Huntington’s disease (HD) is an autosomal dominant, neurodegenerative disease, caused by an abnormal polyglutamine (polyQ) expansion in the N-terminal part of the Huntington protein (Htt). Induced pluripotent cells (iPSCs) derived from patients with Huntington’s disease (HD) have unique features, that make them a useful model system for studying disease pathogenesis and screening novel drug candidates. Therefore, I optimized the conditions to differentiate hiPSCs into cells from neuronal lineages that were used to model neuronal diseases. I also used a proteomic approach to compare Huntington’s disease-specific human iPSCs (HD hiPSC) and a normal human iPSC line (Detroit 551-8 hiPSC) to the H9 human embryonic stem cell (ESC) line. The, HD-hiPSCs had a reduced ability to differentiate and increased apoptotic cell death compared to the H9 hESCs and 551-8 hiPSCs. Therefore, HD iPSCs are an important platform for understanding changes in intracellular protein composition and cell properties before using these cells therapeutically. The results presented here provide important evidence for evaluating the properties of patient derived-iPSCs before being used for therapy and could provide a paradigm for curing several neurodegenerative diseases. Although we know that HD is caused by a faulty gene for Huntington protein, it is unclear how this mutation leads to the loss of medium spiny neurons (MSNs). Therefore, I pursued novel approaches to establishing hiPSCs lines that can be used for cell therapy and tested a novel strategy for inhibiting HDACs in vitro to treat HD. I established multiple hiPSC lines that are safer and more therapeutically relevant than previous cell lines for use as donor cells to transplant healthy MSNs into HD patients to replace cells destroyed by the disease. To do so, I compared the existing cell lines, made using retroviral or lentiviral systems, to hiPSC lines generated using safer methods, such as a non-integrating episomal vector or protein delivery system. Residual exogenous gene expression was detected in lentivirus-based hiPS cells even after they were terminally differentiated into mature neurons. In contrast, expression of the reprogramming genes was not readily detected in the retrovirus-based hiPSCs, protein-hiPSCs, and episomal vector- delivered hiPSCs after neural differentiation, indicating almost complete silencing of exogenous genes. The efficiency of neuronal differentiation appeared to occur normally in all of the hiPSCs lines. While, hiPSCs are a valuable tool for studying HD pathology and have facilitated the search for new drugs, they are not yet ready to be a treatment in their own right. In the second part of this work, I tested the effects of the HDAC6 inhibitor Tubastatin A (TBA) and the general HDAC inhibitor Trichostatin A (TSA) on HD143 hiPSCs derived mature neurons and NSCs from YAC128-TG and WT mice. The HD143 derived mature neurons and YAC128-TG mice derive NSCs have increased α-tubulin acetylation. In this model ,HDAC6 inhibition improved intracellular vesicle trafficking and therelease of BDNF. These results suggested that HDAC6 could be an important regulator of the mitochondrial axonal transport and autophagy systems. Although there is not yet a cure for HD, this study may provide insights that will help delay the onset of symptoms or prevent the progression of the disease. The microfluidics is new technology platform for cellular neuroscience that deals with the flow of liquids in channels of micrometer size. These experimental tools offer adventages for visualizing development of neural structures within a micro-scale device and to allow the guidance of neurite growth into the axonal isolation compartment. For this reason, they can be observed axonal outgrowth by neuronal culture using time-lapse microscopy, and the isolated axon serve as drug screening assay that purpose to find suitable chemical for cell therapeutic affects of neurological diseases. To study be able to generate the neronal cells derived from human ES cells, we differentiated neural precusor cells that were dissociated to the single cells and neurospheres into mature neurons. In this study, we described that the microfluidic technology in vitro can provide a useful tool to neurite outgrowth and axon guidance derived from human ES cells. Furthermore, The uniform population of axonal neurons can studies for screening pharmaceutical molecules using the neurological disease patient derived induced pluripotent stem cells.
