In the present study, the regulatory effects of thyroid hormone (which is known to induce MDR1 expression in vivo), clinical drugs, and oxidative stress on the MDR1 expression were investigated in cultured cell system. In addition, cDNA array experiment was performed to search for new regulatory signals for MDR1 expression among various anti-cancer agents and stresses. Finally, the frequency of functional genetic variant in MDR1 gene polymorphism causing the change of MDR1 expression in vivo was assessed in Korean population. Methods In order to investigate the regulatory effects of environmental stimuli on MDR1 expression, HepG2 cells were treated with thyroid hormone, rifampin, phenobarbital, doxorubicin, or hydrogen peroxide and analyzed for MDR1 expression by RT-PCR method. And to search for additional regulatory signals, cDNA array of various cancer cells treated with anti-cancer agents, UV irradiation, gamma irradiation, heat shock, protein synthesis inhibition, etc. was analyzed for the patterns of MDR1 expression. To assess contribution of genetic factor on MDR1 expression, MDR1 C3435T variant, that is associated with the change of MDR1 expression, was determined by PCR-RFLP method in 200 Korean subjects. Results When HepG2 cells were treated with thyroid hormone, the expression of MDR1 was not affected. This result was confirmed by luciferase-conjugated MDR1 promoter activity assay. Meanwhile, rifampin and phenobarbital induced MDR1 transcription, and oxidative stress, hydrogen peroxide, also induced the expression. The induction by rifampin was apparent at 16 hours and gradually decreased after the time. Other inducers showed different time-dependency. In order to search for novel regulatory stimuli for MDR1 expression, cDNA array was used. MDR1 expression was relatively high in liver cancer cells and colon cancer cells and was induced by various signals such as anti-cancer agents and stresses. Conclusions MDR1 expression was induced by environmental factors such as rifampin, phenobarbital, doxorubicin and hydrogen peroxide.
