Microglia BV2 cell 내에서 염증 유발과 관련된 iNOS와 COX-2의 단백질 발현은 Western blot 방법으로, m-RNA 발현은 RT-PCR로 확인하였다. 연구 결과 : MTT assay 결과 蒼朮은 50ug/ml까지 유의한 세포독성을 보이지 않았으며, 蒼朮 물 추출물과 75%, 100% 에탄올 추출물이 효과적으로 i-NOS와 COX-2의 단백질 발현과 m-RNA 레벨을 감소시켜 결과적으로 NO의 생성을 유의성 있게 감소, 억제함을 알 수 있었다. 그러나 이 과정이 MAPK 의 ERK 나 JUK, P38 pathway를 거치지는 않는다는 점이 확인되었다. 결론 : 蒼朮의 물 추출물과 75%, 100% 에탄올 추출물은 mouse의 microglia BV2 cell에서 LPS자극에 의해 증가된 iNOS와 COX-2의 발현을 억제함으로써 염증에 관여하는 최종산물인 NO의 생성을 억제하였다. 이러한 결과는 蒼朮 추출물이 뇌의 감염성 및 염증성 질환의 치료에 유의한 영향을 미칠 수 있음을 시사하는 것이다.

Objective : In this study, the effect of Atractylodes japonica against LPS induced inflammation in mouse microglia BV2 cells was investigated. Method : Microglia BV2 Cells viability was determined using the MTT assay. We studied on the anti-inflammatory effect of lipopolysaccharide-induced inflammation using reverse transcription polymerase chain reaction (RT-PCR), western blot, and nitric oxide detection on mouse microglia BV2 cells. Result : The MTT assay revealed that Atractylodes japonica extract has no significant cytotoxicity in the microglia BV2 cell. Various solvent extract from Atractylodes japonica inhibited nitrite production, iNOS protein and mRNA expression levels. And also Atractylodes japonica significantly reduced lipopolysaccharide-induced COX-2 activation in RT-PCR and western blot in lipopolysaccharide-induced microglia BV2 cells Conclusion : In this study, Atractylodes japonica was shown to suppress NO production by inhibiting iNOS expression and COX-2 activity. With this effects of anti-inflammation, we suggests that, Atractylodes japonica may be a useful candidate for the development of a drug on the related inflammatory diseases in brain.
