ICR 생쥐중에서 생후 7일경에 과잉 행동이상을 보이며 선회운동을 하는 자연발증 돌연변이 마우스를 발견하였으며, 교배를 통하여 이러한 돌연변이가 상염색체 상에서 하나의 열성유전자 결함을 의해 유전됨을 확인하였다. 이 돌연변이를 Circling 생쥐라 명명하였으며 유전자를 Cir로 표시하였다. 청력검사를 통하여 정상청력을 가진 마우스와는 달리 circling 마우스는 명백한 청력손실을 가지고 있었으며, 또한 circling 마우스의 비정상적인 행동과 관련된 유모세포와 나선신경절의 발생학적 결함을 조직학적 연구로 관찰하였다. 따라서 circling마우스는 내이의 결함과 청력손실의 연구에 유용한 질환 모델동물이 될 것이다. 실험 2. cir 돌연변이는 심한 청력손실, 두부의 과잉 운동, 그리고 양방향의 선회운동을 보였으며, 이는 상염색체 열성유전이라고 판단하였다. 따라서 코르티기관 주위의 내이조직을 관찰한 결과, 나선신경절 세포, 외 유모세포의 결함이 관찰되었다. 본 연구에서는 cir 유전자 근방의 14개 MIT marker를 사용하여 cir 마우스와 C57BL/6J 마우스와의 역교배군에서 유전자 연관지도를 작성하였다. cir 마우스는 상염색체 열성유전을 하며, 원인 유전자는 마우스 염색체 9번에 위치하고 있었다. 또한, 연관지도 작성 결과, cir 유전자는 D9Mit116/ D9Mit15 과 D9Mit38의 사이에 위치하며, 각각의 거리는 0.7 ± 0.40 cM과 0.2cM ± 0.23 cM으로 추정되었고, 표지자와 원인 유전자 간의 순서는 다음과 같다: Certromere- D9Mit182- D9Mit51/ D9Mit79/ D9Mit310- D9Mit212/ D9Mit184- D9Mit116/ D9Mit15- cir- D9Mit38- D9Mit20- D9Mit243- D9Mit16- D9Mit55/ D9Mit125- D9Mit281. 결과적으로 D9Mit38 표지자가 Yeast Artificial Chromosome contig로부터 cir 유전자를 분리하기에 가장 유용한 지표가 될 것으로 판단하였다. 실험 3. 생쥐 9번 염색체 후방의 cir 유전자 좌위는 사람의 염색체 3p21과 높은 상동성을 가지고 있으며, 사람의 유전적인 청력 손실을 보이는 DFNB6의 원인 유전자가 이에 위치한다고 알려져 있다. 따라서 본 연구에서 cir 생쥐의 역교배를 통해 충 427마리의 생쥐로 유전자 지도를 작성할 결과, 마우스 염색체 9번에서 0.93 cM 범위로 그 위치를 좁혔다. 그리고 0.93 cM 안에 존재하는 cir 유전자를 단리하기 위하여, 18개의 YAC 클론으로 약 1960 kb의 물리적인 지도를 작성하였다. 이 물리적 지도는 cir 유전자를 포함하고 있으며, 근방의 유전자 Ltf, Lrrc2, My13, Pthr1, Kif9, 그리고 Map4등의 후보 유전자를 색출하였으나, 이러한 후보 유전자들은 cir유전자와는 관련이 없었다. 그러나 지금까지 본 연구에서 얻어진 결과는 circling 생쥐의 돌연변이 유전자를 규명하고, 청각 발달에서의 역할을 규명함은 사람 DFNB6 유전자의 청력손실의 과정을 이해하는데 유용한 모델동물이 될 것으로 사료된다. 이들 실험 결과를 고려할 때, cir 생쥐는 생쥐 9번 염색체의 60.1 cM에서 열성유전자이며, 사람의 청각손실 유전병인 DFNB6의 모델동물일수 있다. 이들 생쥐는 사람의 청각 상실뿐 아니라 청각 구조 발생을 연구하는데 유용한 모델동물로 짐작된다.

The cir mouse carries an autosomal recessive mutation that arose spontaneously in the breeding proceeds. Homozygous cir mice exhibit characteristic symptoms of Shaker- Waltzer syndrome; circling, tossing of the head, and deafness. This study might offer the basic research which aid in understanding the identification of genes controlling the mechanism of hearing. In experiment Ⅰ , the discovery and pathological characterization of new deafness mice, cir, were examined. In experiment Ⅱ, the genetic region for cir loci were refined and in experiment Ⅲ, the physical map were constructed for cir loci to identifying cir gene by positional cloning. Thus, this study was carried out to characterize and establish a spontaneous animal model for human deafness. The results obtained in this study were summarized as follows; In experiment 1, the new spontaneous mutant mouse was found in the ICR outbred strain and exhibited hyperactive behavior at about 7 days of age, followed by a tight circling behavior. Breeding studies showed that this mutation was inherited via defects of an autosomal single recessive gene. Consequently this mutation is referred to as a cir mouse mutation with the gene symbol cir. The auditory tests identified clearly the hearing loss of the cir mice when compared to wild type mice. Pathological studies confirmed developmental defects in cochlear nerve and hair cells, which were correlated to the abnormal behavior observed in the cir mice. Therefore, cir mice may be useful as a model for studying inner ear abnormalities and deafness In experiment 2, cir mice were recorded to display profound deafness and a head-tossing and bidirectional circling behavior, showing an autosomal recessive mode of inheritance. In addition, the histological examination of inner ears revealed that the region around organ of Corti, spiral ganglion neurons and outer hair cells showed definite abnormality. On the other hand, a genetic linkage map was constructed by positioning 14 microsatellite markers across the cir region in an intraspecific backcross between cir and C57BL/6J mice. The cir mouse was found to harbor an autosomal recessive mutation on the mouse chromosome 9. The cir gene was mapped to a region between D9Mit116/D9Mit15 and D9Mit38. Estimated distances between cir and D9Mit116, and between cir and D9Mit38 are 0.70 ± 0.40 and 0.23 ± 0.23 cM, respectively. The gene in order was defined as follows: centromere-D9Mit182- DQMit51/ D9Mit79/ D9Mit310- D9Mit212/ D9Mit184- D9Mit116/ D9Mitl5- cir- D9Mit38- D9Mit20- D9Mit243- D9Mit16- D9Mit55 D9Mit125- D9Mit281, These data suggest that the nearest flanking marker D9Mit38 provides a useful anchor for the isolation of the cir gene in a yeast artificial chromosome contig. In experiment 3, the distal portion of mouse Chr 9 encompassing the cir region was shared a conserved linkage homology with human chromosome 3p21. Since the human chromosome 3p21 was the region of DFNB6 deafness, cir mice was a valuable the animal model for DFNB6 deafness in human. To identify candidate transcripts in the cir region with the goal of identifying the molecular lesions, a total of 427 backcross progeny were obtained from a backcross segregating for the mouse deafness mutation, cir and linkage analysis provided a detailed genetic map within a 0.93 cM region on Chr 9. To identify further cir gene within a 0.93 cM region, 18 YAC clones were constructed for a contig across an approximately 1960 kb region of cir. They covered the entire region of cir and were therefore useful to facilitate the identification of genes in the cir region. The mouse transcripts were found to place several known genes within cir, including Ltfs Lrrc2, My13, Pfhrl, Kif9, and Map4. However, the all tested candidate genes lied outside of the cir region. However, to identify the genes responsible for circling behavior and elucidate their roles in ear development will provide a clearer understanding of the processes of hearing loss in human DFNB6. 'Taken together, the results from the experiments suggest that spontaneous cir mutant is autosomal recessive one localized to 6Q.l cM of mouse Chr 9 and more likely potential mouse model for human deafness DFNB6. Therefore, cir mutant mice could serve as a valuable experimental model for human deafness as well as development of ear structures.
