결과 : 1) Resveratrol을 첨가한 경우에 2일 및 4일 째 모두 대조군에 비해 세포증식이 유의하게 감소되었고, 증식억제 효과는 resveratrol의 농도에 비례하였다.(p<0.05) 2) Resveratrol을 첨가하여 배양하던 중간에 resveratrol을 제거하여도 HT-29 세포수는 배양 8일 째 까지 대조군 보다 적었다 (p<0.05). 3) Resveratrol과 5-FU를 병용한 경우에 단독으로 사용한 경우에 비해 증식 억제율이 증가하였다. 4) Resveratrol 농도가 증가함에 따라 S기와 G0/Gl기의 세포수가 증가하고, G2/M기의 세포수가 감소하였다. 5) 세포사멸은 resveratrol의 농도에 비례하여 증가하였다.

In the present study the effects of resveratrol on the growth of human colonic carcinoma cell line, HT-29 cells were examined and the combined effects with 5-fluorouracil (5-FU) were also studied. Cell cycle analysis was done to examine the mechanisms for the inhibitory effects of resveratrol.
Materials/Methods : The growth of HT-29 cells were examined by counting cell number on two and four days treatment with 1, 2.5, 5, 7.5, 10, 25 and 50 μM resveratrol, and 0.1 and 0.3 ㎍/㎖ 5-FU, after plating HT-29 cells into 35㎟ plastic dishes at a density of 10 x 10^(4) cells/dish. The reversibility of the effects of resveratrol was examined on one day to seven days treatment with 7.5 μM resveratrol after seeding 2 x 10^(4) cells/dish. To examine the mechanisms for the inhibitory effects of resveratrol, cell cycle analysis was done on the cells after four days treatment with 10, 50, and 100 μM of resveratrol. Induction of apoptosis was evaluated by analyzing the presence of chromatin condensation by fluorescent microscopy using DNA-binding fluorescent dye DAPI. Treatment with 10 to 100 uM of resveratrol induced chromatin condensation and nuclear fragmentation suggesting apoptosis.
Results : 1) Resveratrol significantly inhibited the growth of HT-29 cells in a dose-dependent fashion (p<0.05). 2) After removal of the resveratrol from the media, the suppressive effect of resveratrol on cell proliferation did not returned to normal, and cell count was significantly lower than that of control (p<0.05). 3) Resveratrol enhanced the inhibitory effect of 5-fluorouracil (5-FU) on HT-29 cells. 4) The cells accumulated at the S and G2/M transition of the cell cycle. Resveratrol induced apoptosis of cells and it was correlated with the concentration of resveratrol.
Conclusion : Resveratrol significantly inhibited the growth of HT-29 cells in a dose-dependent fashion and its inhibitory effect was irreversible. The inhibitory effect of resveratrol on the proliferation of HT-29 cells may be derived from inhibition of progression from S to G2/M of the cell cycle, and from the apoptosis that is proportional to the concentration of resveratrol.
