우유 및 육류 중에 잔류하는 합성항균제의 일종인 설폰아마이드류를 신뢰성 있고 정확하게 분석하기 위하여 액체크로마토그래피/ 이온트랩 질량분석법과 동위원소희석법을 적용하였다. 대상 화학종으로 다섯 종류의 대표적인 설폰아마이드들을 선택하였고 우유 및 육류 시료의 전처리방법, 액체크로마토그래피-이온트랩 질량분석기 분석조건의 최적화, 내부표준물법 및 동위원소 희석질량분석법에 의한 정량방법의 확립 및 MS/ MS기법을 이용한 각 성분의 정성분석에 관한 실험들을 수행하였다. 우유와 육류 중에 존재하는 설폰아마이드 성분의 추출에 사용한 용매 중 아세토니트릴이 가장 높은 회수율을 나타내었으며 이 용매는 시료 중의 단백질을 응고시키므로 효과적으로 단백질 성분을 제거할 수 있는 장점이 있었다. 우유와 육류 중 설폰아마이드 추출의 최적 pH조건은 시료의 비극성을 유지하는 pH 4.5 부근이었다. 고체상 추출법에서 방해물질의 제거 및 대상성분의 농축을 위한 고체상으로서 에틸비닐벤젠-디비닐벤젠 공중합체 (ethylvinylbenzene-divinylbenzene copolymer)를 처음으로 적용하였으며 방해물질을 효과적으로 제거함과 동시에 가장 높은 회수율(37∼98 %)을 나타내었다. 액체크로마토그래피 분리조건 변화가 설폰아마이드류 분리에 미치는 영향을 측정하여 최적조건을 결정하였다. 그 결과 정지상은 C_(18), 이동상 중의 완충제는 0.05 M의 아세트산 암모늄염, 이동상 용매는 15 % 아세토니트릴 수용액이 분리의 최적조건이었다. 분리도에 영향을 미치는 주요 파라미터는 정지상 및 이동상의 종류이었고 이동상에 가한 완충제의 종류 및 농도는 시료의 이온화에 대한 주된 파라미터로서 작용하였다. 질량분석기의 조건변화가 설폰아마이드류의 감도에 미치는 영향을 측정하여 설폰아마이드류의 검출을 위한 액체크로마토그래피/ 대기압화학이온화 질량분석법의 최적조건을 결정하였다. 그 결과 액체크로마토그래피의 최적 분리조건에서 가열모세관 온도 150 ℃, 질소기체 유속 45 (기기의 상대값), 가열모세관 전압 10 V, 분무 전압 5.5 kv, 튜브렌즈 전압 25 V, 휘발부분 온도 450 ℃ 이었다. 대기압 이온화방법으로서 대기압화학이온화법과 전자분무이온화법을 설파메타진에 적용한 결과 기기검출한계는 두 방법이 모두 0.05 ㎍/ g 으로 유사한 검출 한계를 보였으나 검정곡선의 직선성은 대기압화학이온화법 (r^2=0.9996)이 전자분무이온화법(r^2=0.9963)보다 우수하여 정량분석에 적합하였다. 액체크로마토그래피/ 질량분석기를 사용한 설폰아마이드류 측정의 정밀도향상을 위한 최적화 실험을 수행한 결과 대기압화학이온화법과 이온트랩의 고분해능 주사모드에서 분석성분과 분석성분에 동위원소가 치환된(^(13)C_6) 내부표준물질의 면적비 측정의 상대표준편차가 0.85 %로 다른 주사방법과 이온화방법들 중 가장 측정 정밀도가 높았다. 한편 정성분석을 위하여 MS/ MS를 수행한 결과 각 분석성분에 대한 생성이온의 질량수가 예상한 값과 잘 일치하였다. 최적 조건에서 액체크로마토그래피/ 대기압화학이온화-이온트랩질량분석법과 동위원소희석질량분석법을 사용하여 복잡한 매질에 미량으로 존재하는 설폰아마이드들을 8 ng/ g의 농도인 우유시료에 대해 5회 측정 시 상대표준편차 2.9 % (95 %신뢰구간에서 확장불확도 8 % (k=2.78)), 75 ng/ g의 농도인 우육시료에 대해 5회 측정 시 상대표준편차 1.1 % (95 %신뢰구간에서 확장불확도 3 % (k=2.45)) 로 측정되었다.

A reliable and simple analytical method using LC/ MS an disotope dilution mass spectrometry (IDMS) has been developed for the determination of selected sulfonamides. Five typical sulfonamides were selected as target compounds, and milk and beef meat was selected as matrix sample. Various parameters in sample-preparation, liquid chromatography, and mass spectrometry were studied for optimization of analysis. Among various solvents tested, acetonitrile was chosen for extraction of sulfonamides in milk and meat. Acetonitrile has a merit as an extracting solvent as it causes coagulation of protein, and thus removes proteins effectively. The optimum pH for extraction was found to be 4.5. For the cleanup and concentration of sample, solid phase extraction (SPE) was used. Among various solid phases tested, ethylvinylbenzene-divinylbenzene-copolymer gave the highest recoveries ranging from 37 to 98 %. Many parameters in fluence LC separation and ionization efficiency (and thus MS sensitivity) including the type of the stationary phase (column), the type and the concentration of the mobile phase additive, and the composition of the mobile phase. It seems the LC separation is more influenced by the type of the stationary phase (column) and the composition of the mobile phase influence, while the ESI MS sensitivity is by the type and the concentration of buffer in mobile phase. It was also found the LC/ MS response of sulfonamides varies with various MS parameters including the capillary temperature, sheath gas flow, heated capillary voltage, spray voltage, tube lens offset voltage, and evaporation temperature, etc. In an atmospheric pressure ionization (API), APCI and ESI methods showed similar detection limit of 0.05 μg/ g. The calibration curve of APCI showed better linearity of r^2=0.9996 th an ESI(r^2=0.9963), so APCI was selected for ionization method for sulfonamides. The high resolution scan mode provides higher precision than the scan mode in an isotope ratio measurement. In the isotope dilution method (IDMS), the interaction and equilibration between target compound and matrix were found to be in significant for the milk but significant for the beef meat. With the IDMS method in LC/ ion trap mass spectrometry, a milk sample spiked with 8 ng/ g of sulfonamides was analyzed with the precision of 2.9 % RSD (the expanded uncertainty in 95 % confidence interval is 8 % (k=2.78)), and a beef meat sample spiked with 75 ng/ g was analyzed with the precision of 1.1 % RSD (the expanded uncertainty in 95 % confidence interval is 3 % (k=2.45)).
