Silica gel TLC prep의 방법을 이용하여 2개의 생성물을 분리한 후 1H-NMR, MALDI-MS를 통해 반응생성물들의 구조분석을 진행하였다. 분석한 결과 compound K와 oleic acid의 에스터화 반응은 진행이 되었으나 다른 화학반응이 일부 함께 일어나 예상했던 구조가 아닌 화합물이 생성된 것으로 확인이 되었다. In the present study, two kinds of organic solvents such as 1-butanol and ethyl acetate were used for esterification reaction. Water was found to be not suitable for the reaction since the lipase would get decomposed in aqueous solution. At first, 1-butanol and oleic acid were esterified with lipase GF. This reaction was carried out at 37℃ by shaking in an incubator. The reaction mixtures were analyzed by both TLC and HPLC. The product was separated by silica gel chromatography (open column), and the structure and purity of the product were confirmed with 1H-NMR, 13C-NMR and ESI-MS. In the second part of the experiment, compound K and oleic acid were esterified with lipase GF in ethyl acetate medium. This reaction was carried out at 37℃ with continuous stirring. The reaction mixtures were also analyzed by both TLC and HPLC. The decrease in the amount of the ginsenoside was clearly shown. Three new reaction products were separated by silica gel prep TLC, and analyzed by 1H-NMR and MALDI-MS for structure determination. In similar experimental conditions, esterification reaction between compound K and palmitic acid was carried out with lipase GF in ethyl acetate medium. New reaction products were also detected by TLC analysis. Esterification reaction between compound K and oleic acid was also investigated by a chemical method in methylene chloride.
