Conditional expression of ATM was observed using immunoblotting with anti-ATM antibody. An amino-terminal FLAG epitope added to the protein allowed purification of the recombinant protein from HeLa nuclear extracts. The recombinant ATM was found to have autophosphorylation activity and to actively phosophrylate p53 and chk2. These results demonstrated that recombinant ATM protein can be expressed efficiently in HeLa cells and that the purified protein contains serine/threonin kinase activity. Using these cell lines conditionally expressing ATM, I can investigate the substrates of ATM and the mechanism of ATM activation by ionizing radiation.
