To compare the effects of various types of dietary fiber on fecal output, short-chain fatty acids(SCFAs) production and colon cell proliferation, groups of 10 male Sprague-Dawley rats were fed one of four fiber-supplemented diets(cellulose, pectin, polydextrose or polydextrose + cellulose) at 6% level by weight for three weeks. As a control, a fiber-free diet was fed to a separate group. Cell proliferation used as an intermediate biomarker for colon carcinogenesis was measured by in vivo incorporation of bromodeoxyuridine into DNA in the proximal and distal colon. Production of SCFAs in the colon was measured by gas chromatography to determine fermentability of the dietary fiber type. The all four fiber-supplemented diets significantly increased fecal output compared to the fiber-free diet, and, among the fiber-supplemented diets, the effect of cellulose supplementation on fecal output was the highest(p<0.05). In contrast, the cellulose-supplemented diet significantly reduced SCFAs production in both proximal and distal colon, whereas the pectin-supplemented diet significantly increased SCFAs production(p<0.05). Even if polydextrose is highly soluble fiber like pectin, the ability of SCFAs production of the polydextrose-supplemented diet was not significantly different from that of the fiber-free diet with exception of propionate production in the distal colon which was significantly higher compared to that in the fiber-free diet(p<0.05). No significant differences in cell proliferation of the proximal colon among the diet groups, whereas, in the distal colon, the labeling index(LI) was significantly higher in the pectin and polydextrose-supplemented diets than the other diet groups(p<0.05). The concentrations of SCFAS were negatively correlated to the fecal output(r=-0.59 in proximal colon and r=-0.48 in the distal colon, p<0.05), whereas they were positively correlated to the fecal moisture content(r=0.30 in the proximal colon and r=0.34 in the distal colon, p<0.05). The concentration of propionate was positively correlated to the LI in the distal colon(r=0.53, p<0.001), whereas no significant correlation was found in the proximal colon. Therefore, these data suggest that specific types of dietary fiber may exert differential effect on colon carcinogenesis, and relationship between fermentability of dietary fibers and cell proliferation may depend on site of the colon.
