초록 수용성 고분자에 의한 난용성 약물의 용출율 향상 및 특성연구 이 현 구 전북대학교 일반대학원 BIN융합공학과 본 연구에서는 난용성 약물인 아토르바스타틴과 페노피브레이트를 폴리비닐피롤리돈, 하이드록시프로필메틸 셀룰로오스, Kollidon VA 64와 같은 수용성 고분자를 사용하여 고체분산체를 제조하였고, 이를 통해 용출율을 향상 시키려 하였다. 또한, 폴록사머 407를 계면활성제로 사용하여 약물과 전달체로 사용한 수용성 고분자간에 잘 혼합되도록 하였다. 두 난용성 약물의 고체분산체를 제조하기 위해 대표적인 방법인 분무건조법을 사용하였다. 용출실험은 인공위액과 장액에서 실시하였고, 대조군으로 시판제인 화이자제약의 리피토Ⓡ와 녹십자의 리피딜 슈프라Ⓡ와 비교하였다. 주사전자현미경을 이용하여 고체분산체의 형태학적인 특성을 분석하였고, 고체분산체의 결정학적 성질은 시차주사열량계와 XRD를 통하여 확인하였다. 고체분산체의 화학구조적 변화를 관찰하기 위해 적외선 분광법을 사용하였다. 분석결과 결정성 약물인 아토르바스타틴과 페노피브레이트가 무정형이 되었고 구조 또한 변하는 것을 확인할 수 있었다. 또한, 아토르바스타틴과 페노피브레이트는 약물과 고분자인 폴리비닐피롤리돈의 비율이 1:3일 때 가장 높은 용출율을 나타내었다. 따라서, 분무건조법을 통하여 난용성 약물의 용출율을 향상 시킬 수 있음을 확인하였다. 주요어 : 아토르바스타틴 칼슘, 페노피브레이트, 고체분산체, 분무건조, 용출율, 수용성 고분자 Student ID Number : 201250896

Abstract A Study on the Property and Improvement of Dissolution Rate of Water-insoluble Drug by Hydrophilic Polymers Hyun Gu Lee Department of BIN Fusion Technology Center Graduate School Chonbuk National University This study was conducted to improve dissolution rate of atorvastatin and fenofibrate with low solubility and high permeability (BCS class II) in water using polyvinyl pyrrolidone (PVP), hydroxyproylmethyl cellulose (HPMC) and Kollidon VA64 as carriers of solid dispersion. Poloxamer 407 has been used as a surfactant. Solid dispersion were prepared by spray drying method using difference ratios of drug and polymer. The characterization of the prepared solid dispersions was characterized by scanning electron microscopy (SEM), Fourier transformation infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC) and powder x-ray diffractometer (PXRD). In these results, the crystallinity and chemical structure of celecoxib was changed. The dissolution behavior of the prepared solid dispersions was compared with pure atorvastatin and fenofibrate, LipitorⓇ (Pfizer, USA) and Lipidil SupraⓇ (Green cross, Korea) as control groups in simulated phosphate buffer at pH 1.2 and 6.8. In atorvastatin and fenofibrate, the prepared solid dispersion as ratios of drug and carriers (1:3) appeared to have the highest dissolution rate. Thus, solid dispersion techniques of spray drying method can be successfully used to improve the dissolution rate of the poorly water soluble drug.
