HPLC를 사용하여 측정한 왁스 물질의 주 구성 성분은 25.5-34.8%가 폴리코사놀, 10.5%-18.8%는 탄화수소, 49.8%-53.0%가 왁스 에스터, 스터릴 에스터와알데히드는 1.7%-2.1% 이었으며, 1.0%-2.9%가 트리아실글리세롤 이었다. 폴리코사놀의 구성 성분을 GC로 측정한 결과, 옥타코사놀이67-68%, 16-17%가 헥사코사놀, 6-9%가 트리아콘타놀로 구성되어 있었다. 비교를 위하여 참깨씨의 왁스 물질도 함께 분석하였다. 참깨씨에 있는 폴리코사놀의 양은 들깨씨와 비교했을 때 매우 적은 5% 이하이었다. 들깨씨 기름의 지방산 조성을 GC로 분석했다. 들깨씨 기름에는 리놀렌산이 49-60%로 가장 많았다. 들깨씨 기름과 다른 식물성 기름(콩기름, 옥수수 기름, 올리브 기름, 유채유, 포도씨 기름)에 함유되어 있는 토콜의 조성을 분석하기 위하여, 기름을 비누화한 후 토콜 조성을 HPLC를 사용하여 분석 하였다. 모든 토코페롤(α, β, γ, δ)이 들깨씨 기름에서 발견되었지만, 토코트리에놀은 발견되지 않았다. 들깨씨 기름에 가장 많이 함유되어 있는 토콜은 γ-토코페롤(31-70 mg/100 g)이었고, 그 다음은 β-토코페롤(4 mg/100 g)이었다. 다음의 몇몇 식물성 기름 시료에 함유되어 있는 토코페롤과 토코트리에놀의 함량의 차이도 뚜렷하였다. 즉, 올리브 기름에 있는 α-토코페롤 함량은 14-16 mg/100 g, 유채유에는 7-18 mg/100 g, 포도씨 기름에는 14-18 mg/100 g이었다. γ-토코페롤은 콩기름에는 47-64 mg/100 g, 옥수수 기름에는 34-49 mg/100 g이 함유되어 있었다. 계산에 의하여 산출한 비타민 E의 활성은 콩기름에서 매우 높게 나타났다. 들깨씨는 우리가 매일 섭취하는 다른 식품들과 비교했을 때, 폴리코사놀이 많이 함유되어 있더라도 이 함량은 상업적으로 이용하기에는 충분하지 못하다. 그러나 본 연구 결과에 따르면 일반 음식물과 더불어 들깨를 충분히 섭취하면 폴리코사놀, 필수 지방산, 토콜을 상당량 공급할 수 있을 것으로 판단된다.

The purpose of this study was to determine the contents and compositions of policosanol and tocols in perilla seeds (Perilla frutescens) using TLC, HPLC and GC. Waxy materials were extracted from perilla seeds using hot hexane. For comparison, waxy materials of sesame seeds were also analyzed. The fatty acid composition of the perilla seed oils were also analyzed by GC. Tocopherols and tocotrienols were extracted from perilla seed oil and different vegetable oils (soybean, corn, olive, canola and grape seed oil) by saponification. Composition of tocols was analyzed by isocratic HPLC. Major components of the waxy materials were policosanols (25.5-34.8%), hydrocarbons (10.5-18.8%), wax esters, steryl-esters and aldehydes (49.8-53.0%), acids (1.7-2.1%), and triacylglycerols (1.0-2.9%), determined by HPLC. Similarly, the components determined by HPLC from policosanols were 67-68% octacosanol, 16-17% hexacosanol, 6-9% triacontanol and others. Result showed that the content of policosanol in sesame seeds was very small (<5%) as compared to the perilla seeds. All tocopherols (α, β, γ, and δ) were detected in the perilla seed oil but tocotrienols were not detected. γ-Tocopherol was most abundant (32-40 mg/100 g oil), followed by β-tocopherol (4 mg/100 g oil) in perilla seed oil. A distinct variation in the contents of tocopherols and tocotrienols among the samples of vegetable oils were observed. It was found that α-tocopherol was 14-16 mg/100 g olive oil, 7-18 mg/100 g canola oil, and 14-18 mg/100 g grape seed. Similarly, γ-tocopherol was 47-64 mg/100 g soybean oil, and 34-49 mg/100 g corn oil. Vitamin E activity was highest in the soybean oil (23-33 mg/100 g oil). Linolenic acid (49-60%) was the most abundant in the perilla seed oil, followed by oleic acid (15-19%). Although perilla seeds contain considerable amounts of policosanols compared to the other major staple foods, the level of policosanols and tocopherols may not be enough for perilla seeds to be commercially used. However, this study indicates that consumption of perilla seeds may be beneficial in supplying policosanols, essential fatty acids and tocopherols in a diet.
