The present doctoral thesis describes observation of early developmental stages of Paralichthys olivaceus obtained from Sangrok-susan, Jeju-city in August 2008. The fertilized eggs were applied to different temperature and salinity conditions, and the survival rate and hatching time were monitored. Moreover, RT-PCR was conducted using a series of primer set involving body form change (metamorphosis) related genes.
The fertilized egg was spherical shape and transparent. The egg size was 0.89-0.95㎜(average 0.92mm, n=60) including one oil globule of 0.16~0.18㎜(average 0.17㎜, n=60), and many pigment cells are scattered on the yolk. During transportation to the laboratory, the fertilized egg was segmented continuously, and formed multiploids at 30 hours after fertilization. Kupffer cell was appeared, and hatched out through egg membrane at 49 hrs and 10 mins after fertilization.
In the different water temperature (15℃, 18℃, 21℃, 24℃ and 27℃), the highest hatching rate was observed at 21℃. The higher the water temperature, the shorter the developmental time was taken. In the different salinity (27‰, 30‰, 33‰, 36‰), the highest and lowest hatching rate were observed at 33‰ and 27‰, respectively.
Total length of just hatched larval fish was 2.43~2.53㎜(2.48±0.03 ㎜ n=10). At this stage, the mouth and anus were not open yet. The yolk was bigger than the body and a oil globule was located behind the yolk. Myotome was 11~14+20~23=31~37.
At 18 days after hatching, full length of the larval fish was 8.81~9.24㎜(9.07±0.16 ㎜, n=10), the end of chorda dorsalis was distinctly bent upward and 13 stems were formed on the tail fin. The stomach was expanded indicating with the development of the digestion system.
At 33 days after hatching, full length of the juvenile was 14.68~15.67㎜(15.36±0.32 ㎜, n=10), the body shape and mottles were similar with adult fish, it turned into juvenile period having complete stripes on the body.
Relative growth from a larval fish just after hatching to a juvenile fish (average total length, 15.36mm) formed breakpoint starting from metamorphosis in body length, anus length, head length, height against the full length, and eye diameter against the head length. The early development stage can be divided into two stages according to body form and size. Body length, anus length, head length, body height, and eye diameter against the full length showed clear gradient between the two straight lines, also the eye diameter against the head length showed clear gradient.
The highest growth rate (17.31±0.81㎜) was observed at 15℃ rearing for 33 days, and the highest survival rate (80.0%) was observed at 21℃. In the different salinity, the highest growth rate (16.45±0.62㎜) was observed at 33‰, and the highest survival rate (66.4%) was observed at 33‰.
A larval Paralichthys olivaceus prior to metamorphosis has two eyes on each side showing similar swimming mode with other fishes. However, the eyes combined together when it starts to live on the bottom.
In order to monitor the regulation of metamorphosis related genes, the present study attempted RT-PCR using cDNAs from larvae and juveniles of the early, mid, late, and complete stages on the process of metamorphosis. A positive control EF-1α showed strong expression signal in the mid, late, and complete stages. Type I procolalgen α1, α2, and α3 chains, consisting of body structure, strongly expressed in the mid and late stages. BMP1 showed an signal in the early and late stages of metamorphosis, the strong expressions of MMP2 and MMP13 were observed in the early, mid, and late stages of metamorphosis, and MMP17 showed a weak expression in the early stage of metamorphosis. TIMP2, which inhibited MMP enzyme activity, showed a similar expression pattern with MMP2. Fibronectin was expressed in the all stages and β-catenin interacting protein was expressed in the early stage of metamorphosis.
Moreover, RT-PCR was tried using cDNAs from various tissues and whole body (9.5cm, 4.9g) to figure out tissue specific distribution and the expression amount. A positive control, EF-1α was observed in all tissues but relatively weak expression was observed in muscle, liver, intestine, gill and the whole body, Type I procollagen α1 and α2 chains were observed in all tissues except for intestine. Type I procollagen α3 chain was found in skin, fin, and eyeball.
BMP2 was observed in fin, rectum, and eyeballs. The expression of MMP2, MMP9, and TIMP2 was observed in various tissues. The expression pattern of MMP2 and TIMP2 was same. MMP13 and MMP17 expressed in fin and rectum. Fibronectin was expressed in all organs and β-catenin interacting protein was expressed in brain, fin, rectum and eyeball.
