In this study, it was evaluated cytotoxic effects of polyphenon 60 (PPE, a green tea polyphenol mixture) or (-)-epigallocatechin-3-gallate (EGCG) in the presence of generic drugs, including ibuprofen (Ibu), aspirin (Asp), and acetaminophen (AAP) on the human liver hepatoblastoma HepG2 cells, human colon carcinoma HCT 116 and HT-29 cells, as well as normal immortalized rat intestinal IEC-6 cells. After 24 hr incubation, PPE and EGCG inhibited growth of these 4 cell lines with IC_(50) ranges from 33.62±2.17 to 208.34±22.42 ㎍/mL, and from 37.80±1.22 to 167.59±3.02 μM, respectively, evaluated by the 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Both Asp and AAP up to 0.5 mM, were not effective in inhibiting HepG2 and HCT 116 cell growth; Ibu at 2 mM, however, inhibited HepG2 and HCT 116 cell growth by 43% and 87%, respectively. There was a significantly increased toxicity observed in HCT 116 cells treated with PPE (10 ㎍/mL) and AAP (100 μM) by 12% as compared with PPE alone. AAP (0-200 μM), however, did not affect on cytotoxic effect of EGCG (25 μM). Lower concentration of EGCG (below 10 μM) in the presence of AAP (10 mM) were significantly decreased AAP toxicity on HepG2 cells by 10% (p<0.01). EGCG stability was significantly increased in a culture medium and 1 M phosphate buffer containing millimolar levels of Asp. Chemical stability of AAP was not modified by incubation with EGCG at pH 2.0, 7.4, and 8.0. Cellular level of AAP after 2 hr incubation with EGCG was not significantly affected either. The present study provides basic information for potential health effects through the interactions of tea polyphenols with generic drugs.
