ESOO 및 ASOO의 혈관 장력 실험은 건강한 Sprague-Dawley계 수컷 백서에서 분리한 흉부 대동맥과 경동맥을 이용하여 실험을 진행하였다. ESOO 및 ASOO는 PE 으로 수축된 백서의 흉부 대동맥과 경동맥에서 농도 의존적인 혈관 이완 효과를 나타내었다. ESOO 및 ASOO에 의한 혈관 이완효과는 내피세포 의존적이었으며, 비 특이성산화 질소합성효소 (nitric oxide synthase)의 억제제인 N^(G)-nitro-L-arginine methyl ester (L-NAME)나 가용성구아닐산 고리화 효소 (soluble guanylate cyclse, sGC)의 억제제인 1H-[1,2,4]-oxadiazole-[4,3-α]-quonoxalin -1-one (ODQ)를 전처리 하면 차단되었다. ESOO 및 ASOO에 의한 혈관 이완효과는 L-형 Ca^(2+) 채널 차단제인 diltiazem 이나 verapamil에 의해서는 억제되지 않았다. 반면에 Ca^(2+) 저장고인 근형질세망 (SR) 의존적 Ca^(2+) 억제제인 ryanodine으로부터 부분적으로 차단되었으며, ATP 의존적 근형질세망으로 부터의 Ca^(2+) 유입 억제제인 thapsigargin으로 차단되었다. 이 두 가지를 함께 전 처리하여 사용하였을 때 차단 효과는 더욱 크게 나타났다. 비 선택적인 K^(+) 채널 차단제인 TEA와 4-aminopyridin을 통해 유의하게 차단되었다, ATP 의존적 K^(+) 채널 억제제인 glibenclamide와 Ca^(2+) 의존적 K^(+) 채널 억제제인 iberiotoxine을 전처리 하여 확인해 본 결과 glibenclamide의 효과만이 유의하게 관찰되어 ESOO 및 ASOO는 ATP 의존적이 K^(+) 채널에 의한 이완효과에 영향을 주고 있음을 확인 할 수 있었다. 반면에 indomethacin, atropine, propranolol 등의 전처리에 의해서는 억제되지 않았다. 또 ESOO 및 ASOO의 전처리는 백서의 흉부 대동맥과 경동맥에서 각각 PE에 의한 수축 반응을 현저하게 억제시켰다.
더욱이 ESOO 및 ASOO를 혈관 조직과 반응시켜 cGMP의 농도를 측정 한 결과 추출물의 농도 의존적으로 현저하게 증가하였으며 이 증가는 L-NAME 및 ODQ의 전처리에 의해 유의하게 차단되었다. 이는 ESOO 및 ASOO의 효과가 sGC의 활성을 통한 NO/cGMP계 신호체계와 밀접한 관계가 있음을 보여 주는 것이다. 또한 ESOO 및 ASOO는 혈관 조직의 cAMP의 생성량을 농도를 증가시켰다.

In the present study, vasorelaxant effect of the extract of seeds of Oenothera odorata (SOO) and its possible mechanism responsible for this effect were examined in vascular tissues isolated from rats. An ethanol extract of seeds of Oenothera odorata (ESOO) as well as methanol extract of seeds of Oenothera odorata (MSOO) relaxed endothelium-intact thoracic aorta and carotid artery in a dose-dependent manner. A dose-dependent vascular relaxation was also revealed by treatment of ethylacetate, n-butanol, and H₂O extracts partitioned from MSOO, but not by hexane extract. However, the vascular relaxation induced by ESOO and ASOO were abolished by removal of endothelium of aortic tissues. Pretreatment of the endothelium-intact vascular tissues with N^(G)-nitro-L-arginine methyl ester (L-NAME) or 1H-[1,2,4]-oxadiazole-[4,3-α]-quinoxalin-1-one (ODQ) significantly inhibited vascular relaxation induced by ESOO and ASOO. Moreover, ESOO and ASOO, respectively, significantly increased cGMP and cAMP production in vascular tissues. However, ESOO- and ASOO-induced increases in cGMP production were blocked by pretreatment with L-NAME or ODQ. The vasorelaxant effect of ESOO and ASOO was attenuated by tetraethylammonium (TEA), 4-aminopyridine, and glibenclamide attenuated. Also, relaxation effect of ESOO and ASOO was blocked by ryanodine and thapsigargin, respectively. On the other hand, the ESOO- and ASOO-induced vasorelaxation was not blocked by atropine, propranolol, indomethacin, verapamil, diltiazem, and iberiotoxin. Taken together, the present study demonstrates that ESOO and ASOO dilate vascular smooth muscle via endothelium-dependent NO-cGMP signaling pathway, which may be causely related with the function of K^(+) channels, and Ca^(2+) channels of the SR.
