Changes in the physiochemical and microbiological characteristics were investigated during alcohol fermentation of Campbell's Early grape and Campbell' Early must by indigenous yeasts. Sugar contents were reduced from 25.0 to 7 °Brix after fermentation for 18 days. Alcohol contents increased as the alcohol fermentation proceeded and reached the maximal level in which the alcohol content was 13.0%. The TTC (2,3,5- triphenyl 2H-tetrazolium chloride) colouring test of the microbiological changes during the fermentation showed that white color colonies appeared as the major group at the early stage of fermentation but theirviable counts decreased as the alcohol formation proceeded. At the late stage of fermentation, red color colonies were the major, which showed high tolerance to alcohol. Total 300 yeast strains were randomly isolated from the fermentation mixtures at the different stages of fermentation. When they were investigated by using PCR with intron splicing site primers, it was found that there were several different yeast strains at the early stage of fermentation. PCR analysis of the yeasts resulted in that the yeasts isolated during the early stage of fermentation showed different patterns. However, the yeasts isolated during the late stage of fermentation showed the same patterns suggesting that the same kind of yeasts play a major role in the wine fermentation. Several yeast strains showing a high tolerance against osomotic pressure in the presence of 50% glucose and high level of alcohol fermentation were selected and characterized. Strains S13, D8 and M12 were identified as Saccharomyces. boulardii and strain S6 was identified as Hanseniaspora uvarum, based on their morphological and physiological characteristics as well as the DNA sequences of there rDNA internal transcribed spacers (ITS). The strains showed the same patterns of growth in a range of pH between 3.0 to 9.0. But strain S6 could grow at pH 2.0 suggesting that it is a acid-tolerant yeast. Their fermentation of carbohydrates except for S6 resulted in the same patterns. When they were used as a starter for the wine fermentation, no big differences were found in the contents of alcohol, phenolic compounds and residual sugar as well as in the viable cell counts.
