In vitro 상에서 흑마늘의 기능성 규명을 위하여 용매 분획물의 항산화활성을 비교 분석하였으며, in vivo 상에서 흑마늘 분말 및 물추출물이 고지혈증 유발 흰쥐의 지질개선에 미치는 영향을 분석하는데 그 결과를 요약하면 다음과 같다. 1. 60, 70, 80 및 90℃로 조절한 incubator에서 1, 3 및 6일간 숙성한 마늘의 L, a 및 b값 모두 숙성온도가 높을수록 숙성기간이 경과할수록 유의적으로 낮아졌는데, 60℃에서 6일간 숙성한 시료의 b값은 22.16±0.76이었으나 90℃에서 숙성한 시료는 1.35±0.21로 급격히 감소하였다. 전단가의 경우 숙성 초기에는 90℃에서 숙성시킨 시료가 85.70±1.44로 가장 낮았으나 숙성기간이 경과할수록 경도가 증가하여 숙성 6일에는 411.30±13.90으로 증가하였다. 2. 마늘의 pH는 숙성온도가 높을수록 숙성 기간이 경과할수록 산성화되었는데, 60℃와 90℃에서 각각 6일간 숙성시킨 시료의 pH는 각각 pH 6.12와 pH 3.90으로 큰 차이를 보였다. 총 페놀과 플라보노이드의 함량은 숙성온도가 높을수록 숙성 기간이 경과할수록 증가하였으며, total pyruvate의 함량은 60℃와 70℃에서 숙성시킨 결과 숙성기간이 경과함에 따라 감소하는 경향을 보였으나 80℃와 90℃ 숙성시킨 시료에서는 숙성 1일에 비해 숙성 6일에 오히려 그 값이 유의적으로 증가되었다. 3. 숙성 온도를 달리한 마늘 열수 및 에탄올 추출물의 DPPH에 대한 항산화활성은 마늘의 숙성 온도가 높을수록, 숙성기간이 경과할수록, 농도가 진할수록 증가하여 90℃에서, 6일간 숙성시킨 시료의 항산화활성은 87.48±0.20%로 타 시료의 43.61~55.76%보다 월등히 높았고 환원력도 이와 비슷한 경향이었다. 4. 흑마늘의 이화학적 특성을 생마늘 및 찐마늘과 비교 분석한 결과 적색도는 흑마늘에서 2.86으로 월등히 높은 값이었고, 명도와 황색도는 흑마늘에서 각각 22.52±0.17와 3.19±0.68로 가장 낮았다. 조지방, 조단백질 및 총당은 흑마늘, 찐마늘 및 생마늘의 순으로 높았고, 수분의 함량은 흑마늘에서 58.20±0.39%로 조지방이나 조단백질과는 상반된 결과를 나타내었다. pH는 흑마늘은 pH 4.36±0.06로 가장 낮았고, total pyruvate 함량은 흑마늘에서 277.85±2.57 μmol/g으로 가장 높았다. Total thiosulfate의 함량은 찐마늘에서 가장 낮았으며, 다음으로 생마늘 및 흑마늘 순이었다. 5. 유리당 중 arabinose와 galactose는 흑마늘에서만 검출되었으며 생마늘에서는 glucose, 찐마늘에서는 sucrose, 흑마늘에서는 fructose의 함량이 가장 높았다. 무기물은 칼륨의 함량이 다른 무기물에 비하여 그 함량이 월등히 높아 전체 무기물 함량의 약 76%를 차지하였다. 구성아미노산은 ammonia를 제외한 모든 아미노산이 생마늘에 비해 흑마늘에서 높게 정량되었다. 유리아미노산은 생마늘과 찐마늘에서는 총 15종이 검출되었으나 흑마늘에서는 taurine, L-aspartic acid, glycine, L-α-aminoiso-n-butyric acid 및 γ-Amino-n-butyric acid를 포함한 총 5종의 아미노산과 urea 및 O-phosphethanolamine이 더 검출되었다. 6. 생마늘, 찐마늘 및 흑마늘의 열수와 에탄올 추출물의 전자공여능은 활성이 가장 높았던 흑마늘의 경우 열수 추출물 10 mg/mL 농도에서 69.40±0.13%의 활성을 나타내었으나 에탄올 추출물은 15 mg/mL 농도에서도 50.55±1.04%의 활성을 나타내었다. 환원력은 흑마늘 추출물에서만 유의적으로 높았으며 에탄올 추출물에서는 흑마늘, 생마늘, 찐마늘 순으로 환원력이 높았다. Oil emulsion 상에서 TBA 생성량은 열수추출물의 경우 흑마늘 추출물이 유의적으로 낮았으며, 에탄올 추출물은 시료간의 유의적인 차이가 미미하였다. 7. 흑마늘의 제조 공정을 4단계로 나누어 이화학적 성상의 변화를 분석한 결과 L, a 및 b값이 모두 감소하였는데 내부색의 경우 가공 초기에 크게 감소하였다. 수분은 4단계시료에서 58.48±0.41 g/100 g까지 감소하였고, pH도 점차 산성화되어 4단계 시료에서 pH 4.22±0.02였다. 총 페놀 화합물과 플라보노이드는 1단계 시료에 비해 최종단계에서 각각 약 1.9배, 2.6배 증가하였다. Total pyruvate와 thiosulfate도 비슷한 패턴으로 각각 약 1.6배, 5.8배 증가하였다. 8. 흑마늘의 제조 단계별 유리당의 변화는 fructose가 1단계에서 1,403.03±6.24 mg/100 g 였던 것이 4단계에서는 2,454.45±4.20 mg/100 g으로 증가하였으나, 반면에 이당류인 sucrose와 maltose는 흑마늘의 숙성과 더불어 서서히 감소하는 경향이다. 무기물의 총량은 4단계 시료에서 1,009.20±6.91 mg/100 g으로 가장 많았고, 흑마늘의 구성아미노산은 glutamic acid의 함량이 가장 많았고, 다음으로 proline과 aspartic acid였으며, 유리아미노산은 taurine, homocysteine 및 δ-hydroxylysine은 1단계 혹은 2단계 시료에서는 검출되지 않았으나, 2단계 혹은 3단계 이후시료에서 미량 검출되었다. 9. 흑마늘 가공 중 물추출물과 에탄올 추출물의 DPPH에 대한 전자공여능은 숙성이 진행될수록, 추출물의 첨가 농도를 높일수록 점차 증가하여 4단계시료의 1,000 μg/mL 농도에서 에탄올 추출물은 45.63±0.43%, 물추출물은 67.40±0.21%로 시험된 시료 중 가장 높은 활성을 보였다. 환원력은 100 및 500 μg/mL 농도에서 1, 2단계 시료를 제외한 모든 시료에서 물추출물이 에탄올 추출물 보다 높은 활성을 보였다. 10. 흑마늘 계통분획물의 항산화활성을 분석한 결과 전자공여능, 환원력, hydroxy radical 소거능, SOD 유사활성, pH 2.5에서의 아질산염 소거능에서 클로로포름 분획물의 활성이 가장 우수하였으며, 부탄올 및 물추출물의 활성이 낮았다. Linoleic acid 상에서 숙성 1일과 7일의 항산화 활성은 클로로포름 및 에틸아세테이트 분획물에서 높은 활성을 보였다. 11. 처리조건을 달리한 생마늘, 찐마늘 및 흑마늘 분말(3%) 및 열수추출물(0.3%)을 고지혈증 유발 흰쥐에 급이하였을 때 혈액 및 간의 지질함량 변화에 미치는 영향을 비교하였다. 마늘 급이군은 대조군에 비해 증체량 및 식이효율에서 유의적인 차이가 없었으며 실험군간에도 유의차가 없었다. 간의 무게는 대조군에 비해 생마늘 분말 급이군에서 유의적으로 낮았다. 12. 혈당은 마늘을 급이함으로써 대조군에 비해 감소되는 경향이었으며, 흑마늘 분말 및 추출물 급이군에서 유의적으로 낮았다. 혈중 총 지질은 생마늘 및 흑마늘 분말 급이군과 마늘 추출물 급이군에서 유의적으로 낮았으며, 총 콜레스테롤 및 중성지방은 마늘 급이군에서 대조군에 비해 모두 유의적인 감소를 보였다. 특히 중성지방의 함량은 마늘 급이군에서 정상군과 유사한 수준까지 감소되었다. 마늘을 급이함으로써 대조군에 비해 HDL-콜레스테롤의 농도는 증가되었고, AI 및 CRF는 흑마늘 급이군에서 가장 낮았다. GOT 활성은 대조군에 비해 유의적으로 저하되었으며, GPT 활성은 마늘추출물 급이군간에 유의적 차이가 없었다. LDH 활성은 흑마늘 추출물 급이군에서 가장 낮았다. 13. 간조직의 총 지질, 총 콜레스테롤 및 중성지방은 고지방식이를 급이한 대조군에 비해 마늘 급이군에서 유의적으로 낮았다. 간 조직의 TBARS 함량은 마늘 급이 유무에 따른 유의차가 없었으나, 항산화 활성은 마늘의 급이로 상승하였다. 고지혈증에 3% 마늘 분말 및 0.3%의 열수추출물 급이시 마늘의 가공처리조건에 관계없이 체내 지질 개선에 효과가 있었으며, 특히 흑마늘의 경우 동맥경화 예방 및 체내 지질 개선 효과가 클 것으로 기대된다.

Garlic (Allium sativum L.) is widely consumed due to its flavor and numerous health-promoting properties. Black garlic was manufactured by heating and aging garlic bulbs. Black garlic has altered physico-chemical properties, including formation of browning compounds, reduction of hot taste, and changes in textural attributes after processing. In this study, we determined physico-chemical characteristics and antioxidant activity of heat-treated garlic bulbs, fresh, steamed and black garlic. In addition, we investigated the effect of black garlic powder and its water extracts on total lipid, cholesterol and triglyceride of blood serum and liver in the rat fed by hyperlipidemic diet. 1. Garlic was aged at 60, 70, 80 and 90℃ for 1, 3 and 6 days in an incubator. The Hunter L, a and b value significantly decreased by increases in aging temperature and period. In the initiation of aging, the shear force of garlic had the lowest value (85.70±1.44 kg/cm2) at 90℃, but it increased after 6 days up to 411.30±13.90 kg/cm2. 2. The pH of aged garlic was lowered as aging temperature and period increased. After 6 days, pH of the garlic aged at 60℃ and 90℃ were pH 6.12 and pH 3.90, respectively. Total phenolic compounds and flavonoids contents in the garlic also increased as aging temperature and period increased. The contents of total phenolic compounds and flavonoids of black garlic aged at 90℃ after 6 days were higher about 3.5 and 9.1 times than those at 60℃, respectively. Total pyruvate contents showed significant difference between black garlics aged at 60~70℃ and 80~90℃. Total pyruvate content of black garlic aged at relatively lower temperature decreased during aging. 3. DPPH scavenging activity of garlic samples increased by the increase in aging temperature and period as well as extract highly added to reaction system. Especially, the activity of the garlic aged at 90℃ for 6 days showed the highest with a value of 87.48±0.20% than any other garlics (43.61~55.76%). Reducing power of the garlic was in good agreement with DPPH scavenging activity. The increase in antioxidant activity of garlic aged at high temperature may be caused by the increase of total phenolic compounds, flavonoids and brownig compounds contents during aging. 4. Physicochemical characteristics of black garlic were compared with fresh and steamed garlic. The contents of chemical components of the garlics, such as crude lipid, crude protein and total sugars were in order of black garlic > steamed garlic > fresh garlic, however moisture contents were viceversa. pH was 6.84±0.01 for fresh garlic, it was lowered for steamed(6.54±0.02) and black garlic(4.36±0.06). The contents of total pyruvate were 188.47±3.03 μmol/g, 77.03±0.97 μmol/g and 277.85±2.57 μmol/g for fresh, steamed and black garlic, respectively. The contents of total thiosulfinates were in order of black garlic, fresh garlic, and steamed garlic. 5. Arabinose and galactose were only detected in black garlic and their contents were 1.62±0.30 mg/100 g and 13.11±1.73 mg/100g, respectively. Contents of free sugars such as glucose, sucrose and fructose were in order of fresh, steamed and black garlic, respectively. Among the minerals, the content of K was the highest and its proportion was about 76% of total minerals. The amino acids such as glutamic acid, arginine and aspartic acid were dominant among detected amino acids in fresh, steamed and black garlic. Twenty kinds of amino acids, o-phosphoethanol amine and urea were detected in black garlic, while 15 kinds of free amino acids were detected in fresh and steamed garlic. 6. DPPH radical scavenging ability of hot water and ethanol extracts from black garlic was the highest among fresh, steamed, black garlics with values of 69.40 ± 0.13% at 10 mg/mL concentration and 50.55 ± 1.04% at 15 mg/mL concentration, repectively. Reducing power of garlic was significantly higher in black garlic, and was in order of black > fresh > steamed garlic. In oil emulsin, formation of TBA was significantly lower in hot water extract from black garlic and has no significant difference among ethanol extracts. 7. Glutamic acid, proline and aspartic acid were higher than other composition amino acids. The taurine, ethanolamine and δ-hydroxy lysine were not detected at the first processing step, but those was formed, up to the level of 1.00±0.69 mg/100 g, 0.31±0.26 mg/ 100 g and 0.68±0.96 mg/100 g at the forth processing step, respectively. 9. DPPH radical scavenging ability of water and ethanol extracts from garlic during its processing steps was significantly different. Water extract from black garlic at forth processing step displayed the strongest DPPH radical scavenging ability with values of 67.40 ±0.21% at 1,000μg/100 mL, followed by ethanol extract(45.63± 0.43%). Reducing power was also significantly higher in water extract than that of ethanol extract, except for 100 and 500 μg/100 mL the first and the second step samples. 10. Antioxidant activity as determined by DPPH radical scavenging ability, reducing power, hydroxy radical scavenging activity, SOD-like activity and nitrite scavenging ability in pH 2.5 were the highest in chloroform fraction and the lowest in buthanol and water fraction among the solvent extracts from black garlic. Antioxidant activity of black garlic extracts was higher in chloroform and ethyl acetate fraction than other fractions garlic during store for 1 and 7 day in linoleic acid. 11. Effect of different types of garlic products such as fresh, steamed, black garlic powder and their water extract on the lipids levels in the dietary hyperlipidemic rats were investigated. Rats were administrated with 1% cholesterol to induce hyperlipidemia and were fed by diet containing 3%(w/w) powder and 0.3%(w/w) hot water extract from fresh, steamed and black garlics for 4 weeks. Body weight gain during 4 weeks slightly decreased in garlic- fed groups, but it was not significant difference between control and garlic-fed groups. Liver weight significantly decreased in fresh garlic powder-fed group(FGP). 12. Blood glucose level decreased in garlic-fed groups, especially was lowered in black garlic powder(BGP) and black garlic extract(BGEx) feedig groups. Total lipids of serum was significantly lowered in the groups fed by FGP, BGP and 0.3% garlic extract. Total cholesterol and triglyceride remarkably decreased in garlic fed groups. Triglyceride content decreased in BGEx as control as. HDL-cholesterol was significantly higher, LDL-, and VLDL-cholesterol were significantly lower in the garlic- fed groups than the control. AI and CRF were the lowest in the black garlic- fed groups(BGP and BGEx). Activities of serum GOT was significantly lower, GPT activity was not different significantly between control and garlic fed groups. LDH activity was the lowest in the BGEx. 13. Total lipid, cholesterol and triglyceride of liver conspicuously decreased in the groups fed by garlic powder and its extract. Hepatic TBARS content was not different significantly by supplying garlic. Antioxidant activity was tended to be increased in garlic fed groups compared with control. 14. Based on above results, we suggest that the supplement of garlic with 3% for powder form or with 0.3% for water extract may improve total lipid, cholesterol and triglyceride of blood serum and liver in dietary hyperlipidemic rats. Especially, we supposed that black garlic had preventive effect against the atherosclerotic process and may improve hyperlipidemia through the removal of cholesterol.
Garlic (Allium sativum L.) is widely consumed due to its flavor and numerous health-promoting properties. Black garlic was manufactured by heating and aging garlic bulbs. Black garlic has altered physico-chemical properties, including formation of browning compounds, reduction of hot taste, and changes in textural attributes after processing. In this study, we determined physico-chemical characteristics and antioxidant activity of heat-treated garlic bulbs, fresh, steamed and black garlic. In addition, we investigated the effect of black garlic powder and its water extracts on total lipid, cholesterol and triglyceride of blood serum and liver in the rat fed by hyperlipidemic diet. 1. Garlic was aged at 60, 70, 80 and 90℃ for 1, 3 and 6 days in an incubator. The Hunter L, a and b value significantly decreased by increases in aging temperature and period. In the initiation of aging, the shear force of garlic had the lowest value (85.70±1.44 kg/cm2) at 90℃, but it increased after 6 days up to 411.30±13.90 kg/cm2. 2. The pH of aged garlic was lowered as aging temperature and period increased. After 6 days, pH of the garlic aged at 60℃ and 90℃ were pH 6.12 and pH 3.90, respectively. Total phenolic compounds and flavonoids contents in the garlic also increased as aging temperature and period increased. The contents of total phenolic compounds and flavonoids of black garlic aged at 90℃ after 6 days were higher about 3.5 and 9.1 times than those at 60℃, respectively. Total pyruvate contents showed significant difference between black garlics aged at 60~70℃ and 80~90℃. Total pyruvate content of black garlic aged at relatively lower temperature decreased during aging. 3. DPPH scavenging activity of garlic samples increased by the increase in aging temperature and period as well as extract highly added to reaction system. Especially, the activity of the garlic aged at 90℃ for 6 days showed the highest with a value of 87.48±0.20% than any other garlics (43.61~55.76%). Reducing power of the garlic was in good agreement with DPPH scavenging activity. The increase in antioxidant activity of garlic aged at high temperature may be caused by the increase of total phenolic compounds, flavonoids and brownig compounds contents during aging. 4. Physicochemical characteristics of black garlic were compared with fresh and steamed garlic. The contents of chemical components of the garlics, such as crude lipid, crude protein and total sugars were in order of black garlic > steamed garlic > fresh garlic, however moisture contents were viceversa. pH was 6.84±0.01 for fresh garlic, it was lowered for steamed(6.54±0.02) and black garlic(4.36±0.06). The contents of total pyruvate were 188.47±3.03 μmol/g, 77.03±0.97 μmol/g and 277.85±2.57 μmol/g for fresh, steamed and black garlic, respectively. The contents of total thiosulfinates were in order of black garlic, fresh garlic, and steamed garlic. 5. Arabinose and galactose were only detected in black garlic and their contents were 1.62±0.30 mg/100 g and 13.11±1.73 mg/100g, respectively. Contents of free sugars such as glucose, sucrose and fructose were in order of fresh, steamed and black garlic, respectively. Among the minerals, the content of K was the highest and its proportion was about 76% of total minerals. The amino acids such as glutamic acid, arginine and aspartic acid were dominant among detected amino acids in fresh, steamed and black garlic. Twenty kinds of amino acids, o-phosphoethanol amine and urea were detected in black garlic, while 15 kinds of free amino acids were detected in fresh and steamed garlic. 6. DPPH radical scavenging ability of hot water and ethanol extracts from black garlic was the highest among fresh, steamed, black garlics with values of 69.40 ± 0.13% at 10 mg/mL concentration and 50.55 ± 1.04% at 15 mg/mL concentration, repectively.
