The dried fruits of Gardenia jasminoides Ellis was extracted with n-hexane, ethylacetate (EtOAc), and methanol (MeOH), successively. The MeOH extract showed antimicrobial activities against Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus mutans, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa, Salmonella typhimurium, Bacillus subtilis, Lactobacillus plantarum, Leuconostoc mesenteroides, Lactobacillus brevis, Enrerococcus faecalis, Micrococcus luteus, Vibrio vulnificus, Pedicoccus cerevisiae, Candida albicans, Saccharomyces cerevisiae. The MeOH extract was purified by solvent fractionation, silica gel absorption column chromatography, Sephadex LH-20 column chromatography, ODS column chromatography. From this purification procedure, 9 active fractions were obtained. Among nine two active fractions were further purified by HPLC. The isolated antimicrobial substances were identified as 3,4-dihydroxybenzoic acid and butanedioic acid by MS, GC-MS, ¹H-NMR, ^(13)C-NMR and 2D-NMR (HMBC). The antimicrobial activity of isolated substances against microorganisms was investigated by paper disc method. As the results, butanedioic acid had stronger antimicrobial activity than 3,4-dihydroxybenzoic acid. The content of 3,4-dihydroxybenzoic acid and butanedioic acid was 32.7 ㎍, 28.5 ㎍/g dry fruit of Gardenia jasminoides Ellis, respectively. This study is the first report on 3,4-dihydroxybenzoic acid and butanedioic acid having antimicrobial activity from Gardenia jsminoides Ellis. The EtOH, MeOH extracts of Gardenia jsminoides Ellis have not only yellow pigments, but also antimicrobial activity. These substances of the fruits of Gardenia jasminoides Ellis may be used as food preservatives, coloring agent of food, dyes of objects craftwork, dyes of cosmetics and coloring agent of clothes.
