The effects of dairy products on the prevention of osteoporosis were studied in vivo and in vivo. In vivo studies, osteoblastic cell line (MC3T3-E1) and osteoclastic cell line (RAW 263.7) were used to determine anti-osteoporotic effect of whey protein concentrate (WPC), lactoferrin fortified whey protein concentrate (WPCL), hydrolysate of whey protein concentrate (WPCH), and fermented product of L. Casei 393 (FMP). All dairy product increased the proliferation activity of undifferentiated MC3T3-E1 cell when supplemented in a 2 % FBS enriched media. The activity of alkaline phosphatase (ALP), the calcium deposition enzyme, was increased by supplementation of dairy product (P<0.05). However, ALP activity was less than that of positive control group that was grown in 10 % FBS enriched media. Differentiation of MC3T3-E1 cell determined by the mRNA expression of CBFA1 was not enhanced by supplementation of all dairy products. The expression of mature mineralization marker, osteocalcin, was neither changed by supplementation of dairy product. The proliferation of osteoclastic cell line, RAW 263.7, was not affected by supplementation of dairy product, and their development to multinuclear cell was neither affected. However, the tartrate resistance acid phosphatase activity, bone resorption enzyme, was decreased by FMP supplementation. In vivo studies, the rats induced osteoporosis by ovariectomy showed increased bone mineral density (BMD) and bone mineral content (BMC) when they consumed diets contained dairy product. This increased bone mineral density and bone mineral content resulted in increased bone breaking force. This increased bone strength is medicated by reduced bone resorption. Bone resorption activity, TRAP activity, was reduced in WPC, FMP, and WPCH supplemented groups, and their mRNA expression was also reduced (P<0.05). However, the anti-osteoclastic factor produced from osteoblast (OPG) was reduced in WPC and WPCL groups, dairy product consumption increased bone strength by mainly reducing osteoclastic activity. The increased mineralized osteoblastic cells also suggest that the anti-osteoporotic effect of dairy product could be mediated by increasing osteoblast cell on bone. However, their molecular effects on osteoclast and osteoblast are still uncertain.
