본 연구는 Estrogen, isoflavone 및 대두 추출물의 농도별 처리를 한 MC3T3-E1 조골세포와 조건배양액에서 파골세포 분화관련 국소인자(IL-1, IL-6, TNF-α, M-CSF, RANKL 및 OPG)의 발현 변화를 단백질과 RNA수준에서 살펴보았으며, 그 결과를 요약하면 다음과 같다. 1)세포 내 MC3T3-E1 osteoblastic cell에서 파골세포 분화관련 국소인자인 IL-1, IL-6, M-CSF, OPG의 mRNA 수준으로 살펴본 결과 표준단일물질 및 대두 추출물 처리 시 -Control에 비해서는 현저한 증가를 보였으나, 대체로 +Control과 비슷하여 발현변화가 미비하였다. 2)MC3T3-E1 조골세포의 파골세포 분화관련 국소인자를 단백질 수준에서 살펴본 결과 IL-1는 표준 물질에서 +contol과 비교하여 유사한 수준의 발현을 보였으며, IL-6는 전반적으로 증가하는 경향을 보였고 두 인자 모두 특히, 대두추출물 처리군에서 높게 나타났다. RANKL 역시 전반적으로 증가하였고 10^(-9)M genistein, didzein과 대두 추출물 처리군에서 높게 발현되었다. 마지막으로 OPG는 10^(-9)M genistein과 10^(-6)M estrogen, S0.001 mg/ml과 S0.1 mg/ml에서 높은 발현을 보였으며 특히 저농도의 대두 추출물 처리군에서 현저히 높은 발현이 나타났다. 3)대두 및 estrogen, isoflavone 표준물질을 처리한 MC3T3-E1 조골세포 조건배양액에서는 IL-1 과 OPG의 발현이 증가되었으며, 대두추출물 처리에 의한 효과가 현저하였다. 결론적으로 대두추출물 처리를 한 MC3T3-E1 조골세포는 파골세포 분화에 관여하는 국소 인자 중 OPG와 IL-1의 발현을 선택적으로 높여주는 것으로 사료된다.

Soybean is of particular interest as a food supplement of isoflavones for inhibiting bone resorption in postmenopausal woman. These beneficial effects of isoflavones are caused by functioning as partial agonists or antagonists of estrogen, of which anti-resorptive effect is mediated indirectly through paracrine factors produced by osteoblasts. To investigate the effect of soybean extracts on the synthesis and secretion of bone resorbing paracrine factors in MC3T3-E1 osteoblasts, 70% methanol extracts of soybean were treated in osteoblastic MC3T3-E1 cells for 6 days, and cell lysates and conditioned medium were collected. In osteoblastic MC3T3-E1 cells, interleukin(IL)-1, IL-6, macrophage -colony stimulating factor(M-CSF), osteoprotegerin-ligand(OPGL) and OPG, an inhibitor of bone resorption with binding to OPGL, were expressed. Soybean treatment at 0.001 mg/ml - 0.1 mg/ml did not alter mRNA and protein expressions of IL-1, IL-6, M-CSF, OPGL and OPG in MC3T3-E1 cells. However, in the conditioned medium, the protein expression of IL-1 and OPG with soybean treatment was specifically higher in a dose dependent manner than with 10^(-9) M - 10^(-6) M of estrogen, and isoflavone standards such as genistein or daidzein. Taken together, we demonstrate that soybean enhances the secretion of OPG and IL-1 protein.
