We estimated the oxidative stress effect on nDNA and mtDNA via comet assay and DNA fragmentation assay. The expression levels of Mn-SOD, Cu/Zn-SOD and CytC were estimated by western blot. Tissue morphology was examined by H&E staining and mitochondrial morphology was analyzed by transmission electron microscope.
Our results showed that expression levels of DNA (mtDNA, nDNA) damage and Cu/Zn-SOD were higher in LVH rabbit hearts in comparison with control rabbit hearts. We also found lower mtDNA damage, higher CytC and Mn-SOD expression levels in mitochondria of regional tissues were corresponded to the lower O_(2)^(-) production in IPC compared with IR.
In conclusion, our results will contribute to the investigation of molecular mechanisms of hypertrophy, IR - induced oxidative stress to cardiovascular disease and the protective role of IPC against ischemic injuries.
