For this reason, topical treatment with ceramides has been comprehensively used for skin disorders such as psoriasis, itching, atopic dermatitis, irritant contact dermatitis, allergic contact dermatitis, and so on. However, we have a lot of difficulties in applying ceramides in topical treatments or cosmetics due to their poor solubility and instability. From this view point, we prepared nanoliposome composed of ceramide in order to improve solubility and stability of ceramide and found that the poor-soluble ingredient in nanoliposome showed better stability in this study. The formulas not containing ceramide and containing nanoliposome composed of ceramide were not observed any recystallization after cycling and freeze/thaw conditions. But the formula containing dispersed ceramide showed recrysallization at the interface between internal and external phase after cycling and freeze/thaw conditions. To investigate the potency of nanoliposome composed of ceramide for reinforcement of skin barrier function, we examined the recovery rate of transepidermal water loss (TEWL) after tape stripping, moisturizing effect and the inhibitory effect of flush induced by methyl nicotinate. Subsequently, clinical studies such as human patch test and stinging potential test were conducted to evaluate the anti-irritation effects of nanoliposome composed of ceramide using lactic acid as a chemical irritant. According to the results, nanoliposome composed of ceramide showed certainly better effects on reinforcement of skin barrier function and anti-irritation than dispersed ceramide. And then, we performed in vitro skin penetration test using horizontal Franz diffusion cells equipped with skin membrane prepared from hairless mouse to evaluate the influence of nanoliposome composed of ceramide on the skin penetration of lactic acid in formulations. From the results, we found that the anti-irritation effects of nanoliposome composed of ceramide were due to the reduced penetration rate of irritants. There were no significant differences in TEWL reduction and hydration effects among the formulas containing nanoliposome composed of ceramide with a variety of different mean particle sizes, but we could confirm that the smaller mean particle size of nanoliposome composed of ceramide was, the higher anti-irritaion effect was. Natural ceramides showed slightly better anti-irritation effects than pseudoceramides, and nanoliposome composed of ceramide and hydrophilic actives together results in synergistic anti-irritation effects by means of various anti-irritation mechanisms with the ability in modulation of either activation or damage of keratinocytes, and release of inflammation mediators. Another ultimate objective of this study is to develop cosmetic products for the treatment of atopic dermatitis. The overproduction of the inflammation- mediating cytokines such as interleukin-8 (IL-8) and tumor necrosis factor-α (TNF-α) is also observed in atopic dermatitis due to the ceramide deficiency. And it has been known that elastase is released from the skin surface after skin irritation caused by UV-exposure or environmental stress as well as dry skin conditions on atopic dermatitis. We screened some cosmetic ingredients which have excellent inhibitory effects on the production of IL-8 and TNF-α, and on the activity of elastase, and prepared nanoliposome composed of ceramide and the selected cosmetic ingredients together to improve stability, penetration and efficacy at usage levels. Finally, we manufactured a cosmetic product for the treatment of atopic dermatitis and evaluated the efficacy of a cosmetic product with atopic dermatitis patients. The cosmetic product containing nanoliposome composed of ceramide and selected actives was very safe and exhibited very excellent hydration effect and TEWL reduction effect compared to those of placebo and two comercialized atopy care creams. The topical application of the product also showed remarkable effects on the improvement of atopy skin. Conclusively, we could characterize nanoliposome composed of ceramide in detail and apply this new system to develop the final cosmetic products successfully.
