batch fermentations for lactic acid production. Stable lactic acid production was possible in batch culture by cells immobilized in chitosan-coated Ba-alginate capsules. The cell leakage from the capsules was maintained relatively low during repeated batch fermentations.
