Hyperoxidation state of peroxiredoxins (Prxs) by peroxides is known to be Prx-SO₂H, which is retroreducible to Prx-SH. However, no systematic study has been performed on other forms of hyperoxidized Prxs until now. Visualizing hyperoxidized Tsa1p (yeast peroxiredoxin) with anti-Tsa1p-SO₃H antiserum, I observed an obvious reactivity difference depending on the hydrogen peroxide concentrations to which yeast cells were exposed. On the basis of the retroreducing capacity of Srx 1, I identified two different types: reversibly and irreversibly hyperoxidized Tsa1p. Irreversibly hyperoxidized Tsa1p was determined to be Tsa1p-SO₃H by electrospray mass spectrometry. Tsa1p-SO₃H was not an autoxidation product from Tsa1p-SO₂H and was maintained in yeast cells at least two doubling cycles after recovery form H₂O₂ exposure. Tsa1p-SO₃H not only forms self-assembled high molecular weight complexes with homogeneously ring-shaped structure, but it also has about 4-fold higher chaperone activity than Tsa1p-SH. In addition, hyperoxidation to Tsa1p-SO₃H in response to H₂O₂ in yeast cells was significantly faster than in vitro hyper-oxidation in the presence of the thioredoxin system. In this study, I identified Tsa1p-SO₃H and its function as an effective and enduring molecular chaperone, and I also identified its function as a cellular marker that retains a cell's cumulative index of oxidative stresses. Since the discovery of Srx/sestrin, which catalyzes the reduction of Prx-SO₂H to -SH, several retroreduction mechanisms of Prx-SO₂H have been studiedon yeast and mammalian systems. Despite little difference on the retroreduction process of Prx-SO₂H between two species, sulfinic phosphoryl ester, thiosulfinate, and sulfenic acid have been considered in common as intermediates derived from the retroreduction process of Prx-SO₂H. However, retroreduction mechanism of Srx on Prx-SO₂H is still controvertial. In Saccharomyces cerevisiae, I observed that the reduction catalysis of Tsa1p-SO₂H by Srx 1 had been accomplished via a prominent 50 kDa Tsa1p band on non-reducing PAGE gel. The alkylation with 4-acetamido-4'-maleimidylstilbene-2,2'-disulfonic acid (AMS) and SDS-PAGE analysis following the reduction of 50 kDa Tsa1p band determined that 50 kDa Tsa1p does not have any free cysteine thiols and its oxidation states is sulfinic acid. Non-reducing 2D-PAGE analysis revealed that 50 kDa Tsa1p is joined through a disulfide linkage between two resolving cysteine residues from two independent Tsa1p-SO₂H polypeptide chains. This 50 kDa Tsa1p band did not appear in the process of the retroreduction of C170S mutant Tsa1p-SO₂H. Also, 50 kDa Tsa1p was correlated with the increasing protein expression of Srx 1 in yeast cell recovered from H₂O₂ and triggered structural change from heterogenous higher oligomers to lower oligomers. I propose that 50 kDa Tsa1p is an additional intermediate derived from retroreduction process of Tsa1p-SO₂H by Srx 1 and it may be important for understanding the retroreduction kinetics of Tsa1p-SO₂H.
