본 연구에서는 시중에 유통되고 있는 인삼농축액(whiteginseng extract, WG), 발효인삼농축액(fermented ginseng extract, FG), 홍삼농축액(red ginseng extract, RG)에 대해 사포닌, 고형분, 점성도, 회분, 조지방, 조단백, 탄수화물, 나트륨, 열량, 산성다당체, 향기성분 등을 분석 하였고, 인삼농축액, 발효인삼농축액, 홍삼농축액이 항산화, 면역활성, 혈관이완작용에 미치는 영향을 규명함으로써 인삼농축액, 발효인삼농축액, 홍삼농축액의 효능 차이를 비교 평가하고자 하였다. 인삼농축액, 발효인삼농축액, 홍삼농축액의 고형분 조사 결과 홍삼농축액, 인삼농축액, 발효인삼농축액 순으로 발효인삼농축액이 가장 높았으나, 점성도 조사 결과 홍삼농축액이 가장 높게 나타났다. 탄수화물과 나트륨의 함량을 조사한 결과 인삼농축액이 가장 높았고, 회분과 조단백질의 함량은 발효인삼농축액이 가장 높았다. 조지방과 산성다당체는 홍삼농축액이 가장 높은 것으로 나타났다. 그러나, 열량에 있어서는 큰 차이가 없는 것으로 조사되었다. 인삼농축액, 발효인삼농축액, 홍삼농축액으로부터 향기성분을 채취한 후 조사한 결과 furfuryl alcohol, maltol, 2-acetyl pyrrole 성분이 인삼농축액, 홍삼농축액에 비해 발효인삼농축액이 현저하게 감소하였으며, 반면에 2,6-dimethyl pyrazine, trimethyl pyrazine, borneol 성분의 증가가 관찰되었다. 또한, 인삼 속에는 존재하지 않고 홍삼에서만 존재하는 물질로 알려진 maltol 성분이 인삼농축액, 발효인삼농축액, 홍삼농축액 측정된 것은 인삼농축액과 홍삼농축액 제조시 열이 가해져 maltol 성분이 생성되는 것으로 생각된다. 인삼농축액, 발효인삼농축액, 홍삼농축액에 대해 TLC 분석을 실시한 결과 인삼농축이 발효인삼농축액, 홍삼농축액에 비해 major 사포닌(ginsenoside Rb1, Rb2, Rc, Rd, Rg1)이 높았고, minor 사포닌인 ginsenoside Rg3는 홍삼농축액, compound K는 발효인삼농축액이 가장 높게 나타났다. 사포닌의 정확한 함량을 판별하기 위해 ginsenoside표준물질을 사용하여 HPLC 조건을 확립한 후 분석한 결과 인삼농축액에서 발효인삼농축액과 홍삼농축액에 비해 많은 양의 magor 사포닌 함량이 존재하였다. 특히 인삼농축액 및 발효인삼농축액에서 홍삼 특유의 사포닌으로 알려진 ginsenoside Rg3도 HPLC 분석한 결과 존재하였는데, 이는 추출과정중 가열에 의해 생성된 것으로 홍삼과 백삼모두 가공 조건에 따라 2차산물이 생성 될 수 있음 을 의미한다. Compound K는 인삼농축액과 홍삼농축액에서는 그 존재를 확인 할 수 없었으나, 발효인삼농축액에서는 다량 존재 하였다. 인삼농축액, 발효인삼농축액, 홍삼농축액의 항산화, 면역활성, 혈관이완작용데 대한 활성을 분석한 결과 HFD에 대한 항산화 효소인 SOD, CAT의 활성은 인삼 추출물 투여군에서 전반적으로 낮았으며, GPx의 활성은 WG1000, FG1000 RG1000군에서 가장 높은 결과를 보였다. H2O2 함량은 모든 군에서 거의 변하지 않는 유사한 결과를 보였으며. GSH 함량은 모든 군에서 증가하였다. 자유라디칼에 의해 생성된 지질과산화의 최종 산물인 MDA의 함량은 모든 투여군에서 낮게 나타났으며, 인삼 농축액을 다량으로 투여할수록 GPx의 활성은 증가하고 MDA의 함량은 줄어드는 것을 알 수 있었다. HFD에 대한 인삼 농축액은 SOD, CAT의 활성 증대보다는 자유라디칼 억제제로서 항산화 효과를 보였다. 전반적으로 백삼 농축액과 발효인삼농축액 그리고 홍삼농축액이 비슷한 활성을 나타내었지만, 발효인삼농축액이 좀 더 유용하게 작용하였다. 이와 같은 결과로 볼 때 발효농축액은 면역활성, 동맥확장, 항산화 작용에 의한 노화방지 등의 효능을 지닌 기능성 인삼제품으로의 개발이 가능할 것으로 예측된다.

In the recent decades, many studies have focused on pharmaceutical activities of minor ginsenosides, such as ginsenosides Rg3, Rh2 and compound K. Their activities were found to be more effective to those of major ginsenosides. It has been claimed that ginseng has many beneficial bioactive effects on human health, such as antistress, antiaging and enhancing immune functions. The objective of this study is to compare the composition and efficacy of white, fermented and red ginseng extracts. Ginsenoside contents of white ginseng, fermented and red ginseng extracts were analyzed by TLC. Contents of major saponin (ginsenoside Rb1, Rb2, Rc, Rd, Rg1) of white ginseng extract were higher than fermented or red ginseng extracts. Contents of the ginsenoside Rg3 of red ginseng extract and the compound K of fermented ginseng extract were higher than the others, respectively. Ginsenoside contents of three ginseng extracts were quantitatively analyzed by HPLC. Average contents of total saponin of white ginseng extract were higher than fermented and red ginseng extract. Red ginseng extract contains compound K in large quantities, but it does not exist in white or fermented ginseng extract. Content of extractable solids of fermented ginseng extract was the higher than other ginseng extracts, but viscosity was the lower. Contents of carbohydrate and sodium of white ginseng extract were higher than other ginseng extracts, but contents of ash and crude protein of fermented ginseng extract were higher than other ginseng extracts. Contents of crude fat and acidic polysaccharide of red ginseng extract were higher than other ginseng extracts. The calories of three ginseng extracts were not significant. Results of flavor components analysis determined low amount of furfuryl alcohol, maltol and 2-acetyl pyrrole. In opposition, 2,6-dimethyl pyrazine, trimethyl pyrazine, borneol was found significantly high in fermented ginseng extract. When white ginseng extract (WG), fermented ginseng extract (FG) and red ginseng extract (RG) (500, 1000 mg/kg/day for 4 weeks, orally) were used as oral administration, the activity of SOD and CAT were generally decreased. But Gpx activity was increased. The antioxidant effect of three ginseng extracts acted as inhibitor of free radical than increasing of SOD or CAT activity. The hydrogen peroxide (H2O2) was not induced in all groups. The GSH levels were increased dose dependently. The level of MDA (malondialdehyde) were generally decreased in the white, fermented and red ginseng extract groups. Mainly, the white, fermented, red ginseng extracts acted to increase the antioxidative enzyme activity. Especially, fermented ginseng extract showed the higher antioxidant effect than other extracts. On the other hand, white or red ginseng extracts showed inhibiting effect of lipid peroxidation. In this study, many effects of fermented ginseng extracts were determined. For example, the immune enhancing, artery expanding, antioxidant and anti-aging effects were demonstrated. These kinds of effects of the fermented ginseng extracts are expected to develope the functional ginseng products.
