PCV2의 재조합 capsid 단백질, A. pleuropneumoniae의 toxin인 Apx I과 Apx II, 그리고 iron-uptake에 관여하는 막단백질인 Tbp A 및 Tbp B, 그리고 점막부착과 관련된 M. hyopneumoniae의 P97단백질의 30kDa 및 50kDa 분획을 재조합 단백질로 발현시켜서 denaturing condition에서 발현 여부 확인 및 순수분리를 실시하였다. 순수분리된 PCV2 capsid 단백질을 접종하여 생산된 mouse 항체는 PCV2에 감염된 돼지의 호흡기 조직내의 viral particle과 반응함으로써 재조합 단백질과 viral protein 사이에 공통항원성이 있음을 보여 주었다. 마찬가지로 각각 준비된 A. pleuropneumoniae 및 M. hyopneumoniae의 재조합 단백질들을 mouse에 접종하여 준비한 항체들은 원래의 균체단백질과 반응함으로써 모두가 공통항원성을 띄고 있음을 알 수 있었다. Chitosan의 점막부착성을 확인하기 위해 chitosan 농도를 설정하는 실험에서 2% chitosan이 가장 부착성 및 침투성이 좋은 것으로 나타나 이 농도를 이용하여 면역실험을 실시하였다. 한편 tracheal route를 통해 항원을 직접투여 하더라도 오연성 폐렴과 같은 부작용이 일어나지 않음을 육안 및 조직학적으로 확인 할 수 있었다. Chitosan과 혼합된 재조합 단백질 항원을 tracheal route를 통하여 접종하였을 경우 항원 특이적인 IgA 및 IgG가 생성됨을 Western blot assay를 통하여 알 수 있었다. 한편 항원 특이적 IgA 및 IgG의 생성 유도 여부를 알아보기 위해 실시한 ELISA 실험 결과, chitosan 혼합제재를 tracheal route를 통해 투여한 실험군은 PBS와 혼합하여 투여한 비교군에 비해 항원특이적 IgA는 pulmonary lavage 시료에서는 비교군에 비해 통계적으로 유의성 있게 약 4배, 그리고 lung tissue grind supernatant에서는 약 3배의 흡광도 수치를 나타내었다 (P<0.01). 특히 tracheal route를 통하여 유도된 IgG의 수치는 IM route를 통해 유도된 IgG titer와 거의 유사한 수치를 나타내었다. 또한 IM route를 통한 IgG 생성 유도 실험의 결과, chitosan 혼합 제재를 투여한 경우 PBS 혼합 투여 비교군에 비해 통계적으로 유의성 있게 약 2배의 흡광도 수치를 나타내었다 (P<0.01). 위의 결과들을 종합해본 결과, chitosan 혼합제를 tracheal route를 통하여 투여하면 효과적으로 IgA 및 IgG 생성 유도가 이루어지고 이로 인한 병원균의 침입에 대한 방어효과가 기대된다.

This study describes the cloning and expression of 7 recombinant proteins, porcine circovirus type 2 (PCV2) capsid protein, Actibobacillus pleuropneumoniae Apx Ⅰ, Apx Ⅱ, Tbp A, and Tbp B, and 30kDa and 50kDa fragments of Mycoplasma hyopneumoniae P97 adhesin protein in an Escherichia coli expression system to produce a fusion protein for subsequent immunological studies. The genes encoding each protein were amplified from the genomic DNA of original pathogens by PCR and cloned into a pRSET prokaryotic expression vector, which is under the control of the T7 promoter, downstream of a 6X histidine sequence tag. The presence of each insert was confirmed by restriction analysis and by sequencing after the propagation of recombinant DNA in E. coli JM109. Two new nucleotide sequences of the PCV2 capsid gene were found (GenBank accession numbers: AY672600 and AY672601). E.coli BL21(DE3)pLysS was transformed with constructs containing each protein gene sequence. Recombinant proteins were expressed by the addition of IPTG and the expression was visualized by SDS-PAGE. Each protein was purified under denaturing condition using a Ni^(2+)-resin affinity column. Mouse polyclonal antibody raised against purified recombinant PCV2 capsid protein reacted with viral particles in a frozen section of lung on immunofluorescence assay (IFA). And the antibody from the pig with PCV2 infection reacted with the recombinant protein on a Western blot assay. These results indicate that the recombinant protein can induce an immunological response and possess the common antigenicity with the viral capsid protein. Mouse antisera raised against the 30kDa and 50kDa fragments of M. hyopneumoniae adhesin protein, P97, reacted with the mycoplasmal 97kDa protein each, indicating those two fragments are both immunogenic and can induce immune responses. Bacterial toxins (Apx Ⅰ and Apx Ⅱ) and membrane-bound proteins (Tbp A and Tbp B) of A. pleuropneumoniae were reacted with polyclonal antibodies raised against each recombinant protein, indicating that those recombinant proteins share the common antigen properties with the bacterial proteins. Chitosan formulation was found to be the most effective in adherence and infiltration into the respiratory tract when prepared with 2% concentration. In addition, no side-effect such as inhalation pneumonia was observed though the antigen was administrated directly to the respiratory tract. Antigen-specific IgA production was induced 4 times more when conjugated with chitosan than with PBS by the administration via tracheal route. Also, when administrated via tracheal route, high titer of antigen-specific IgG was induced, even as much as that of intramuscular route.
