Levan, a polysaccharide produced by plants or microorganisms, is a sugar polymer consisting of fructose with β-2,6 linkages. In order to investigate the possibility of Levan produced by Zymomonas mobilis as cosmeceutical ingredient, we measured its moisturizing effect, cell cytotoxicity, cell proliferation effect, and anti-inflammation effect. Levan showed almost same moisturizing effect and cell proliferation effect as hyaluronic acid in human fibroblast and keratinocyte cell lines. Moreover, in cell proliferation test on bio-artificial skin constructed by 3-dimensional (3-D) culture after inducing primary skin inflammation with 0.5% sodium lauryl sulfate (SLS) cell viabilities in the presence of Levan (0.01 mg/㎖, 0.05 mg/㎖) was higher than that in the absence of Levan. In anti-inflammation test on 3-D artificial skin evaluated by measuring secreted quantity of interleukin-1α (IL-1α) which is a pre-inflammatory mediator induced by SLS, the quantity of IL-1α on the 3-D artificial skin treated with 0.01 mg/㎖, 0.05 mg/㎖ of Levan was less than that of one treated with SLS only. In this study, Levan showed anti-inflammation effect against inflammatory reaction by a skin irritant as well as cell proliferation effect on bio-artificial skin indicating its potential applicability as cosmeceutical ingredient.
