Polymeric nano-/microcarrires have been used in various applications such as optics, electrics, or drug delivery, etc. Various techniques such as emulsion, suspension, precipitation, or spray drying have been used to fabricate these carries. However, these technologies have some drawbacks to generate the novel functional nano-/microcarriers. Recently, electrohydrodynamic method (referred as to electrospraying or electrospinning) has been recognized as a technology to introduce interesting morphologies, shapes, additional functions into carriers. The study is focus on the fabrication and application of functional nano-/microcarriers via electrohydrodynamic jetting. Electrospraying technology has been studied in many fields to produce particles of various substances in nano-/microsize with using properly controlled processing variables and solution properties. Unlike pure liquids, polymer solution droplets in electrospraying have complicated solidification mechanisms of solvent evaporation, which mainly determine particle size and morphology. Herein, the effects of the governing parameters on the morphology and dimension of solidified particles were systematically studied. On the whole, the size of solidified spherical particles of smooth surfaces reflects that of droplets, which can be estimated by the theoretical equations developed for pure liquids. The particle size increases with an increase in polymer content and a decrease in the boiling point of a volatile solvent. Because of inhomogenous drying processes such as phase separation or skin formation, the particle sizes and morphologies of many cases are outside of the scope of the theoretical treatments. Surface charge, solvent evaporation rate, the development of inhomogeneous drying, etc. determine the particle formation mechanisms. Protein discovery in biotechnology is one of the most rapidly advanced areas, whereas the improvement of its stability should be achieved. The protein carries incorporating with biocompatible and biodegradable polymers should have the controlled released rate and stability improvement. To fabricate these particles, emulsion process was generally used. However, the method has some drawbacks such as the fact that the mechanical or thermal energy can denature the protein during the process. Electrohydrodynamic technology has been recognized as a useful method to produce the protein-encapsulated carriers without significant denaturation of proteins. The objective in the design of a controlled release system is to release a pharmacologically active protein in a predetermined, predictable, and reproducible fashion. Originally, the underlying rationale of controlled release formations was that a drug is more effective and exhibits less side effects when the drug concentration in circulation is kept constant at some optimum levels. Controllable intelligent nano-/microparticles were fabricated by co-axial electrospraying because of core/shell structure. The primary diffusion was controlled by the interaction and hydrophobicity between the proteins and components in the core. Pores in/on the shell polymer acted as the secondary diffusion channel. It is studied that porosity in polymeric carriers produced by electrohydrodynamic jetting becomes a channel to diffuse a material. In dental restorative materials, fluoride release is recognized as one of the most important properties because it may prevent development of secondary carious lesion at the restoration tooth interface. However, until recently, the release rate of fluoride ions was controlled and understood by empirical results without a scientific prediction. Therefore, we investigated the dental composites with porous channels produced by tri-nozzle co-axial electrospinning. Composites with nanoporous fibers did not show the improved release of fluoride because the hydrophobicity of poly(methyl methacrylate, PMMA) and Bis-GMA might not be enough to induce a capillary force. Using PAA in the porous fiber improved the release rate. This study suggests that the release of fluoride dominantly depends on water diffusion rather than the capillary force. Methods to in-situ construct nanostructures are the key tools of nanotechnology. Herein, for the enforced combination of two nanomaterials, a novel technique, ‘electrohydrodynamic jetting of countercharged nozzles (EJC)’, was explored. EJC used an attraction force between negative and positive surface charges generated by electrohydrodynamic spinning and spraying. The obtained nanostructures were dependent on the types of materials and jetting conditions. PMMA nano- or micro-fibers were uniformly combined with polystyrene nano- or microparticles. When lipoic acid (an antioxidant drug) was used instead of polystyrene, composite structures of fibers and surface nanodots resulted. The method could also enforce gold nanoparticles stick onto the surface of fibers. EJC is an efficient technique to fabricate composite nanostructures using materials with a wide range of properties. Nano-comminution was investigated as an alternative technology. The technology is a milling method that the shear force mills solid materials in the liquid medium. Especially, it is useful to fabricate nano-drug carriers. However, it is not easy to use the method for materials of poor crystal and chemical stability, especially, such as polymerizable drugs. Nevertheless, this study shows the successful case to generate nanodrugs as controlling polymerization conditions. Poorly soluble alpha-lipoic acid (ALA) induces significant body weight loss by appetite suppression, but has a short plasma half-life and low intrinsic bioavailability. To enhance the bioavailability and reduce the dosage requirement, ALA was fabricated into micro- and nanoparticles, and the efficacy and in vitrorelease were investigated. Due to the self-polymerization of ALA into poly[3-(n-butane carboxylic acid)propyl]disulfide (PBCPD) by the processing heat, low-speed rotation comminution was used to control the PBCPD content. The ALA particle size initially decreased and then increased after 10 h of nano-comminution, indicating aggregation related to PBCPD formation. The in vitro release of ALA was significantly reduced by the existence of PBCPD. Interestingly, the reduction was not followed by a decrease in efficacy. Alternatively, the food intake was significantly reduced by ALA particles containing more than 30 mol% PBCPD. The efficacy on appetite suppression was superior to water-soluble ALA salt, and the ALA particles might have a composite nanostructure of ALA and PBCPD. Stabilizers such as hydroxylpropyl cellulose (HPC), Pluronic F127, and polyvinylpyrrolidone (PVP) used in nano-comminution reduced the increased surface energy and controlled the chemical reactivity of ALA. Nano-ALA in suspension is more unstable than that in powder state produced by freeze-drying method. From In Vitro results, the release rate of ALA nanocrystal powders was slower than that of ALA suspension. In vivo results revealed that this nanocrystal formulation had an improved bioavailability related to the appetite suppression.

Composites with nanoporous fibers did not show the improved release of fluoride because the hydrophobicity of poly(methyl methacrylate, PMMA) and Bis-GMA might not be enough to induce a capillary force. Using PAA in the porous fiber improved the release rate. This study suggests that the release of fluoride dominantly depends on water diffusion rather than the capillary force. Methods to in-situ construct nanostructures are the key tools of nanotechnology. Herein, for the enforced combination of two nanomaterials, a novel technique, ‘electrohydrodynamic jetting of countercharged nozzles (EJC)’, was explored. EJC used an attraction force between negative and positive surface charges generated by electrohydrodynamic spinning and spraying. The obtained nanostructures were dependent on the types of materials and jetting conditions. PMMA nano- or micro-fibers were uniformly combined with polystyrene nano- or microparticles. When lipoic acid (an antioxidant drug) was used instead of polystyrene, composite structures of fibers and surface nanodots resulted. The method could also enforce gold nanoparticles stick onto the surface of fibers. EJC is an efficient technique to fabricate composite nanostructures using materials with a wide range of properties. Nano-comminution was investigated as an alternative technology. The technology is a milling method that the shear force mills solid materials in the liquid medium. Especially, it is useful to fabricate nano-drug carriers. However, it is not easy to use the method for materials of poor crystal and chemical stability, especially, such as polymerizable drugs. Nevertheless, this study shows the successful case to generate nanodrugs as controlling polymerization conditions. Poorly soluble alpha-lipoic acid (ALA) induces significant body weight loss by appetite suppression, but has a short plasma half-life and low intrinsic bioavailability. To enhance the bioavailability and reduce the dosage requirement, ALA was fabricated into micro- and nanoparticles, and the efficacy and in vitrorelease were investigated. Due to the self-polymerization of ALA into poly[3-(n-butane carboxylic acid)propyl]disulfide (PBCPD) by the processing heat, low-speed rotation comminution was used to control the PBCPD content. The ALA particle size initially decreased and then increased after 10 h of nano-comminution, indicating aggregation related to PBCPD formation. The in vitro release of ALA was significantly reduced by the existence of PBCPD. Interestingly, the reduction was not followed by a decrease in efficacy. Alternatively, the food intake was significantly reduced by ALA particles containing more than 30 mol% PBCPD. The efficacy on appetite suppression was superior to water-soluble ALA salt, and the ALA particles might have a composite nanostructure of ALA and PBCPD. Stabilizers such as hydroxylpropyl cellulose (HPC), Pluronic F127, and polyvinylpyrrolidone (PVP) used in nano-comminution reduced the increased surface energy and controlled the chemical reactivity of ALA. Nano-ALA in suspension is more unstable than that in powder state produced by freeze-drying method. From In Vitro results, the release rate of ALA nanocrystal powders was slower than that of ALA suspension. In vivo results revealed that this nanocrystal formulation had an improved bioavailability related to the appetite suppression.
