In vitro test를 진행한 결과 시료를 용매별 추출수율은 70% Ethanol 추출물에서 가장 높은 추출수율인 0.11%를 나타냈으며 용매별 추출물의 Free radical 소거능과 총 페놀함량, 항균효과 역시 70% Ethanol이 가장 우수한 효과와 가장 높은 총 페놀함량을 보여주었다. 이를 종합하여 70% Ethanol 추출물을 추출용매로 선정하고 실험 동물로 rat를 사용해 In vivo test를 실시하였다. In vivo test를 위해 시료를 일반식이 섭취군(Cont)과 고지방 식이 섭취군(HFG) 그리고 고지방식이 섭취 및 연잎추출물 투여군으로 구분하였으며, 연잎 추출물 투여군은 rat 의 체중 (kg) 대비 고형분 40 mg 투여군(HL40), 80 mg 투여군(HL80), 120 mg 투여군(HL120) 으로 세분하여 총 4주간 사육하였다.4주간 사료 섭취량 대비 체중증가량을 측정한 결과 HL40, 80, 120의 경우 고지방 식이를 하였기에 Cont 보다는 높은 체중증가량을 보여주었지만 동일한 고지방 식이를 행한 HFG와 비교하여 유의적으로 낮은 체중증가량을 보여주었다. 연잎추출물 섭취량이 증가 할수록 그 폭이 낮아져 HL 120의 경우는 Cont 와 오차범위 내에 있을 만큼 그 효과가 두드러지게 좋았다. 실험 동물 각 군의 간, 신장의 무게를 측정하여 체중 대비 비중을 측정한 결과 역시 체중 증가량과 비슷한 경향을 보여주었다, 간의 총 지질 함량 측정결과 HL120군이 HFG군보다 지질생성량이 낮게 나왔으나 유의적인 차이를 보여주지 못했다. 총 콜레스테롤 함량의 경우에는 Cont군과 HL120군에서 동일한 수치로 가장 낮게 나타났다. HFG군과 비교하여 HL120군에서 수치가 40% 가량 떨어졌고 HL40, 80 군과는 큰 차이가 없는 것으로 미루어 일정 수준 이상의 연잎 추출물 급이시 총콜레스테롤 수치를 떨어뜨리는데 효과가 있는 것으로 판단되었다. 중성지방의 경우 연잎추출물을 급이 한 모든 군이 HFG군 뿐만 아니라 Cont군에 비해서도 낮은 수치를 보였다. 특히 HL120군에서는 HFG군에 비해 그 수치가 절반 가량 줄어듦을 볼 수 있었다. 혈청 내 지질 패턴을 분석하기 위해 총 지질, 총 콜레스테롤, 중성지방 농도를 측정한 결과, 혈청 내 총 지질은 Cont 군을 제외한 나머지 군들은 오차범위 내에 있었으며 변별력을 보여주지 못했다. 중성지방의 경우 HL40, 80, 120군이 HFG보다 낮게 측정되었으며 총 콜레스테롤의 경우에도 동일한 경향을 보여주었다. Rat의 간을 채취하여 Malondialdehyde (MDA)의 양을 구하여 지질 과산화도를 측정한 결과 Cont 이 가장 적었으며 HL 120, 80, 40, HFG 순으로 그 양이 증가되어 연잎 추출물이 항산화능을 가지고 있으며 이는 섭취량에 비례함을 알 수 있었다. 이 후 생리활성을 확인한 연잎 추출물을 사용하여 이의 유효성분을 분리하고자 연구를 진행하였다. 연잎을 70% ethanol을 사용하여 80℃에서 추출 한 후 여과하여 추출액을 얻었다. 추출액은 감압농축 하여 water에 희석한 후 hexan, chloroform, ethyl acetate, butanol, water 순으로 각각 분액 깔대기에서 3회 반복 추출하여 용매별 분획물을 얻었다. 용매별 분획물의 추출수율과 Free radical 소거능을 측정하여 비교해본 결과 타 용매분획은 물론 합성항산화제인 BHA 보다도 우수한 Free radical 소거능을 보여주었으며 추출수율도 25%로 비교적 높게 측정된 ethyl acetate분획물을 취하여 silica gel column chromatography로 정제하였다. Silica gel column은 chloroform - methanol을 사용하였으며 용출하였으며 methanol농도를 1/10, 1/9, 1/8, 1/7 로 증가시키며 분획하였다. 용출물은 10 ml 씩 획득했으며 이를 538 nm에서 흡광도를 측정하여 흡광도가 변하는 시점을 임의로 묶어 총 30개의 분획물을 얻었다. 각 분획물의 free radical 소거능을 측정하여 활성이 인정되는 분획을 얻었다. 이들 중 Fr 13, 16, 21을 임의로 취하여 GC-MS를 측정하였다. GC-MS 측정결과 pyrocarechol 등 두수의 물질이 검출되었으며 이들 중 Fr. 16 에서 유사도 90% 이상을 보여준 pyrocatechol 과 phloroglucinol 로 추정되는 물질을 얻기 위한 목적으로 정제를 진행하고자 하였다. HPLC를 사용하여 pyrocatechol과 phloroglucino 표준물질의 peak와 Fr. 16의 peak를 측정한 후 Fr. 16의 peak 중 pyrocatechol과 phloroglucino 표준물질의 peak와 비슷한 구간에서 검출된 3.2 min, 3.8 min 구간의 물질을 정제하였다. 이들 정제물을 1H-NMR, 13C-NMR,을 사용하여

In vitro test를 진행한 결과 시료를 용매별 추출수율은 70% Ethanol 추출물에서 가장 높은 추출수율인 0.11%를 나타냈으며 용매별 추출물의 Free radical 소거능과 총 페놀함량, 항균효과 역시 70% Ethanol이 가장 우수한 효과와 가장 높은 총 페놀함량을 보여주었다. 이를 종합하여 70% Ethanol 추출물을 추출용매로 선정하고 실험 동물로 rat를 사용해 In vivo test를 실시하였다. The lotus (Nelumbo nucifera) leaf is known to be effective for 'over-coming body heat', and stopping bleeding. It has been used as a traditional curing plant for hematemesis, epistaxis, hemoptysis, hematuria and metrorrhagia in traditional Chinese medicine. The aim of the present study is to investigate the analysis chemical composition, antimicrobial, antioxidant and antiobesity potential of lotus (Nelumbo nucifera) leaf. The lotus leaf contained 63.8% of crude carbohydrate, 16.9% of crude protein, 1.0% crude fat and 9.3% of crude ash as dry basis. The main mineral of the leaf was calcium(2,2%) and showed much higher content than green tea etc. The antioxidative effect of several solvents extracts of lotus (Nelumbo nucifera) leaf was investigated. Among them 70% ethanol extract showed relatively higher extraction yield and higher total phenol content as well as the highest electron-donating ability (EDA) by using 1,1-diphenyl-2-picryl hydroxyl (DPPH). As the results the 70% ethanol exract was chosen for the further investigation in vivo test. Experimental rats were divided into 5 different experimental groups such as general diet group(Cont), high fat diet with lotus (Nelumbo nucifera) leaf extract oral adminstration group(HL 40, HL 80, HL 120) and high fat diet group(HFG). Body weights were significantly decreased in the sample of HL 120 than in Cont. The results of liver and kidney weight of rats were similar to the increase of body weights. Total cholesterol and triglyceride contents in liver tissue of rats were lowest in the sample HL 120. The level of total lipid, total cholesterol and triglyceride in serum were lower in the sample of HL 120 than HFG. The livers of rats were taken and MDA test was conducted for the evaluation of peroxidation degree. The results of MDA value of the control was lowest and increased as the followings order as HL 120, 80, 40, HFG, which means that the lotus leaf contained free radical scavenging compounds and the effectiveness of diet for the rats was in correlation with the taken amount. 70% Ethanol extract from lotus (Nelumbo nucifera) leaf was fractionated by using a variety of solvents, including hexane, chloroform, ethyl acetate, butanol and water. Only ethyl acetate fraction exhibited appreciable antioxidant activity, with the highest total phenol content and antioxidative activity. To isolate the component responsible for the antioxidative activity, the ethyl acetate fraction was further fractionated by using column chromatography. 30 Fractions were gained through column chromatography and among them the 16th compound showed the strongest antioxidant activity. It was further refined by means of HPLC and the result of GC-MS analysis. The active compounds were proved to be pyrocatechol (1,2-benzenediol) and Phloroglucinol (1,3,5-trihydroxy benzene). In rat brain homogenate peroxidation system the antioxidative activity of pyrocatechol was higher than BHA. Inhibition effect of the compound pyrocatechol in the peroxidation of linoleic acid reached to 80% and was higher than BHA. The growth inhibition effect of pyrocatechol on some pathogenic microogranisms in different concentrations in liquid culture were as follows.
