Thirty-two soil samples were collected from different layers of quercus and coniferous forest soil throughout the one year and analyzed pH, water content, and C/N ratio in each layer, and from the results, we confirmed that forest soil contains ecologically various habits. The average number of bacteria on the 10-2 diluted NB medium(DNB) was higher than that from the full strength NB medium with all samples taken at different layers throughout the year. Results suggest that oligotrophic bacteria exist abundantly in bacterial populations of rendzina forest soil. The number of colonies on DNB plates increased with incubation time following triple colony formation curves(CFCs). According to CFC, we obtained 459 isolates from CFC-I(about 24 to 216 hr), 401 from CFC-II(about 216 to 480 hr), and 363 from CFC-III(about 480 to 1200 hr), so a total of 1223 isolates. Seventy-three isolated successfully in the 1st isolation but did not grow in subculture. We temporarily designate as ‘temporarily culturable bacteria'. Based on the 16S rDNA sequences, 49 temporarily culturable isolates were classified into 6 phylogenetic groups: α-, β-, γ-proteobacteria, Acidobacteria, Actinobacteria and Firmicutes. A large proportion of these isolates not closely related to any genus in the current 16S rDNA sequence database, indicating the possibility of new genera. Among bacteria isolated from forest soil, soil bacteria that formed small colonies very slowly after 1000 hours’ culture and 18 bacteria were isolated from CFC-III. We temporarily designate as ‘slow growing oligotrophic bacteria.’ These were classified into 4 phylogenetic groups: α-, β-, γ-Proteobacteria and Actinobacteria. Slow-growing oligotrophic bacteria were suggested as novel order (6 isolates), novel family (4 isolates), and novel species (4 isolates). According to the results of the purification and identification experiment, strains L5, L8 L9, and L70 belonging to α-proteobacteria were suggested as novel order Rhodospirillales, strain L34 as a novel species belonging to family Rhodospirillaceae, and strain L55 as family Bradyrhizobiaceae. Strain L27, which belongs to family Burkholderia of β-proteobacteria, was suggested as a novel species. Strain L64 belonging to Actinobacteria was suggested as a novel species of family Solirubrobacter. Oligotrophic bacteria strain To13, which promotes plant growth, was suggested as a novel species of family Sphingomonas, and isolate To47 was suggested as a novel family of order Sphingomonadaceae. In order to design culture medium for the oligotrophic novel bacteria purified and identified in this study as above, we investigated the physiological characteristics of the oligotrophic bacteria. The amino acid content in soil was 200 ~ 600 times lower than that in NB medium. When oligotrophic bacteria were seeded in medium, which was prepared by adding 14 kinds of amino acid to basal medium, and analyzed their multiplication, amino acids showing a multiplication effect at concentration of over 1mM were glutamic acid, leucine, threonine, proline, and isoleucin. When glutamic acid and isoleucin amino acid, which showed high multiplication of oligotrophic bacteria, were added to basal medium at concentration of 1mM and the soil bacterial count was measured, the count was 1.5 ~ 2 times higher than that in DNB medium. Based on the results, we seeded oligotrophic bacteria in mediums to which glutamic acid, which showed high multiplication of oligotrophic bacteria, was added at concentration of 1mM, 5mM and 10mM respectively, and compared the multiplication of the oligotrophic bacteria. In the result, multiplication was over 4 times higher than that in DNB medium. The results of this study are expected to provide important basic materials necessary to design medium for the mass culture of industrial usable oligotrophic bacteria based on the physiological and biochemical characteristics of slow-growing oligotrophic bacteria.
