Basic characteristics of astaxanthin and Haematococcus pluvialis cracked cell, including solubility and stability were investigated, and the extraction conditions were optimized for the astaxanthin production from H. pluvialis cracked cells. Additionally, inclusion complexes of astaxanthin with various cyclodextrins were prepared and characterized with respect to water-solubility and stability. Finally, in vitro biological activities such as antioxidant activity, whitening activity, anti-hangover activity and anticancer activity were evaluated. First, the chemical compositions of H. pluvialis cracked cell, based on dry matter were 55.5% of carbohydrate, 16.64% of crude lipid and 13.41% of crude protein. Sugar composition of H. pluvialis cracked cell was glucose (62.3%), mannose (16.6%), galactose (10.5%) and fucose (5.6%). Astaxanthin showed a very poor solubility in water, but it was highly soluble in organic solvents such as acetone and acetic acid. The solubility of astaxanthin in acidic condition was enhanced as 10-20 times as in neutral and basic conditions. Astaxanthin was very unstable in acidic condition under UV irradiation and in the presence of oxygen. Also heating even for a very short time accelerated the degradation of astaxanthin. Second, the extraction of astaxanthin from H. pluvialis was examined using various solvents, such as acetone, ethanol, dichloromethane and methanol. Acetone was selected as the best solvent for the extraction of astaxanthin from H. pluvialis. Response surface methodology (RSM), based on a two-factor interaction model, was applied, and optimization of the extraction parameters such as β-glucanase concentration and sonication time was performed. Astaxanthin yields was the highest when it was extracted after H. pluvialis cell was treated by β-glucanase(2%) and the sonication was carried out spontaneously with the extraction for 75 min. Third, various inclusion complexes of astaxanthin with various cyclodextrins were prepared. The inclusion characteristics were determined through SEM (scanning electron microscope) and FT-IR spectrometer. The best host material for inclusion of astaxanthin was selected as β-cyclodextrin. The best ratio of astaxanthin and cyclodextrin for inclusion complex formation was found to be 1 : 200. The stability of the astaxanthin in inclusion complex was examined under storage at various pHs, temperatures, under UV irradiation and in the presence of oxygen for 4 weeks by measuring the astaxanthin concentration. Also, effects of heating on the stability of inclusion complex were investigated at various conditions (65℃ for 30 min, 85℃ for 15 sec and 100℃ for 5 sec). The stability of the inclusion complexes against heating, light and oxidation were greatly enhanced compared to free astaxanthin without inclusion complex formation. The water-solubility of the inclusion complexes was also investigated at various temperatures(4℃, 25℃ and 45℃) and pHs(2,6.5 and 8). As a result, the solubility of the inclusion complex (As-β-CDIC) was about 110 times higher than free astaxanthin at pH 6.5 and 25℃, respectively. Finally, the biological activities of astaxanthin and H. pluvialis extract such as antioxidant activity, whitening activity, anti-hangover activity and anticancer activity were investigated. The antioxidant activities of astaxanthin and H. pluvialis extract were significantly higher than that of α-tocopherol when the antioxidant activity were determined as the nitrate scavenging activity, hydroxyl radical scavenging activity and DPPH radical scavenging activity. The whitening effect of H. pluvialis extract was about two times as kojic acid. H. pluvialis extract indicated an anticancer activity on a lung cancer cell line and a cervical cancer cell line. Astaxanthin showed anti-hangover effect of 1.5 times as jiguja extract. The anti-hangover effect of the inclusion complex (As-β-CDIC) was about 1.2 times of jiguja extract. And, the inclusion complex of H. pluvialis (H.p.-CycloBIC) showed almost the same whitening effect as kojic acid. In conclusion, the inclusion complex of astaxanthin and β-CD, showinga highly improved water-solubility and stability against heating, light,oxidation and acidic pH.
