In vivo와 In vitro에서 혈전분해 효과를 확인하기 위하여 Fibrin과 Rat의 Platelet rich plasma에서 반응시킨 결과 Fibrin과 Rat의 PRP 음성대조군에서는 fibrin섬유가 그물모양으로 얽혀 있었고, Plasmin과 버섯으로부터 추출된 효소를 처리한 곳에서는 Fibrin섬유가 분해되어 있음을 확인 할 수 있었다. 또한 뽕나무 버섯 균사체로부터 혈전분해효소 유전자를 크로닝하여 염기서열 및 아미노산서열을 분석하여 NCBI에 등록되어 있는 뽕나무 버섯 자실체 유래 AMMEP 유전자 및 아미노산서열을 비교분석하여 본 결과 유전자의 크기는 1,056 bp로 염기서열 상동성은 약 98%를 보였으며 아미노산 서열 상동성은 99%의 높은 상동성을 보였다. 따라서 뽕나무버섯 자실체와 뽕나무버섯 균사체에서 분리·정제된 혈전분해효소가 99%이상 동일함을 확인하였다. 향후 혈전분해 관련 유전자 연구와 새로운 혈전분해물질에 대한 연구를 통하여 기능성 버섯류 개발과 인체 부작용을 최소화할 수 있는 새로운 혈전분해제 개발이 가능하리라 사료된다.

In this respect, we tried to develop highly valuable nutraceuticals or drug candidate, fibrinolytic agents from artificially cultivated mushroom sources. Blood clots are formed through the conversion of fibrinogen into fibrin by the proteolytic action of thrombin, and subsequently, insoluble fibrin clots are formed. The fibrin clots are dissolved by the hydrolytic action of plasmin, which is activated from plasminogen by tissue-type plasminogen activator (tPA). Generally, the hydoiysis of fibrin is called fibrinolysis. The fibrin clot formation and fibrinolysis are well balanced in the biological system. However, when the fibrin is not hydrolyzed due to any disorder, thrombosis such as myocardial infarction can occur. Intravenous administration of urokinase (UK), streptokinase (SK), and tPA, which are capable of degrading fibrin, has been widely used for the thrombosis therapy. Unfortunately, these enzymes have a low specificity for fibrin and are also expensive. Recently, investigations have been conducted on the isolation of fibrinolytic enzyme from natural extract, because the thrombolytic therapy meet the demand of the fibrinolytic proteases having high specificity for fibrin and cutting cost. Therefore, we have purified the fibrinolytic enzyme and cloned the gene encoding the purified enzyme from the mushroom, Armillaria mellea mycelium in this study. A. mellea, honey mushroom is well known as a symbiotic fungus with Gastodia elata and is reported that fruit body and mycelium are used to treat geriatric patients with palsy, dizziness, headache, neurasthenia, insomnia, numbness in limbs and infantile convulsion. Because cultivation need not be a limitation when active principles can be derived from mycelial cultures, we performed research on functional properties such as fibrinolytic activity with A. mellea mycelium. A strong fibrinolytic enzyme, produced by A. mellea mycelium, was purified and characterized. The purified fibrinolytic protease from A. mellea mycelium showed molecular mass of 22 kDa, both on SDS-PAGE and in fibrin zymography. It has been isolated and purified using a series of chromatographic column such as anion exchange, cation exchange and gel filtration. The protease from A. mellea mycelium showed a level of fibrinolytic activity that was about 2.0 fold higher than 1.0 unit of plasmin. The optimum pH and temperature for the enzyme activity were pH 6.0 and 30℃, respectively. The enzyme activity was relatively stable at pH 5.0~7.0 and temperature below 30℃. The activity of enzyme was activated by Ca^(2+) and Mg(2+). In addition, the enzyme activity was significantly inhibited by EDTA, indicating that the enzyme is a metalloprotease. The purified enzyme had fibrinolytic activity, hydrolyzing α, β and γ-γchains with preference in this order. It also had fibrinogenolytic activity, rapidly hydrolyzing Aα ,Bβ ,and γ chains with preference in this order. Consequently, we cloned a cDNA encoding fibrinolytic enzyme from A. mellea mycelium. The sequence of a DNA fragment encoding fibrinolytic enzyme contained an open reading frame of 1,056 base pairs encoding 352 amino acids, which is near perfect homologous with that of a fibrinolytic enzyme, AMMEP produced by A. mellea fruiting body (identities 99%). These results suggest that the purified enzyme can be a strong candidate of a therapeutic agent for the treatment of thrombic diseases.
